[Ochratoxin A contamination of cereal grain harvested in 2003 and 2004 years].

The analysis of ochratoxin A--mycotoxin produced by widely distributed Aspergillus and Penicillium--of cereal grain harvested in 2003 - 2004 years were performed by immunoaffinity column clean-up and HPLC with fluorescence detection. This survey examined 282 samples of raw grain wheat, rye, barley and oat 13.8% of all samples contaminated by ochratoxin A in the range 0.2-33.3 mg/kg. Calculation made on the basis of the obtained means showed that the daily ochratoxin A intake of human from cereal grain were from 1.58 to 2.84 ng/kg b.w. Scientific Committee for Food of the European Commission suggested that it was prudent to reduce exposure to ochratoxin A as much as possible below 5 ng/kg bw/day. Codex Alimentarius and European Commission have established maximum permissible level of 5 mg/kg for ochratoxin A in raw cereal grains.

[The method of the simultaneous determination of the citrinin and ochratoxin A content in cereals and cereal products].

The simplifyed method of simultaneous determination of the citrinin (CT) and ochratoxin A (OTA) in cereals is described. The extraction of mycotoxins was carried out by small volumes of solvents (water/aceton/hexane) without additional sample clean up and concentration. The extracts were analysed by HPLC using mixture of methanol/ethylacetate/phosphoric acid, pH 2.2 as a mobile phase and fluorescence detection (lamda ex 330 nm, lamda em 495 nm). The reliability and reproducibility of results were improved by usage of internal standard--methyl-derivative of ochratoxin A. Average recovery of CT and OTA was 70%, relative standard deviation 12% and 7%, respectively, limit of detection 0.003 mg/kg.

[Food supplements and medicines of vegetative origin. Safety estimation and standardization].

The article describes the modern legislative and normative base of both Russian made and foreign made food supplements turnover. The main requirements to them. The reasons for banning the use of some particular ingredients in food supplements formula are substantiated. The ways of improving the normative base for food supplements as well as for medicines of vegetative origin are proposed. The conclusion is made, that this base is more perfect than for the medicines, produced from herbs.

[Complex approach to evaluation of natural quality of coloured juices].

A novel multidimensional approach to estimation of fruit and berry colored juice authenticity is proposed. It includes determination of quality (determination of 5-hydroxymethylfurfurol) and authenticity (determination of organic acids, hydrocarbons, anthocyanin pigments and artificial colorants) parameters. Both experimental and literature data on chemical composition of 12 colored fruits and berries are summarized. New composition characteristics of pomegranate, bilberry, red currant, blackberry and lingonberry are presented.

[5-hydroxymethylfurfural as indicator of quality of juices for child nutrition].

132 samples of domestic and imported juices and juice concentrates for child nutrition were analyzed on content of 5-hydroxymethylfurfurol (5-HMF), which is considered as an indicator of violation of juice manufacturing technology. The concentrations of 5-HMF in 15.9% of domestic juice samples and 10.3% of imported juice and concentrate samples were found higher than 20 mg/L--maximum level established by AIJN for some juices and nectars. The results confirmed the expediency of including of HMF maximum concentration in hygienic characterization of fruit juices and concentrates as an indicator of violation of their manufacturing and storage procedures.

Evaluation of hydromatrix and magnesium sulfate drying agents for supercritical fluid extraction of multiple pesticides in produce.

The simultaneous extraction of relatively polar and nonpolar pesticides has been problematic in multiresidue analysis using supercritical fluid extraction (SFE) with carbon dioxide. In fruit and vegetable samples, which typically contain 80-95% water, moisture acts to increase SFE recoveries of many polar pesticides, but a drying agent should be used to control water in SFE. Hydromatrix, a prevalent drying agent, has many desirable characteristics, but it reduces recovery of certain important pesticides, such as methamidophos, acephate, and omethoate. MgSO4 has been shown previously to be applicable for the extraction of methamidophos and six other pesticides, but MgSO4 has practical disadvantages in its use. In this study, properties and SFE results with the individual drying agents and their combination were evaluated. Simultaneous recoveries for polar and nonpolar pesticides were achieved for 71 pesticides fortified in apple using a mixture of 2 + 1 + 2 MgSO4-H2O-Hydromatrix-sample for extraction. The advantages of each drying agent were maintained by their combination. The analysis of real samples, however, showed that more study was needed to improve recoveries of nonpolar pesticides.

[Determination of histamine and serotonin in whole blood using the method of high performance liquid chromatography].

The authors have developed a method for measuring histamine and serotonin in whole blood, based on reverse-phase high-performance liquid chromatography of 5-dimethylaminonaphthyl sulfonic (DNS) amine derivatives with fluorometric detection. Chromatography in a mini-column packed with polyamide was employed for the isolation and purification of DNS-histamine and serotonin. The lowest detectable level of histamine is 2-2.5 ng/ml of blood, that of serotonin 1-1.5 ng/ml. The method was tried with whole blood samples from children suffering from various allergic conditions. Whole blood histamine and serotonin concentration were found particularly elevated in the patients with gastrointestinal allergies.

A survey using normal-phase high-performance liquid chromatography of aflatoxins in domestic and imported foods in the USSR.

A survey of the occurrence of aflatoxins B1, B2, G1 and G2 in Soviet domestic and imported cereals and nuts (totalling 4532 samples) collected in 1985-87, showed that 26.9% of imported peanuts, 2.2% of corn and 28.3% of cotton seeds were contaminated by aflatoxins at levels exceeding the maximum tolerated level established in the USSR (5 micrograms/kg for aflatoxin B1 in foodstuffs of all types excluding baby foods); maximum concentrations were 3650, 600 and 153 micrograms/kg, respectively. A highly sensitive normal-phase high-performance liquid chromatographic method was developed. The detection limit was 0.1 microgram/kg and the coefficients of variation were 11% and 8.5% at contamination levels 10 and 100 micrograms/kg of aflatoxin B1, respectively.

A survey of the occurrence of deoxynivalenol in wheat from 1986-1988 harvests in the USSR.

The occurrence of deoxynivalenol (DON; vomitoxin) in wheat from 1986-1988 harvests in the USSR was surveyed. A significant frequency of DON contamination (81.3% of samples analyzed) was observed. A correlation between levels of DON contamination and percentage of Fusarium-damaged kernels was demonstrated. It was shown that DON contamination did not exceed the maximum tolerated level (MTL) established in the USSR (1.0 mg/kg) if samples contained no more than 0.6% of Fusarium-damaged kernels.

[Optimization of analytical methods for assay of ochratoxin A in the foodstuffs].

Ochratoxin A is a mycotoxin produced by widespread mold fungi of the genera Aspergillius and Penicillium. It is a common contaminant of cereals, coffee, wine, dried fruits, and spicery. There is evidence that ochratoxin A has nephrotoxic, immunosuppressive, embryotoxic, teratogenic, and carcinogenic effects on many species of mammals. Its maximum allowable concentration established by the Alimentarius Code and the European Union is 5 microg/kg for cereals and 3 microg/kg for ready-made cereals. Two optimized techniques (silica gel column chromatography (CC) or immunoaffinity CC for extract purification and high performance liquid chromatography with fluorescence detection) are proposed for the detection, identification, and assay of ochratoxin A in food commodities. The detection limit was 0.05 and 0.5 microg/kg, respectively. The developed techniques were used to assay the content of ochratoxin A in 46 samples of the grain gathered in different regions of Russia. Nine samples were contaminated by ochratoxin A at concentrations of 0.05 to 5 microg/kg.
