Cerebrosides from Longan Arillus.

Abstract

From the pulp of Euphoria longana (Longan Arillus), three cerebroside molecular species have been isolated. Six known cerebrosides, soyacerebrosides I and II, 1-O-beta-D-glucopyranosyl-(2S,3R,4E,8E)-2-(2'-lignoceroylamino)-4,8-octadecadiene-1,3-diol (longan cerebroside I) and its 8Z isomer (longan cerebroside II), momor-cerebroside I, and phytolacca cerebroside, were identified as major components of these cerebroside molecular species. All the cerebrosides were shown to be a mixture of geometrical isomers (8E and 8Z) of sphingosine-type or phytosphingosine-type glucocerebrosides possessing 2-hydroxy fatty acids. The structures of these cerebrosides have been determined on the basis of chemical and spectroscopic evidence.

Longifoside-A and -B: two new cerebrosides from Mentha longifolia (Lamiaceae).

Two new N-acyl-glycosphingosines (cerebrosides) named longifoside-A {6'-tetracosenamide, N-[3,4-dihydroxy-1-(hydroxymethyl)-2-(beta-D-glucopyranosyl) heptadecyl]-(6'E)} and B {6'-tetracosenamide, N-[3-hydroxy-1-(hydroxymethyl)-2-(beta-D-glucopyranosyl)-4Z-heptadecenyl]-(6'E)} have been obtained from the methanolic extract of Mentha longifolia belonging to the family Lamiaceae. Both the cerebrosides were purified as their acetate-derivatives: 6'-tetracosenamide, N-[3,4-diacetoxy-1-(acetoxymethyl)-2-(tetraacetoxy-beta-D-glucopyranosyl) heptadecyl]-(6'E) and 6'-tetracosenamide, N-[3-acetoxy-1-(acetoxymethyl)-2-(tetraacetoxy-beta-D-glucopyranosyl)-4Z-heptadecenyl]-(6'E). They were characterized with the aid of 1D and sophisticated 2D-NMR spectroscopic techniques.

Ethanol-induced changes in glycolipids of Saccharomyces cerevisiae.

Total glycolipid content of Saccharomyces cerevisiae cells increased in ethanol-treated yeast cells. Sialic acid and hexosamine contents of glycolipids from ethanol-treated cells decreased, whereas those of hexoses increased. Increased sialidase activity in the presence of ethanol may be responsible for the decrease in sialic acid content of glycolipids. The saccharide moieties of glycolipids of S. cerevisiae consisted of fucose, mannose, galactose, and glucose. Ethanol treatment of yeast cells caused an increase in glucose and a decrease in galactose content of glycolipids. The changes in glucose content can be related to changes in beta-glucosidase activity under alcohol stress. The content of cerebrosides, sulfatides, and monoglucosyldiglycerides was enhanced following ethanol treatment. An increase in cerebroside as well as in sulfatide content during alcohol stress might play an important role in stabilizing the membrane both physically and structurally. Such variations in glycolipid content and composition of S. cerevisiae cells may represent an adaptive response to ethanol stress.
