[Cytogenetic changes in Ph(+) chronic granulocytic leukemia under the influence of interferon alpha (IFN-alpha) therapy].

Chromosomal changes during therapy with IFN-alpha were analysed in 21 patients suffering of chronic granulocytic leukemia. Complete or major cytogenetic response (CgR) was obtained in 4 patients, minor in 5 minimal in 7, and no response in 5 patients. Patients showing a low Sokal index more often disclosed CgR. In 6 persons additional chromosomal aberrations were present at diagnosis or during the disease course. They disappeared on IFN-alpha therapy. This fact may be in favour of the influence of IFN-alpha not only on disappearance of Ph chromosome, but also of secondary aberrations, some of them indicating the possibility of an acceleration of the disease. No relation of the CgR, as well as Sokal index value to the survival time in presented small cohort could be observed, which may depend on short observation time. CgR did not depend on a type of fusion of BCR/ABL gene. However, survival time was longer in patients with b3a2 fusion.

[Observation of D-dimer levels in serum of patients with acute leukemia].

The blood coagulation and fibrinolysis disorders are common complications observed in patients with acute leukemias, particularly in acute myelogenous leukemia. These abnormalities are mediated by thromboplastic substances released from the blast cells and the alteration of hemostatic properties of vascular endothelium. The plasma concentration of D-dimer (cross-linked fibrin degradation products), measured by enzyme immunoassay, using monoclonal antibodies, serves as a specific marker of the coagulation activation and fibrinolysis system. In our study, the plasma concentration of D-dimer was investigated in 142 patients with acute leukemia during clinical course--at the time of initial diagnosis, complete remission, relapse or in cases resistant to chemotherapy. It has been revealed that, at the time of initial diagnosis, the plasma level of D-dimer was elevated in most patients, irrespective of the type of acute leukemia. However, the initially elevated plasma concentration of D-dimer was significantly lower when complete remission had been achieved. Furthermore, in the majority of cases of relapse or resistance to chemotherapy, a further increase of plasma concentration of D-dimer is commonly observed.

Lactoferrin-induced up-regulation of zeta (zeta) chain expression in peripheral blood T lymphocytes from cervical cancer patients.

Alteration of T-cell-associated signal transduction molecules has recently been implicated in immune suppression in tumour-bearing hosts. Here we report the immunoregulatory effects of human lactoferrin (LF) on zeta-chain expression in peripheral blood T lymphocytes from cervical cancer patients and healthy donors. By quantitative flow cytometry analysis, we demonstrated that the mean zeta-chain expression was significantly higher in freshly-isolated T lymphocytes from healthy donors (69%), compared with the patients (38%). Following 3-day culture under standard conditions, zeta-chain expression in T lymphocytes from the patients increased significantly, whereas it dropped in the cells from healthy donors. Anti-CD3 MoAb as well as LF, significantly increased expression of zeta-chain in T cells both from patients and control subjects. The addition of LF to the anti-CD3 MoAb cell cultures resulted in an even higher stimulation of the zeta-chain expression. The results suggest that, in patients with cervical cancer, zeta-chain defects could be corrected by the therapeutic application of LF.

[Activity of natural cytotoxic cells in non-Hodgkin's lymphoma].

Cytotoxic activity of NK cells in the peripheral blood has been determined in 30 patients with malignant non-Hodgkin lymphomas prior to and following therapy. In the whole group as well as in subgroups classified according to the criteria of Working Formulation as lymphomas of the low, moderate and high degree of malignancy, activity of NK cells has been statistically significantly lower than that in healthy individuals. Marked increase in this activity has been noted in 19 patients in the state of clinical remission after the treatment with cytotoxic agents, and sometimes radiotherapy. The value of mean cytotoxic activity reached normal limits in the lymphoma of high degree of malignancy, and exceeded these limits in the lymphomas of moderate and low malignancy.

[Effect of brown enriched Livex on the general condition of patients with proliferative diseases of the bone marrow and lymphatic system].

Brown Livex, a preparation of animal blood in doses of 5.0 g twice daily was given to 33 patients with proliferative diseases of bone marrow and lymphatic system during and after treatment with cytostatic agents. The course of treatment was 3 months. The effect of the preparation was studied on the wellbeing of the patients, body weight, general fitness (Karnofsky scale) and the levels of haemoglobin, haematocrit and beta-lipoproteins. The control groups comprised 20 patients with similar diseases and similar cytostatic treatment. In the studied group a positive effect was observed of the preparation on weight gain (26 out of 33 patients), well-being (24 of 33) and general fitness (17 of 33). In the control group during the same-time of observation weight loss was noted in 11 of 20 cases, well-being deterioration in 7 of 20, and decreased general fitness in 7 of 20. Livex treatment had no effect on haemoglobin level, haematocrit value, total serum protein, albumins and beta-lipoproteins.

[Richter's syndrome and cervical cancer in the course of a rheumatoid arthritis].

The authors describe a rare case of Richter's syndrome (transition of chronic lymphatic leukaemia into reticulosarcoma) developing during rheumatoid arthritis in a women aged 53 years. Chronic antigenic stimulation and disturbances of interaction between lymphocytes T and B are suggested as a cause of Richter's syndrome.
