HB Handa [alpha 90 (FG 2) Lys replaced by Met]: structure and biosynthesis of a new slightly higher oxygen affinity variant.

Hb Handa, an abnormal hemoglobin which isoelectrofocused anodally to Hb A, was found in a 15-year-old Japanese girl and her mother, who were apparently healthy. Clinical and hematological examinations showed no abnormality in individual heterozygous for this mutant gene except for a slight increase of reticulocyte count (2.1-2.4%). Structural studies disclosed an alpha chain anomaly and substitution of alpha 90 (FG 2) Lys replaced by Met. The content of the abnormal hemoglobin in the hemolysate was 17.5-18.3% of the hemoglobin. The isopropanol precipitation test was negative. The purified abnormal hemoglobin showed a slightly higher oxygen affinity than that of Hb A, but Hill's n constant, Bohr effect and organic phosphate effect were within the normal range. Globin chain biosynthesis in reticulocytes indicated that the production rate of the abnormal alpha chain was suppressed to 38% of the normal level.

Physicochemical changes during haemorrhage and following infusion.

Interstitial fluid pressure (IFP) and plasma colloid osmotic pressure (plasma COP) were measured in dogs following acute haemorrhage and following the infusion of saline, 10% dextra 40 in saline and 3% dextran 40 in lactated Ringer's solution. Exsanguination decreased IFP, plasma COP and total plasma protein and albumin concentrations, and increased plasma glucose concentration and osmotic pressure. A massive infusion of physiological saline increased IFP, and decreased plasma COP and total plasma protein and albumin concentrations. The volume of saline infused correlated with the increase in IFP. When a 10% dextran 40 saline solution was used, there was a marked increase in plasma COP but a decrease in IFP. When 3% dextran 40 in lactated Ringer's solution was infused, IFP was little affected in the early stage, but gradually increased thereafter. Plasma COP increased slightly immediately after infusion, but remained near the pre-exsanguination value for at least 3 h.

Hemoglobin Yusa (beta 21 (B3) Asp leads to Tyr), a new abnormal hemoglobin found in Japan.

An electrophoretically slowly moving hemoglobin variant, Hb Yusa, was found in a 48-year-old man, his younger sister and mother. No clinical or hematological abnormalities were seen in the individuals heterozygous for this mutant gene. Structural studies of this abnormal hemoglobin showed a new type substitution of beta 21 (B3) Asp leads to Tyr. The percentage of the abnormal hemoglobin in the hemolysate was 40.8. The hemoglobin had neither instability nor functional abnormality.

Changes of the cell bodies in the facial nucleus after facial nerve block.

Facial nerve blocks were produced in four monkeys by insertion of a needle into the facial nerve trunk. The sequential changes of nerve cells in the facial nucleus were investigated 4 days, and 2 and 7 months later. Although the facial paralysis recovered in 2 months, considerable changes of cell bodies in the facial nucleus were still obvious which recovered completely only after 7 months. It is postulated that an optimum discharge is generated by those nerve cells in the facial nucleus during the recovery period which in clinical practice could result in 6 months of clinical remission from facial spasm.

Effects of differentially modeled stimuli on vicarious autonomic arousal.

The present study examined effects of variations in modeled pain cues on level of vicarious arousal, using heart rate and blood-volume amplitude as psychophysiological measures. Students were assigned randomly to one of four groups in which they either (a) underwent directly the cold-pressor test, (b) observed a model exposed to the cold-pressor test and expressing pain by facial and bodily movements, (c) heard only the modeled verbal expressions of pain, or (4) observed the model express neither verbal expressions nor physical movements while undergoing the painful stimulation. The results show aversive stimulation increases heart rate whereas vicarious aversive stimulation produces reductions in heart rate. The deceleration of heart rate was not found in response to all three forms of modeling. Both direct and vicarious aversive stimulation produced decreases in the amplitude of vasomotor responses. The size of the decrement between direct stimulation and the minimal modeling condition of vicarious arousal showed significant differences, but differences between direct stimulation and the maximal modeling condition were not significant. The vasoconstriction was found in response to two forms of modeling.
