Identification of syn- and anti-anethole-2,3-epoxides in the metabolism of trans-anethole by the newly isolated bacterium Pseudomonas putida JYR-1.

Bacterial strain JYR-1, which utilizes high concentrations (up to 100 mM) of trans-anethole as the sole source of carbon and energy, was isolated from soil. It grew to OD(600)(nm) = 2.6 with a doubling time of 8 h when grown on 20 mM trans-anethole. Strain JYR-1 was identified as Pseudomonas putida based on the partial gene sequence of its 16S rDNA. Elution profiles of culture extracts were examined by high-performance liquid chromatography and showed that four metabolites were produced from the bacterial culture containing trans-anethole that were not detected in control experiments. LC-MS analysis showed molecular weights of 138.2, 164.5, 164.3, and 152.3. The metabolites with molecular weights at 152.3 and 138.2 were confirmed to be p-anisic acid and p-hydroxybenzoic acid, respectively, when compared with HPLC retention times and molecular weights of authentic compounds. The metabolites with molecular weights at 164.5 and 164.3 were further analyzed by NMR and were proved to be stereoisomer syn- and anti-anethole epoxides. Therefore, strain JYR-1 most likely initiates the metabolism of trans-anethole through the formation of epoxides on the propene group of the compound.

A benzenoid from the stem of Acanthopanax senticosus.

Seven compounds were isolated from the stem of Acanthopanax senticosus by repeated column chromatography. Their structures were elucidated as isovanillin (1), (-)-sesamin (2), isofraxidin (3), (+)-syringaresinol (4), 5-hydroxymethylfurfural (5), eleutheroside B (6), and eleutheroside E (7) by spectral analysis. Among them, isovanillin (1) was isolated for the first time from the family Araliaceae.

Anti-oxidant activities of Acanthopanax senticosus stems and their lignan components.

The antioxidant activities of Acanthopanax senticosus stems were evaluated in CCl4-intoxicated rats. The n-butanol fraction from the water extract of the stems, when pretreated orally at 200 mg/kg/day for 7 consecutive days in rats, was demonstrated to exhibit significant increases in antioxidant enzyme activities such as hepatic cytosolic superoxide dismutase, catalase and glutathione peroxidase by 30.31, 19.82 and 155%, respectively. The n-butanol fraction whereas showed a significant inhibition of serum GPT activity (65.79% inhibition) elevated with hepatic damage induced by CCl4-intoxication. Eleutheroside B, a lignan component, isolated from the n-butanol fraction was found to cause a moderate free radical scavenging effect on DPPH, its scavenging potency as indicated in IC50 value, being 58.5 microM. These results suggested that the stems of A. senticosus possess not only antioxidant but also hepatoprotective activities.

New lanostane-type triterpenoids from Ganoderma applanatum.

Four new lanostane-type triterpenes were isolated from the MeOH extract of the fruiting bodies of Ganoderma applanatum. Their structures were established as 3beta,7beta,20,23xi-tetrahydroxy-11,15-dioxolanosta-8-en-26-oic acid (1), 7beta,20,23xi-trihydroxy-3,11,15-trioxolanosta-8-en-26-oic acid (2), 7beta,23xi-dihydroxy-3,11,15-trioxolanosta-8,20E(22)-dien-26-oic acid (3), and 7beta-hydroxy-3,11,15,23-tetraoxolanosta-8,20E(22)-dien-26-oic acid methyl ester (4), respectively, by extensive spectroscopic analyses.

Cerebrosides from Longan Arillus.

From the pulp of Euphoria longana (Longan Arillus), three cerebroside molecular species have been isolated. Six known cerebrosides, soyacerebrosides I and II, 1-O-beta-D-glucopyranosyl-(2S,3R,4E,8E)-2-(2'-lignoceroylamino)-4,8-octadecadiene-1,3-diol (longan cerebroside I) and its 8Z isomer (longan cerebroside II), momor-cerebroside I, and phytolacca cerebroside, were identified as major components of these cerebroside molecular species. All the cerebrosides were shown to be a mixture of geometrical isomers (8E and 8Z) of sphingosine-type or phytosphingosine-type glucocerebrosides possessing 2-hydroxy fatty acids. The structures of these cerebrosides have been determined on the basis of chemical and spectroscopic evidence.
