A simple, rapid and effective method for total RNA extraction from Lentinula edodes.

A rapid, inexpensive and reliable method for total RNA extraction from fruiting bodies of Lentinula edodes containing large quantities of polysaccharides and secondary metabolites is described. An initial extraction step using saturated NaCl solution facilitates the separation of nucleic acids from contaminants and, after further extraction with organic solvents and precipitation with 2-propanol, total RNA of high purity and suitable for applications such as cDNA synthesis, RT-PCR and Northern blot hybridization was obtained. The procedure may also have wider applicability for total RNA extraction from the tissues of other mushrooms.

[Study on application of Fourier transformation near-infrared spectroscopy analysis with support vector machine (SVM)].

Support Vector Machine (SVM) is a method for the research on identifying two types of problem. It is the latest branch in the statistics study theories, and the identification model has a strict mathematics foundation. In this paper, the basic principle and method of SVM are not only introduced, but also applied to chemometrics. One hundred and three rhubarb samples were used as experimental materials. The identification models were established with near-infrared spectroscopy and SVM training method with the intention of identifying whether the rhubarb samples are true or false. The thirty-three samples in training set were identified by the identifying models with the accurate rate of 100%, while seventy estimate samples had an accurate rate of 96.77%. The research result provided the method of identifying the traditional Chinese medicine rhubarb quickly. So, it shows the feasibility of establishing the models with near-infrared spectroscopy and SVM method to identify biological samples. This paper introduced the theme of SVM training method in order to beget the attention of the research members who deal with chemometrics.

[An applied study on Fourier transform near-infrared whole spectroscopy regression analysis].

In the present paper, 66 wheat samples were used as experimental materials, 33 of them were used for building the quantitative analysis model of protein content, and the rest composed the prediction set. Using Moore-Penrose matrix, we estimated directly the regression coefficients of the regression analysis model with Fourier transform near-infrared (FTNIR) whole spectroscopy. The samples of prediction set were analyzed, and the correlation coefficient is 0.979 9 between the prediction values of the near-infrared model and the standard chemical ones by Kjeldahl's method, and the average relative error is 1.76%. Using Moore-Penrose matrix, we can not only get the near-infrared spectroscopy analysis model's regression coefficients, but also know their contribution at every wavelength point. Consequently we can understand and explain the physical and chemical significance of the FTNIR whole spectroscopy regression model.

[Study on bone marrow megakaryocytes in children patients with myelodysplastic syndrome].

The study was aimed to observe morphological characteristics and hematopoiesis function of bone marrow megakaryocyte in children patients with myelodysplastic syndrome (MDS), and analyse the cause and mechanism of thrombocytopenia. CD41 McAb immunohistochemical technique was used to detect micromegakaryocytes of bone marrow smear. Plasma clot culture and CD41 McAb immunohistochemical technique were used for the MK-colony forming assay. The colony formations of CFU-MK and BFU-MK were measured. The results showed that there was no significant difference of CFU-MK colony formation rate between groups of MDS and control. But, in 62.5% of children patients the colony formation rate of CFU-MK decreased, in 25% increased, and in 12.5% was normal while BFU-MK formation rate decreased in MDS group significantly. The number of micromegakaryocyte and the positive rate of type I lymphoid micromegakaryocyte were significantly higher than those of the control group. In conclusion, there may be two kinds of megakaryocyte clones in bone marrow of children patients with MDS. One is supposed to be pathologic and potentially malignant micromegakaryocytes, the another may be the normal megakaryocytic precursors. The thrombocytopenia in MDS patients induced by increase of pathologic MK leads to abnormal development and maturation of MK in bone marrow.

[Study on bone marrow megakaryocytes in children patients with idiopathic thrombocytopenic purpura].

To observe the morphological characteristics and hematopoietic function of bone marrow megakaryocyte (MK) in children patients with idiopathic thrombocytopenic purpura (ITP), and to preliminary analyse the cause and mechanism of thrombocytopenia. CD41 McAb immunohistochemical technique was used to detect micromegakaryocyte in bone marrow smear. Plasma clot culture and CD41 McAb immunohistochemical technique were used for the MK-colony forming assay. The results showed that there was no statistical difference of the positive rate of micromegakaryocyte between groups of ITP and control, but type I lymphocyte-like micromegakaryocyte was infrequent. The number of micromegakaryocyte and the formation rates of CFU-MK and BFU-MK in ITP group were significantly higher than those in control group. The normal MK releasing platelet could be easily found in the culture system. The MK colony formation rate was decreased in a patient with chronic ITP. In conclusion, the increment of type II, III, IV micromegakaryocytes is one of pathologic phenomenon of ITP. These small megakaryocytes can develop and mature to normal megakaryocytes in the condition of ex vivo culture. The developmental abnormity of MK is a possible reason for thrombocytopenia among partial patients with ITP, especially the chronic cases.

[A basic research to share Fourier transform near-infrared spectrum information resource].

A method to share the information resource in the database of Fourier transform near-infrared(FTNIR) spectrum information of agricultural products and utilize the spectrum information sufficiently is explored in this paper. Mapping spectrum information from one instrument to another is studied to express the spectrum information accurately between the instruments. Then mapping spectrum information is used to establish a mathematical model of quantitative analysis without including standard samples. The analysis result is that the relative coefficient r is 0.941 and the relative error is 3.28% between the model estimate values and the Kjeldahl's value for the protein content of twenty-two wheat samples, while the relative coefficient r is 0.963 and the relative error is 2.4% for the other model, which is established by using standard samples. It is shown that the spectrum information can be shared by using the mapping spectrum information. So it can be concluded that the spectrum information in one FTNIR spectrum information database can be transformed to another instrument's mapping spectrum information, which makes full use of the information resource in the database of FTNIR spectrum information to realize the resource sharing between different instruments.
