[Physical and chemical effects and plant growth suitability of substrates in subsurface flow wetland].

Substrate, which not only takes part in the pollutant-removing, but influences the plant growth, plays an important role in subsurface flow wetland. With X-ray fluorescence measurement and X-ray diffractometer, the elements and minerals in zeolite and shale were confirmed, and the removal mechanics of nitrogen, phosphate and hydrogen ion in substrates were explained respectively. The investigation show that the zeolite has abounded with micropores and mesopores, while the shale has only mesopores, which causes the NH4+ -N adsorption capability of the shale is less than zeolite. The PO4(3-) -P removal and hydrogen ion buffer capacity of shale are greater than those of zeolite because CaCO3 is one of the main contents of shale. In pilot-plant system to treat starch waste water, the reeds and acorus aclamuc were either planted in shale and zeolite, and the phytum's indexes in shale including relative green concentration, average plant height, root stem ratio were higher than those in zeolite. The root vitality of reeds and acorus aclamuc planted in shale were 3.7 and 1.6 times of those in zeolite respectively. Total nitrogen and phosphorus contents in the plant organization of acorus aclamuc in shale were 7.8 and 3.4 times of those in zeolite; total nitrogen and phosphorus contents in the plant organization of reeds in shale were 3.3 and 2.2 times of those in zeolite. The results indicate that shale provides a steady pH for the plant's root in the acid waste water and it is more suitable for plant growth than zeolite.

[The correlated research of acute graft versus host disease after allogeneic stem cell transplantation].

OBJECTIVE: To analyze the relationship between the expression of FasL, Perforin and Granzyme B and the development of acute graft versus host disease (aGVHD) after allogeneic hematopoietic stem cell transplantation (allo-HSCT). METHODS: The peripheral blood mRNA expression of granzyme B, perforin, fasL from 17 patients after allo-HSCT was detected by competitive quantitative RT-PCR and the relationship between FasL, Granzyme B and Perforin expressions and clinical symptom of aGVHD was analyzed. RESULTS: The expression level of Granzyme B, Perforin and FasL was 4.6 +/- 0.2, 4.5 +/- 0.1, 1.4 +/- 0.1 before aGVHD occurrence respectively, and was 98.7 +/- 2.5, 91.8 +/- 3.4, 61.5 +/- 2.2, after the occurrence in 14 patients (P < or = 0.05). Over expressions of Granzyme B, Perforin, and FasL during acute GVHD were detected in 13 of 14, 12 of 14, and 12 of 14 patients respectively. The upregulated expressions occurred prior to clinical symptom of aGVHD. CONCLUSION: The expressions of Granzyme B, Perforin, and FasL were significantly high in patients with acute aGVHD. Monitoring of the expressions, might predict the occurrence of clinical aGVHD and it severity and prognosis.

[Inhibitory action of propyl gallate on the activation of SAPK/JNK and p38MAPK induced by cerebral ischemia-reperfusion in rats].

AIM: To explore the protective effect of propyl gallate against neuronal injury in the boundary zone of the infarction area in the rat cerebral ischemia-reperfusion model and its possible mechanism. METHODS: Transient focal ischemia induced by middle cerebral artery occlusion in the rats was established by ligation of the left internal carotid artery for 2 h. Rats were treated by propyl gallate with different doses (23.5, 47 and 94 micromol x kg(-1)) for three days before operation. Coronal brain sections were collected after 1 , 2, 4, 6, 12 and 24 h of reperfusion, neuronal injury in the boundary zone of the infarction area was evaluated by TUNEL and Nissl staining. The expression of activated Caspase-3, total SAPK/JNK, p38MAPK and their phosphorylation (Thr183/Tyr185, Thr180/Tyr182) was investigated by immunohistochemistry and Western blotting with corresponding antibodies. RESULTS: Although SAPK/JNK immunoreactivity did not increase at each time point in the boundary zone of the infarction area after reperfusion, p-SAPK/JNK immunoreactivity increased significantly at 1 h and then decreased gradually, and p38MAPK immunoreactivity was enhanced at each time point, peaked at 6 h. Expression of p-p38MAPK peaked at 6 h. Activated Caspase-3 immunoreactivity appeared at 6 h in the boundary zone of the infarction area and peaked at 12 h. TUNEL positive neurons were observed at 12 h and became more abundant at 24 h. The number of Nissl positive neurons decreased gradually and apoptosis ratio of neurons peaked at 24 h. Propyl gallate reduced the immunoreactivity of SAPK/JNK, p-SAPK/JNK, p38MAPK and p-p38MAPK markedly at 1 and 6 h. Propyl gallate with doses of 47 and 94 micromol x kg(-1) were more effective. CONCLUSION: Inhibition on the activation of SAPK/JNK and p38MAPK is the possible protective mechanism of propyl gallate against neuronal injury induced by cerebral ischemia-reperfusion.

[Effect of mesenteric lymph duct ligation on nitric oxide content and its expression in lung in rats with hemorrhagic shock].

OBJECTIVE: To observe the effect of mesenteric lymph duct ligation on nitric oxide (NO) and its expression in lung in rats with serious hemorrhagic shock at different periods, and explore the role of intestinal lymphatic pathway in acute lung injury (ALI). METHODS: Seventy-eight male Wistar rats were randomly divided into the sham operation group (n=6), shock group (n=42), and ligation group (n=30). The model of serious hemorrhagic shock was reproduced in shock group and ligation group. Mesenteric lymph was diverted by ligating mesenteric lymph duct in ligation group after resuscitation. All rats were executed and their lungs were harvested. NO and nitric oxide synthase (NOS) were determined in lung homogenate 90 minutes after shock, and 0, 1, 3, 6, 12 and 24 hours after resuscitation. The expression of inducible nitric oxide synthase (iNOS) mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: The content of NO, activity of NOS and expression of iNOS mRNA in lung tissue in shock group were increased 3 hours after transfusion and resuscitation, and they became higher 6-12 hours after resuscitation. The values were significantly higher than those in sham operation group, 90 minutes after shock and 0 hour after transfusion and resuscitation (P<0.05 or P<0.01). The values were only increased 3 hours and 6 hours after transfusion and resuscitation, and all the values on 6, 12 and 24 hours after resuscitation were significantly lower in the ligation group compared with those in shock group at the same time points (P<0.01 or P<0.05). CONCLUSION: The results demonstrate that the ligation of mesenteric lymph duct could reduce the NO and expression of iNOS mRNA in the lung tissue, thus ameliorate ALI in rats with serious hemorrhagic shock. Mesenteric lymph might play an important role in the pathogenesis of ALI.

[Soil microbial ecological process and microbial functional gene diversity].

Soil microbes in terrestrial ecosystem carry out a series of important ecological functions, such as geo-chemical cycling of elements, degradation of pollutants, and buffering to the acute changes of environment, etc. Soil microbial ecological function has a close relation with soil function, and the changes in the structure and composition of soil microbial populations can directly affect the realization of soil function. Through their produced enzymes, soil microbes take part in a series of metabolic activities, and the functional genes of coded enzymes are the functional markers of microbes. In recent ten years, molecular ecology focusing on the functional gene diversity has been developed rapidly, which gives us a new cut-in point to understand soil microbial ecological function from the point of functional gene. This paper reviewed the research advances in the functional gene diversity correlated to soil microbial ecological function, with the perspectives in this field discussed.

[Survival analysis on liver failure patients treated with an artificial liver support system].

OBJECTIVE: To evaluate the efficacy of artificial liver support system (ALSS) in the treatment of liver failure patients. METHODS: This is a prospective, multi-center, controlled, large sample clinic trial. 518 patients with liver failure from 5 hospitals were studied and followed. All the patients received similar pharmacological manipulation according to one and the same protocol but were divided into an ALSS treatment group and a control group without ALSS treatment. The ALSS treatment procedures included plasma exchange, molecular adsorbent recirculating system (MARS), plasma exchange plus hemofiltration and other combined nonbioartificial methods. The analysis of survival time was computed using the Kaplain-Maier method, and comparison among groups was done using Log-Rank, Breslow and/or the Tarone-Ware test. RESULTS: Survival time of acute liver failure patients was prolonged from 4.0+/-0.2 days to 8.0+/-0.4 days (P=0.004). ALSS was shown to be two times more effective. ALSS increased the survival time of acute on chronic (A on C) liver failure patients from 27.0+/-1.6 days to 39.0+/-4.0 days (P less than 0.01). In addition, it increased the survival time of the patients in the middle and end stage of subacute liver failure and A on C liver failure, but had no significant effects on early stage patients. The survival time of middle stage patients was 38.0+/-17.5 days in the control group vs 66.0+/-18.6 days in the ALSS group (P less than 0.05). The survival time of end stage patients of the control group and the ALSS group was 18.0+/-4.0 days vs 26.0+/-2.5 days (P less than 0.01). CONCLUSIONS: Multi ALSS treatment is more effective than the standard medicinal liver care treatment. Multi-ALSS treatment could increase survival time of patients suffering from acute liver failure or A on C liver failure, especially in their middle and end stages. It is important and necessary to treat these patients with ALSS.

[The endocrine function of skeletal muscle].

Skeletal muscle has long been considered as an effector organ under the control of neural and humoral regulation. Recent studies indicated that skeletal muscle is not only an effector organ but also an endocrine organ. It is reported that the skeletal muscle can express, synthesis and secrete a lot of active factors, such as regulatory peptide, cytokine, growth factor and so on. The active factors play important roles not only in the growth, metabolism and motor function of skeletal muscle per se by paracrine/autocrine pathway but also in the function of peripheral tissues and organs by endocrine pathway. It is worthwhile to pay close attention to the significant effects of the skeletal muscle active factors on locomotor disease and some systemic diseases. This paper reviews the active factors secreted by skeletal muscles and their physiological and pathophysiological significance.

[Determination of three nitroimidazole residues in royal jelly by high performance liquid chromatography-tandem mass spectrometry].

A method for analysis of trace metronidazole (MTZ), dimetridazole (DMZ) and ronidazole (RNZ) residues in royal jelly was developed by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). After samples were dissolved in sodium hydroxide solution to disassociate target analytes from matrix, liquid-liquid extraction methods by ethyl acetate solvent were used. Matrix effects were minimized and good quantitation results were obtained by using highly-selective reaction monitoring (H-SRM) technology when deuterated dimetridazole (dimetridazole-D3) was selected as internal standard. Limits of detection (LODs) were 1.0 microg/kg for DMZ, 0.5 microg/kg for MTZ and RNZ (S/N > 5). Limits of quantitation (LOQs) were 2.0 microg/kg for DMZ, 1.0 microg/kg for MTZ and RNZ (S/N > 10). The linear ranges were 2.0 - 200 microg/L for all target analytes. Recoveries and relative standard deviations (RSDs) were in the ranges of 96.6% - 110.6% and 2.1% -7.4%, respectively. This method is suitable for statutory residue testing in the National Residue Surveillance Plan in China and meets the requirement for export.

[Study on the characterization of beta-cyclodextrin-diferrocenylenone inclusion complex and micro-environmental effects].

The structure of the inclusion complex of diferrocenylenone and beta-cyclodextrin prepared by a novel method of solid-grinding was studied by UV, FTIR and powder X-ray diffractometry. Continuous variation plot proved that 2 : 3 (DFE : beta-CD) inclusion complex has been formed. The binding constant of the inclusion compound was found to be 1.48 x 10(15) (L4 x mol(-4)) with the data of UV spectroscopy. Furthermore, the influence of different solvents and pH on the UV spectrum of DFE and beta-CD-DFE was discussed respectively. The results show that different micro-environment has apparently different effects on the electric spectra of guest and inclusion complex.

[Echocardiographic changes and anticardiolipin antibodies in patients with systemic lupus erythematosus].

OBJECTIVE: To analyze the echocardiographic abnormalities and the prevalence of anticardiolipin antibodies (aCL) in systemic lupus erythematosus (SLE) patients and to evaluate the relationship between aCL and cardiac valvular abnormalities in SLE patients. METHODS: Ninety SLE patients were performed M-mode, 2-dimensional and Doppler echocardiography and aCL IgG and IgM were measured by an enzyme-linked immunosorbent assay (ELISA). According to the abnormalities in the echocardiography, the patients were assigned into valvular abnormality group and non-valvular abnormality group. Chi-square method was used to compare the difference of aCL prevalence between the two groups. RESULTS: The prevalence of echocardiographic abnormalities was 53.33%, and valvular abnormality (38.89%) and pericardial effusion (34.44%) presented most frequently. The aCL prevalence was 32.56% in the 43 SLE patients. The prevalence of aCL in the valvular abnormality group was significantly higher than that in non-valvular abnormality group (52.94% vs 19.23%, P<0.05). CONCLUSION: The incidence of echocardiographic abnormalities is high in SLE patients, most often in valves and pericardium. The aCL is probably related to valvular damage in SLE patients.

[Mutations of tumor suppressor gene PTEN mutations in hepatocellular carcinoma and its implications in tumor proliferation and apoptosis].

OBJECTIVE: To study mutations of tumor suppressor gene PTEN in human hepatocellular carcinomas and its effects on the proliferation and apoptosis of hepatocellular carcinoma cell line HHCC. METHODS: (1) PCR-SSCP and sequence analysis were used to detect the mutations of the 5th and 8th exon of PTEN in 42 cases of human primary hepatocellular carcinoma. (2) Eukaryotic expression vectors of the wild-type (pEGFP-wt-PTEN) and the mutant type (pEGFP-PTEN, G129R) of PTEN were constructed. Lipofectamine 2000 mediated gene transfection was used to transfect hepatocellular carcinoma cell line HHCC, in which the PTEN protein is not expressed. Culture medium containing G418 was used to select stable transfectants. MTT colorimetry was used to analyze the proliferation ability of selected cell lines. Naive HHCC cells and HHCC cells transfected with empty vector (pEGFP-C1) served as controls. (3) TNF-alpha was used to induce apoptosis of selected cell clones. RESULTS: (1) Point mutation involving the 5th exon of PTEN was detected in 4 of 42 primary hepatocellular carcinomas. (2) Compared with the control groups, the proliferation of hepatocellular carcinoma cells was significantly inhibited by the transfection of wild-type PTEN gene, while the transfection with mutant PTEN construct did not significantly change the proliferation. (3) The apoptosis indices of cells transfected with the wild-type and the mutant PTEN genes were 13.8% and 8.1% respectively. Compared with the control, the apoptosis index of HHCC cell transfected by the wild type PTEN was significantly lower (P < 0.05). There were no significant differences between HHCC cells transfected with mutated PTEN gene and the control (P > 0.05). The expression of internal 473-phosphorylated Akt of HHCC was weak, but was enhanced when the cells treated with TNF-alpha. However, it was down regulated by the wild type PTEN. CONCLUSIONS: (1) First time report that PTEN mutations can be found in 9.5% human primary hepatocellular carcinomas. (2) The expression of the wild-type PTEN can suppress the proliferation of HHCC cells, and such suppression was lost when PTEN gene was mutated. (3) PTEN inhibition of the proliferation and the enhancement of apoptosis of hepatocellular carcinoma cells is likely related to a down-regulation of the TNF-alpha induced activation of protein kinase Akt pathway.

[Combined use of transmyocardial stents with gene therapy in the treatment of acute myocardial infarction].

OBJECTIVES: To determine the efficacy of combined use of transmyocardial stent with gene therapy to treat acute myocardial infarction in porcine model. METHODS: 24 Chinese mini swines have been divided into 4 groups randomly: group myocardial infarction (group MI n(1) = 6), group transmyocardial stent (group ST n(2) = 6), group vascular endothelial growth factor (group VEGF n(3) = 6), group transmyocardial stent and VEGF (group ST + VEGF n(4) = 6). In group MI, acute myocardial infarction animal model has been established by the ligation of the left descending coronary artery. In group ST, after the establishment of the model, 3 transmyocardial stents were implanted. In group VEGF, an expression plasmid containing the gene-encoding VEGF(165) (300 microg) was administered directly in the myocardium at 6 sites. In group ST + VEGF, both transmyocardial stents and expression plasmid containing the gene-encoding VEGF(165) are applied. 4 weeks later, the animals are sacrificed and echocardiography and pathological analysis have been done. RESULTS: The density of blood vessel in group ST, VEGF and ST + VEGF are significantly higher than group MI. And capillary density in group ST + VEGF is the highest in these groups statistically. Expression of VEGF was detected in group ST, VEGF and ST + VEGF, but in group VEGF and ST + VEGF the level of expression are higher. CONCLUSION: Combined use of transmyocardial stent with gene therapy has synthetic effect for the treatment of acute myocardial ischemia in porcine model and can significantly increase the vascular density.
