[Alfalfa growth and its relation with soil water status in loess hilly and gully region].

This paper studied the dynamics of alfalfa growth and its relation with the excessive depletion and resumption of soil water in loess hilly and gully region. The results showed that alfalfa could grow in this region for 10 years, being most flourishing at the forth and fifth year, but declined gradually because of the excessive depletion of soil water. From 0 to 35 cm above ground, the stem biomass was higher than the leaf biomass, and the ratio of stem to leaf was 1.7:1; at 40 cm above ground, the biomass of stem and leaf was equal; and from 45 to 90 cm above ground, the leaf biomass was higher than the stem biomass, with the ratio of leaf to stem 1.42:1. The soil dry layer was not distinct in first two years of alfalfa growth, but getting thicker with time, being from 110 cm in third year to 260 cm in seventh year, with a 4.6%-6.2% of soil water content. It took 5 years for the natural resumption of soil water in degraded alfalfa grassland, and soil water content was increased with time.

PAHs pollution from traffic sources in air of Hangzhou, China: trend and influencing factors.

PAHs pollution in air of arterial roads was investigated from October 1998 to October 2001 in Hangzhou, China. The results showed that sigma10 PAHs was 13-36 microg/m3, among which, BaP, a strong carcinogenic kind ranged from 0.034 microg/m3 to 0.12 microg/m3. PAHs pollutions in four seasons were winter > autumn > spring-summer. The annual averages of sigmaPAHs concentration were 25 microg/m3 for 1999, 28 microg/m3 for 2000, and 29 microg/m3 for 2001, respectively. Leaded gasoline was banned in December 1998 in Hangzhou, thus comparative measurements with PAHs in leaded and lead-free gasoline powered motor exhausts made it certain that the use of lead-free gasoline leaded to a heavier PAHs pollution in roadside air from December, 1998, in China, and sigmaPAHs in air samples after the lead-banning were more than twice of that in samples before the action. For the large contribution of vehicle discharge to air pollution in roadside, further research was performed to suggest the factors influencing PAHs distribution in vehicle exhaust in order to control air pollution effectively. Compared to gasoline engines, emissions from diesel engines were less toxic, although they might produce more PAHs. Of the same vehicular and oil type, automobiles of longer mileages produced more toxic PAHs. PAHs distributions in the vehicular exhausts were related to the oil type. Large difference was found in the abundance of 3-, 5- and 6-ring PAHs between exhausts from gasoline and diesel oil engines. Diesel oil engines produced relative lighter PAHs such as NAPH, ACEN, FLUOR, while gasoline engines emitted heavier kinds such as BkF, IN and BP. The automobile produced more PAHs with the increase of mileage especially FLUR, PY, BaP, BP. Some significant ratios for traffic source in Hangzhou such as PHEN/AN, FLUR/PY, IN/BP were 0.50-4.3, 0.58-7.4, 0.51-1.5, respectively. A source fingerprint for vehicle exhausts of a mixture of vehicle and oil types in the city district for light-duty vehicle was the abundance of BaA, followed by NAPH, BP, IN. 4-ring PAHs such as FLUR, PY, BaA and CHRY were the most predominant kinds followed by 6-ring PAHs(BP, IN).

Biodegradation of azo dyes by genetically engineered azoreductase.

A azoreductase gene with 537 bp was obtained by PCR amplification from Rhodobacter sphaeroides AS1.1737. The enzyme, with a molecular weight of 18.7 kD, was efficiently expressed in Escherichia coli and its biodegradation characteristics for azo dyes were investigated. Furthermore, the reaction kinetics and mechanism of azo dyes catalyzed by the genetically engineered azoreductase were studied in detail. The presence of a hydrazo-intermediate was identified, which provided a convincing evidence for the assumption that azo dyes were degraded via an incomplete reduction stage.

METHODS: The levels of IGF-1 and IGFBP-1 in serum and placenta were detected from 60 patients with hypertensive disorder complicating pregnancy (hypertensive disorder complicating pregnancy group) and 18 normal pregnant women (control group) by enzyme linked immunosorbent assay (ELISA) and immunohistochemistry. RESULTS: (1) Serum level of IGF-1 (229 +/- 100) microg/L in hypertensive disorder complicating pregnancy group was significantly lower than that in control group (336 +/- 120) microg/L, (P < 0.01). There were significant differences among gestational hypertension (303 +/- 80) microg/L, mild preeclampsia (233 +/- 77) microg/L and severe preeclampsia groups (155 +/- 73) microg/L, (P < 0.05). (2) The expression of IGF-1 in the placenta of hypertensive disorder complicating pregnancy group (48%, 29/60) was significantly reduced than that in control group (83%, 15/18; P < 0.01). Its expressions in mild and severe preeclampsia patients were lower than that in control group (P < 0.05, P < 0.01). (3) Serum IGFBP-1 from hypertensive disorder complicating pregnancy group (161 +/- 90) microg/L was significantly higher than that in control group (98 +/- 75) microg/L, (P < 0.01). There were significant differences among gestational hypertension (97 +/- 73) microg/L, mild preeclampsia (157 +/- 69) microg/L and severe preeclampsia groups (225 +/- 81) microg/L (P < 0.05). (4) The expression of IGFBP-1 in hypertensive disorder complicating pregnancy group (77%, 46/60) was higher than that in control group (39%, 5/18), (P < 0.01). Its expression in mild and severe preeclampsia patients were higher than that in control group (P < 0.05). (5) The levels of IGF-1 in serum and placenta had a significant negative relationship with that of IGFBP-1 in hypertensive disorder complicating pregnancy group (r = -0.269, P < 0.05; r = -0.369, P < 0.01). Levels of IGFBP-1 in maternal serum were positively related with that of placenta (P < 0.01). CONCLUSION: The changes of levels of IGF-1 and IGFBP-1 are related to the occurrence and development of hypertensive disorder complicating pregnancy.

Novel constructs of tuberculosis gene vaccine and its immune effect on mice.

A novel tuberculosis (TB) gene vaccine containing mouse granulocyte macrophage-colony stimulating factor (mGM-CSF) and a TB antigen (Ag85A) was developed in this study. The genes encoding Ag85A and mGM-CSF were amplified by PCR respectively from the Ag85A-containing pBSby5 and pC-mGM-CSF. The genes were then cloned into two different polylinker sites of plasmid pIRES, forming a novel TB gene vaccine construct pI85AGM. Following transfection of pI85AGM plasmid into 7721 cell line by Lipofectamine(TM), the expression of Ag85A and GM-CSF proteins was identified by Western blotting or RT-PCR. Then Balb/c mice were inoculated with the recombinant pI85AGM, pI85A, pIGM or plasmid alone, respectively. The activities of CTL, NK cells and the Ag85A-stimulated proliferation of spleen cells were measured by MTT method. The serum antibody against Ag85A was detected by ELISA. The results showed that the Ag85A and GM-CSF proteins could be expressed in 7721 cell line and the activity of CTLs and the proliferation of spleen cells were significantly increased in the pI85AGM-immunized mice, indicating that the pI85AGM-immunized mice could generate specific immune responses to Ag85A. This study might provide possibility for developing novel anti-TB gene vaccine.

[Clinical results of circumferential pulmonary vein linear ablation in 100 patients with atrial fibrillation].

OBJECTIVE: To investigate the feasibility and efficacy of treating atrial fibrillation (AF) with circumferential pulmonary vein (PV) linear ablation guided by 3 dimensional mapping system and single circular mapping catheter. METHODS: From April 2004 to January 2005, PV isolation with circumferential PV linear ablation guided by CARTO system (in 76 patients) or EnSite-NavX system (in 24 patients) was performed in 100 consecutive patients with significantly symptomatic, drug refractory AF. The procedural end-point was complete electrical isolation of bilateral PV. RESULTS: Up to 200 linear circles were produced around each ipsilateral PVs in all 100 cases, and 95.0% (190/200) of PV isolation rate was achieved with a mean procedure time of 150-365 (240 +/- 65) min and a mean fluoroscopy time of 23-61 (37 +/- 12) min, respectively. Eight cases with recurrent AF (8.0%) underwent second session. Cumulative atrial tachyarrhythmias-free rate was 85.0% (85/100) during a mean follow-up of 5.5-12 (10.2 +/- 5.7) months. Atrial tachyarrhythmias-free rate was 66.0% (66/100), 82.0% (82/100), 87.0% (87/100), 85.0% (85/100), 85.0% (85/100), and 88.6% (70/79) during the follow up at 1 month, 2 months, 3 months, 4 months, 5 months and 6 months, respectively. There were 2 complications (1 tamponade and 1 PV stenosis), which were rehabilitated after conservative treatment. CONCLUSION: PV isolation with circumferential PV linear ablation guided by 3 dimensional mapping system is safe and effective for treating AF.

PAHs in aquatic sediment in Hangzhou, China: analytical methods, pollution pattern, risk assessment and sources.

Eleven surface sediment samples, from Hangzhou section of Qiantang River and Jinghang Canal, west Lake the inland river were collected to investigate 17 polycyclic aromatic hydrocarbons (PAHs) pollution in aquatic sediments of Hangzhou. Accelerated solvent extraction(ASE) was used to extract PAHs from sediments with satisfactory recoveries. It was found that the total PAHs in the sediments ranged from 308.4 to 3037 ng/g dw, and PAHs pollution in sediments from Jinghang Canal were the heaviest. Lowest effect level (LEL) and severe effect level (SEL) sediment quality guidelines were introduced to perform risk assessment for PAHs pollution in aquatic sediments. Only one sample in Jinghang Canal had adverse impact on benthic organism. 2-3 ring PAHs had a noticeable contribution to total PAHs, especially NA, PHEN. A quantity method was used to determine the major source, the results showed petroleum origin was the chief source to PAHs pollution in all sediments with the exception of sediments from Jinghang Canal where combustion sources had a larger contribution.

[Study of achieving white organic LED by fluorescence dye].

Some hybrid organic LEDs are made by using fluorescence dye and InGaN blue-light chip and the possibility of achieving white organic LED is investigated according to light conversion theory. Firstly, the LEDs made by normal method and double-dotting glue method are studied. It is found that the double-dotting glue method is too complex and the LEDs made by this way is low-luminance, so it isn't fit to make LED. Secondly the different weight ratio of 1/1/100, 1/1/200, 2/1.5/100 ax-17/zq-13/AB LEDs are manufactured. The 2/1.5/100 LED's color coordinate is (0.32, 0.30), approaching to white point and the correlative color temperature is 6290K which is close to sunlight. So it is a relatively ideal white lamp-house.

[Synthesis and characteristic of cobalt bearing ferrite particles at room temperature].

Cobalt bearing magnetite particles were synthesized by aerial oxidation of ferrous and cobalt ions at alkaline condition and room temperature. The samples obtained at different time intervals during reaction were subjected to XRD and IR, and it was indicated that the cobalt-bearing ferrites were produced through green-rust intermediate phase. Different compounds were synthesized using 1/20, 1/15, 1/10, 1/7, 1/5 and 1/3 Co2+/Fe2+ starting ratios, and Co-ferrites with Co/Fe of 0.05, 0.06, 0.08, 0.12, 0.17, 0.18 were obtained. The chemical composition, microstructure and micrograph of those precipitates were characterized by chemical analysis, IR, XRD and SEM. The results revealed that Co2+ replaced Fe3+ at octahedral position in spinel structure, and the increase of Co in initial solution showed deleterious effect on the crystallinity of the precipitates.

[Prospective 2-year clinical study of patients with positive IgG-antibodies after recovering from severe acute respiratory syndrome].

OBJECTIVE: To investigate the clinical characteristics of patients recovering from severe acute respiratory syndrome (SARS) during 2 years after the infection. METHODS: The antibody of SARS-IgG, T cell subsets, chest CT, and the pulmonary function were observed in patients 1 month, 3 months, 6 months, and 2 years after convalescence from SARS. RESULTS: In the 20 SARS cases, the level of antibodies was found to descend gradually and slowly during 2 years after convalescence. In the majority of patients T cell subsets recovered completely to normal range at the second examination. At the first re-examination, the rate of abnormal chest CT was 65%, and the main abnormal images included ground glass opacities, thickening of inter-lobular and intra-lobular septa, distorted lobular structure, thickened bronchovascular bundles, thickened pleura, arc shadow under the pleura, bronchiolar dilation, and honey comb like shadows. The rate of abnormal chest CT was 30% at the 4 fourth examination. At the first re-examination, the abnormal rate of KCO was highest, accompanied by abnormalities of forced expiratory volume in 1 second (FEV1) and the diffusing capacity of the lung for carbon monoxide (DLCO), and it began to recover since the third examination. CONCLUSION: The level of SARS-IgG descends slowly, and it may last for a long time. The recovery of chest CT to normal may take a long time. The abnormality in pulmonary functions manifests mainly as impairment of diffusion function. Further research on SARS is necessary.

[CD4+CD25high regulatory cells in peripheral blood of nasopharyngeal carcinoma].

OBJECTIVE: The current study was designed to investigate the changes in peripheral CD4+ CD25high regulatory T cells (CD4+ CD25high Tr) in nasopharyngeal carcinoma (NPC). METHOD: The peripheral was collected blood from 54 patients with NPC and 15 healthy controls, by using monoclonal antibodies, the blood samples were evaluated with the flow cytometry for lymphocyte subsets and Tr cells. RESULT: The proportion of CD4+ T cells, and the ratio of CD4+/CD8+ in NPC group was significantly less than those of the healthy controls (P < 0.05), but the proportion of CD4+ CD25high Tr cells was higher than those in controls (P < 0.01). The proportion of CD4+ CD25high Tr cells in the late stage was higher than those in the early stage [(stages I + II (2.28 +/- 1.07) %; stage III (3.65 +/- 1.21) %; stage IV (4.72 +/- 0.87) %)]. Comparing with the stages I + II, the proportion of CD4+ CD25high Tr cells in the stage III or IV was higher than those in controls (P < 0.01); and more, the proportion of CD4+ CD25high Tr cells in the stage IV was higher than those in the stage III (P < 0.01). CONCLUSION: The relative increase in CD4+ CD25high Tr cells in peripheral blood may be related to immunosuppression and tumor progression in patients with NPC. This finding suggests that CD4+ CD25high Tr cells in peripheral blood of NPC may be positive for prognosis analysis. Deletion of the CD4+ CD25high Tr cells may induce tumor immunity effectively and may be a new tumor treatment method.
