Experimental treatment of malignant gliomas with human monoclonal antibody-drug conjugates.

This study was designed to determine whether the monoclonal antibody-drug conjugates are more potent than the free drugs in killing human glioma cells in vitro and in vivo. The anticancer drugs doxorubicin (DXR) and 4'-epi-doxorubicin (epi-DXR) were separately conjugated to the human monoclonal antibody (mAb) CLNIgG, which binds strongly to human malignant glioma cells. The cytotoxic activity of the mAb-drug conjugates was assessed by a 3H-thymidine assay in vitro. The efficacy, biodistribution and autography of the immunoconuugates were examined using the subcutaneous glioma model (nude mouse). The epi-DXR-CLNIgG conjugate was found to be 11 times more potent than free epi-DXR in killing glioma cells in vitro, and epi-DXR-CLNIgG clearly achieved the most favorable antitumor effects. A biodistribution study using [14-14C]DXR-CLNIgG at the conjugate indicated that the immunoconjugates delivered DXR to glioma tissues at least five times more than the free DXR alone in nude mice without increasing the concentration in other tissues. An autoradiographic study also showed good accumulation of the antibody-drug conjugate in the subcutaneously transplanted glioma. Thus, the human monoclonal antibody-drug conjugates constitute a potential new approach for the treatment of malignant gliomas.

[Effect of swimming on asthmatic children].

Thirteen asthmatic children were enrolled in a swimming program during five days. Pulmonary function test before and after swimming training was evaluated. Training did not change pulmonary function values, and asthmatic children improved swimming distance and enhanced swimming ability. No side effects were observed during study. After asthma can questionnaires indicated a high degree of enthusiasm and acceptance of the program by children and parents.

Simple analytical method for organophosphorus pesticide residues in milk.

A simple method for determination of organophosphorus pesticide residues at the parts per million level in milk was developed. Pesticide residues were extracted with acetonitrile added to aqueous milk, fat was removed by zinc acetate addition and dichloromethane partition, and analytes were concentrated and analyzed by wide-bore capillary column gas chromatography. Recoveries of 6 pesticides spiked in milk samples at levels of 0.1 and 1.0 micrograms/mL were 82.1-93.8% and 79.7-96.6%, respectively. Triplicate samples spiked with 6 pesticides at 1 microgram/mL were analyzed independently by 3 laboratories. Average recoveries were greater than 80%, and the mean coefficients of variation for the complete study were 2.9% for diazinon, 5.4% for dimethoate, 4.6% for malathion, 4.6% for parathion, 4.9% for EPN, and 6.1% for phosalone.

Simple analytical method for organophosphorus pesticide determination in unpolished rice, using removal of fats by zinc acetate.

A rapid and simple method is developed for the determination of organophosphorus pesticides in unpolished rice. The new method incorporated acetonitrile-water (1 + 1) extraction, removal of fats by zinc acetate, and further cleanup on an activated charcoal chromatographic column. The higher fatty acids in the extract react rapidly with zinc acetate to form insoluble zinc carboxylates, which precipitate. Additional interferences were cleaned up on an activated charcoal chromatographic column, and organophosphorus pesticides adsorbed on the activated charcoal were eluted with acetone-hexane. Dimethoate is not retained on the activated charcoal and must be extracted with dichloromethane from the first acetonitrile-water eluate. Pesticides are measured by flame photometric gas chromatography. Recoveries from 50 g unpolished rice samples fortified with 5-50 micrograms diazinon, 6-30 micrograms parathion, 8-40 micrograms fenitrothion and IBP, 10-50 micrograms dimethoate and fenthoate, 20-100 micrograms malathion, or 40-200 micrograms EPN ranged from 75.7 to 95.8%.
