Comparison of autoradiography and DNA histograms in assessing S-phase cells from patients with myeloid leukemias.

The studies reported here were designed to determine whether DNA histogram analysis provided the same estimate for the percentage of cells in S-phase as does 3H-TdR labeling index (LI). For DNA histogram analysis high resolution flow cytometry was employed together with 3 different computer based methods for histogram analysis. When tissue culture cells were employed there was a good correlation (r = 0.89) between the estimates for the percentage of cells in S-phase by the LI and DNA histogram techniques. In contrast, when 1138 bone marrow or peripheral blood specimens obtained from leukemic patients were studied the correlation was poor. For example when 243 pretherapy bone marrow aspirate cells were studied by both methods the correlation was only 0.46. A direct comparison of the clinical relevance of the two methods for measuring S-phase cells when high dose cytosine arabinoside therapy was used to treat patients demonstrated that only LI measurements provided clinically useful information. In conclusion, DNA histogram analysis provides an inaccurate estimate of the percentage of cells in S-phase.

Natural killer (NK) activity in normal donors undergoing leukapheresis.

The effect of leukapheresis performed with intermittent flow blood separator on normal donors NK activity is presented. There was statistically significant decrease of NK activity immediately after leukapheresis (p less than 0.001) which did not return to predonation values 1 day after leukapheresis (p less than 0.05), however, none of the donors values fell outside the normal range. It seems that NK cells are not as quickly replaced as T lymphocytes.

Comparison of in vitro assays for detecting subpopulations of P388 leukemic cells resistant to AraC.

The kinetics of cytosine arabinoside (AraC) were studied in vitro using P388 murine leukemia lines sensitive (P388-S) and resistant (P388-R) to the drug. Using P388-S cells, the 3H-thymidine index (L.I.) was paralleled by the 3H-AraC index, i.e. any cell in the S-phase of the cycle also incorporated the 3H-AraC into DNA. With the resistant line, the 3H-AraC index was zero in spite of high L.I. This discrepancy between the L.I. and 3H-AraC index could be used to predict the percentage of resistant cells when known mixtures of sensitive and resistant populations were used. The reason for lack of incorporation into DNA by the resistant cells was shown to be the presence of very low levels of the enzyme deoxycytidine kinase in P388-R cells resulting in an inability to phosphorylate AraC to its active form AraCTP. Immediate inhibition of DNA synthesis caused by AraC was directly proportional to the number of sensitive cells present in the population, but was not as accurate a predictor of sensitivity as the 3H-AraC index. Cloning in methylcellulose was found to be the least sensitive predictor of the percentage of sensitive and resistant cells, most likely related to the difference in cloning efficiency of the sensitive and resistant lines.

Examination of leukaemic blasts sensitivity to cytostatic drugs using two short-term laboratory methods.

The authors examined acute leukaemia cells sensitivity to anticancer drugs using 2 short-time methods: 1. Measurement of the inhibition of 3HTdr incorporation during 2 h incubation with cytostatic; 2. Evaluation of the cell viability in short-term (5 days) liquid culture. The 37% decrease in living cells count or in the rate of 3HTdr incorporation has been evaluated as indicating sensitivity to examined drugs. Out of many determinations performed by these use of 2 mentioned methods, those executed simultaneously were compared. Comparable results were obtained to arabinoside-cytosine testing, showing ca. 94% of sensitive cases. In relation to vincristine the 3HTdr incorporation method showed more often - in 82% of cases - sensitivity as compared to only 46% tested in culture method, whereas to daunorubicine more common effect on cultured cells viability (86%) than on tritiated thymidine incorporation (40%) could be observed. These differences remain to be elucidated as to their technical or biological sources. The sensitivity to L-asparaginase was shown only in about the half of cases by both methodes.

[Lead concentration in the blood and aminolevulinic acid dehydratase (ALAD) activity in the erythrocytes depending on sex, age, tobacco smoking and alcohol drinking in the group of persons exposed to industrial dust].

A population of 399 persons (180 women and 219 men) has been examined. Anamnesis included detailed inquiries about smoking habit and alcohol drinking. In the laboratory, lead concentration in blood and ALAD activity in erythrocytes have been determined on empty stomach. No differences have been found in the mean lead concentration determined by sex, whereas the mean ALAD activity is higher in women than in men. The subjects' age has affected the test parameters neither in men nor women. In smoking men no changes in the mean lead concentration in blood and mean ALAD activity in erythrocytes hav been found. In smoking women, the mean lead concentration is not changed, but the mean ALAD activity is lower. Alcohol drinking in men does not change the values of the test parameters. Whereas drinking women have revealed higher mean blood lead concentration.

[Hematological studies of the populations of Lower Silesia living in different ecological conditions].

Haematological investigations were carried out in different population groups in Lower Silesia. The total material investigated included 439 men and 229 women. The investigations demonstrated that the haemoglobin level, erythrocyte count haematocrit index were lower in the population of villages lying around copper processing plants. The mean reticulocyte count exceeded significantly that in the control group. The highest parameters of erythrocyte system were found in the group of men from a large city despite their occupational exposure to exhaust gases. The leucocyte count was 6-7000/mul in all groups with the exception of the urban group in which it was 1000-1500/mul lower than the mean values in the remaining groups. Lymphocytosis was observed most frequently in this group. The highest mean percent of granulocytes and the lowest incidence of lymphocytosis were observed in the control group from a rural population in a non-industrialized area while rural groups around industrial centres showed intermediate values. In the female group from a locality exposed particularly to industrial pollution low platelet count was frequent.

Interleukin-17 in acute myeloid leukemia.

There are several reports that angiogenesis plays important roles in hematological malignancies including acute myeloid leukemia (AML). Human interleukin-17 (IL-17) is a proinflammatory cytokine produced by activated CD4 T cells. IL-17 plays a potential role in T cell mediated angiogenesis. The role of IL-17 in pathologic angiogenesis has not been evaluated yet. The aim of the study was to determine plasma level of IL-17 in patients with AML. IL-17 levels were measured by ELISA in plasma samples taken from 68 adult patients with AML before chemotherapy was administered. In addition 20 out of 68 patients were reanalysed after achieving complete remission (CR). Ten samples from healthy volunteers were evaluated as the control. In this study we have demonstrated that serum level of IL-17 is not elevated in AML patients. These results suggest that angiogenesis in AML is not mediated by CD4 T cells. To our knowledge this is the first report about IL-17 serum level in acute leukemias. We are currently evaluating IL-17 levels in others haematological malignancies.
