Effects of ethanol and arachidonic acid pathway inhibitors on the effectiveness of gastric mucosa cytoprotection.

Cytoprotection in the stomach, consisting in the mucus secretion, mucous circulation intensification and bicarbonate secretion to the gastric lumen, is highly dependent on the products of arachidonic acid pathway and peroxidative-antioxidative balance. The aim of the paper was to examine the effects of selected inhibitors of arachidonic acid pathway on the natural protective system of the gastric mucosa exposed to 50% ethanol. The results show that leukotrienes, thromboxane and oxygen reactive forms significantly impair the protective function of the gastric mucosa while prostaglandins and antioxidant enzymes act protectively.

Histological changes in placenta villi in some types of pathological pregnancy.

The examinations were carried out on 3 placentas from normal pregnancies and 12 placentas from pathological pregnancies (gestosis, cholestasis, type A diabetes pregnancies and premature outflow of amniotic fluids). Placenta sections were fixed in glutaraldehyde and subsequently embedded in Epon 812. Semithin sections 1 mu thick were stained with methylene blue and azure. Moreover, changes in terminal villi were assessed. In comparison with the control placentas changes in blood vessels, stroma and syncytiotrophoblast were found.

Ultrastructural changes in the syncytiotrophoblast in some types of pathological pregnancy.

The studies were performed on 12 placentas from pathological pregnancies (gestosis, cholestasis, type A diabetes pregnancies, premature outflow of amniotic fluids) and 3 placentas from normal pregnancies. Ultrathin sections were stained by the Reynold's method and evaluated under the electron microscope BS-500 (Tesla). The changes found in syncytiotrophoblast were the following: the increase or the decrease in its thickness, increase in basal membrane thickness, decrease in the number or lack of pinocytic vesicles and superficial microvilli, damage of mitochondria.

The influence of nifedipine (calcium channel blocker) and Bay-K-8644 (calcium channel agonist) on the development of experimental acute pancreatitis.

The aim of this paper was to evaluate the influence of nifedipine (calcium channel blocker) and Bay-K-8644 (calcium channel agonist) on cerulein acute pancreatitis (AP) in rats. AP was induced according to the Lampel and Kern method (1) by the continuous intravenous infusion of cerulein in the doses of 5 x 10(-6) g/kg/h for 12 hours. There was obtained a statistically significant decrease in serum amylase activity and pancreatic weight in the groups treated with higher doses of nifedipine before infusion of the cerulein compared with rats treated only with cerulein. However, in the group treated with Bay-K-8644 before infusion of cerulein statistically significant increase was obtained in serum amylase activity and pancreatic weight compared with the group treated only with cerulein. The investigations suggest a beneficial effect of higher doses of nifedipine on cerulein induced AP. The inflammatory changes in the pancreas in the groups treated with nifedipine observed under the light microscope were smaller than in the group treated only with cerulein.

Ultrastructural changes in the receptor parts of retinal rods in experimental alloxan-induced diabetes in rabbits.

Mature rabbits were administered a single dose of alloxan at the dose 100 mg/kg b.m. After 3 and 6 weeks and after 3 and 6 months, the samples of the retina were taken from the areas immediate to the papilla of the optic nerve. Ultrathin sections were dyed according to the Reynold's method, and the receptive parts of the rods were examined under electron microscope BS-500 Tesla. After 6 weeks following alloxan administration, distinct morphological changes in the form of enlargement of certain discs in the receptive parts of rod cells were observed. After 3 months the majority of the discs was damaged, and after 6 months only single, quite well preserved rod cells were found to be present in the retina.

Alloxan↗

Histological changes in the retina in experimental alloxan-induced diabetes in rabbits.

Mature rabbits were given a single dose of alloxan at the dose of 100 mg/kg of b.m. After 3 and 6 weeks and after 3 and 6 months the retina samples were taken from the areas immediate to the papilla of the optic nerve. Half-thin sections were stained with Swiss blue and azure, and the changes in the particular layers were evaluated under the light microscope. Morphological changes in the form of decreased number of neurones (especially ganglionic) and narrowed layer of rods and cones occurred after 6 weeks. After 3 months even further increase in the number of neurones and atrophy of the pigmented epithelium cells were observed. After 6 weeks nearly total atrophy of ganglionic neurones and distinct narrowing of all the layers were found.

Histological examination of the thyroid after experimental administration of Metizol.

The investigations were carried out on thyroids of white Wistar rats which were given Metizol for 3 days, 3 and 6 weeks at the dose of 1 mg/kg of b.m. for 24 hrs. The samples were stained with hematoxylin and eosin and by the PAS method. The following changes were observed: 1. Three-day administration of Metizol resulted in the decrease in the quantity of colloid; the secretory epithelium became uniform--i.e. cubic; the quantity of the interfollicular tissue increased. 2. Three-week administration of Metizol resulted in appearing of colloid stained differently than in the control group; secretory epithelium cells were of different height (cubic and cylindrical), a considerate congestion of the gland was visible, the quantity of the interfollicular tissue increased. 3. After 6 weeks of Metizol administration the morphological picture of the thyroid became similar to that of the control group.
