Comparative measurements of urinary polyamines in early morning and 24-hour urine specimens.

Detailed comparative measurements of urinary polyamines were made between an early morning spot urine and 24-hour urine from normal controls and from patients with cancers. Urinary putrescine and spermidine concentrations in samples of morning spot urine were similar to those in 24-hour urine samples, and they were significantly increased in patients with various types of cancer. A morning spot urine can replace 24-hour urine in polyamine assay.

Urinary 4-hydroxy-3-methoxymandelic (vanillylmandelic) acid, 4-hydroxy-3-methoxyphenylacetic (homovanillic) acid, and 5-hydroxy-3-indoleacetic acid determined by liquid chromatography with electrochemical detection.

We describe a simple liquid-chromatographic assay of urinary 4-hydroxy-3-methoxymandelic (vanillylmandelic) acid, 4-hydroxy-3-methoxyphenylacetic (homovanillic) acid, and 5-hydroxy-3-indoleacetic acid with electrochemical detection, with direct injection of the sample. The first two analytes are measured simultaneously; 5-hydroxy-3-indoleacetic acid is measured separately. Chromatographic conditions for assay of the three were: column temperature, 65 and 60 degrees C; mobile phase, potassium phosphate buffer (0.2 mol/L, pH 3.0) for 6 min, then potassium phosphate buffer plus acetonitrile (9/1 by vol) for 20 min; flow rate, 0.7 mL/min; oxidation potential, 600 and 450 mV vs an Ag/AgCl reference electrode; and sensitivity, 40 and 160 nA at full scale. Values so obtained agreed well with those obtained for samples that were first solvent-extracted.

Effect of arterial infusion of bleomycin on esophageal carcinoma--an evaluation by nuclear cytophotometry.

In order to determine the histological changes after a single infusion of bleomycin (BLM) into the aortic esophageal artery (BLM A.I.), resected specimens from 58 patients with esophageal carcinoma (27 cases with BLM A.I., 14 untreated cases, 13 cases with radiation therapy and four cases with combined therapy of radiation and BLM) were examined and evaluated by Feulgen microspectrometry for nuclear deoxyribonucleic acid (DNA) content. The histological changes following the BLM A.I. were characterized by degeneration and necrosis at the front of the invading carcinoma tissue accompanied by inflammatory cell infiltration with foreign body giant cells and fibrosis. In comparison with untreated cases, BLM-treated cases showed an increase in nuclear DNA content and a wide dispersion of the DNA values especially at the perinecrotic area and at the front of advancing carcinoma nests. Therefore, a histological effect of the BLM A.I. was shown by the measurement of the nuclear DNA content of carcinoma cells.

Binding of Ulex europaeus agglutinin-I in polyposis coli: comparative study with solitary adenoma in the sigmoid colon and rectum.

Our previous lectin-binding study of the human colorectal mucosa revealed that, in the distal large bowel, Ulex europaeus agglutinin-L (UEA-l) binding was detectable in adenocarcinomas and adenomas, whereas it usually was not found in the normal epithelium. The aim of the present study was to obtain further information about the chemical nature of the constituents of the epithelium in polyposis coli and solitary colorectal adenoma. UEA-l binding to demonstrate alpha-L-fucosyl residue-containing glycoproteins was conducted on entire colon specimens from 4 patients with polyposis coli and on polypectomy specimens from 23 patients with solitary adenomas of the sigmoid colon and rectum with the use of a highly sensitive lectin-antilectin immunoperoxidase method. In the rectosigmoid area of all 4 patients with polyposis coli, positive UEA-l staining was observed not only in the adenomatous epithelium but also in the nonneoplastic epithelium intervening between the polyps. In contrast, in 18 of 23 patients with solitary adenomas, positive UEA-l staining was observed only in the adenomatous epithelium but not in the adjacent nonneoplastic epithelium. In the remaining 5 solitary adenomas, UEA-l staining was shown neither in the adenomatous epithelium nor in the adjacent nonneoplastic epithelium.

[Carcinoma of the colon coexisting with tuberculosis--report of 2 cases].

Two cases of colon carcinoma coexisting with tuberculosis are reported. Both carcinoma were adjacent to scarring areas of ulcerative tuberculosis, these areas were covered by regenerated mucosa, the muscularis mucosae were lacking and there was occurrence of atypical regenerative tubules and adenomatous glands. These findings suggest that ulcerative lesions of tuberculosis may be precursors of the carcinomas. It is interesting that such carcinomas derived from a chronic inflammatory process with repetition of erosion, ulcer and consequent regeneration.

Urinary p-aminobenzoic acid determined in the pancreatic function test by liquid chromatography, with electrochemical detection.

We describe a liquid-chromatographic procedure for determining urinary p-aminobenzoic acid (PABA) in the exocrine pancreatic function test. The urine specimen, collected for 6 h after oral administration of N-benzoyl-L-tyrosyl-p-aminobenzoic acid, is hydrolyzed in 4 mol/L NaOH containing m-hydroxybenzoic acid as internal standard, to free PABA metabolites, then diluted and injected into the chromatograph. PABA and the internal standard are eluted from a reversed-phase C18 column with phosphate buffer (0.2 mol/L, pH 3.5)/acetonitrile, 87.5/12.5 (by vol), and detected with an electrochemical detector at +1100 mV. Urinary PABA concentration as measured by this method agreed well with that by colorimetry involving p-dimethylaminocinnamaldehyde. The present procedure is specific for PABA, precise, and fairly rapid. We hope that the present method will minimize instances of interference from drugs, which cannot be avoided in the colorimetric method.

Improved analysis for urinary polyamines by use of high-voltage electrophoresis on paper.

This method for assay of polyamines (putrescine, spermidine, and spermine) in large numbers of urine samples is based on preliminary analyte isolation by use of a Dowex 50W column, separation by high-voltage paper electrophoresis, reaction with ninhydrin to detect the separated amines, and subsequent direct assay with a dual-wave-length thin-layer chromatography scanner by the zigzag scanning method. By optimizing the conditions for the Dowex 50W column and high-voltage paper electrophoresis, putrescine, cadaverine, spermidine, and spermine were completely separated. The major technical improvements are room-temperature collection of urine with toluene, shorter hydrolysis time, complete separation of putrescine from cadaverine and histamine, and elimination of interference with spermine measurement. Polyamine concentrations as measured by this method agreed with those obtained by use of an amino acid analyzer. Reference intervals by this method were similar to those reported by Russell (Clin. Chem. 23: 22, 1977) with an automated amino acid analyzer. Putrescine, spermidine, and spermine excretion in urine from patients with blood cancers and solid cancers was significantly increased; information on concentrations of putrescine and spermidine were especially useful for following the clinical efficacy of cancer treatments.

Increase of urinary putrescine in 3,4-benzopyrene carcinogenesis and its inhibition by putrescine.

A significant increase in putrescine was noted in the urine of mice with experimental s.c. tumors induced by a single injection of 3,4-benzopyrene solution (2.52 mg of 3,4-benzopyrene in 0.5 ml of tricaprylin). When 10 mg of putrescine were added to the 3,4-benzopyrene solution, the development of tumors was completely inhibited and the increase of urinary putrescine in mice was suppressed simultaneously. Animal weight data of a control group receiving only putrescine indicated that the inhibitory effect of putrescine is not due to its toxicity.

Urinary putrescine, spermidine, and spermine in human blood and solid cancers and in an experimental gastric tumor of rats.

An improved method of assay of urinary polyamines (putrescine, spermidine, and spermine) was applied to the study of cancer patients and an experimental gastric tumor of rats. Although total polyamines (putrescine, spermidine, and spermine) in urine of patients with blood and solid cancers were significantly high, putrescine concentrations also increased significantly and were shown to be of diagnostic aid even in solid cancers. A significant increase in putrescine was also noted in the urine of rats with experimental stomach tumors induced by N-methyl-N-nitro-N'-nitrosoguanidine.
