Linear relationship between lethal mutation yield and intake of ethyl methanesulfonate in Drosophila melanogaster.

Males of Drosophila melanogaster were fed sucrose solutions containing various concentrations of EMS (from 0.25 to 10 mM) for 24 hr. To measure the intake of an EMS solution, 3H-labeled sucrose was added to the feeding solution, and the 3H activity inside the flies was used as a measure for the intake volume of EMS solution. Each absorbed dose of EMS was estimated from the intake volume of an EMS solution multiplied by its EMS concentration in the feeding solution in a way similar to that described in a previous report [Ayaki et al, 1984]. The relationship between the estimated absorbed dose and the exposure concentration was almost linear in a low concentration range but became concave with a downward curvature in a high concentration range. The dose-response relationship between the frequency of sex-linked recessive lethals and the estimated absorbed dose showed no deviation from linearity at all the five absorbed doses tested. It may be concluded that the absorbed doses thus estimated were very close to true absorbed doses, indicating the usefulness of the present method for dosimetry of chemicals to be given to flies.

Scanning electron microscope study on the biodegradation of IOL and suturing materials.

The biodegradation of the IOLs (27 cases: mainly prepupillary lens with pupillary fixation, irido-capsular and angle supported lens and a J loop lens for posterior chamber lens) and suturing materials of four cases have been studied by Scanning Electron Microscope (SEM). The longest duration in the eye was 7.5 years and the shortest 0.3 year. The materials used for optics were PMMA with the exception of one case of glass. The materials for haptics were Nylon 6, Isot.PP, PMMA, PVDF, Polyimid and Pt-Ir. The suturing materials were Nylon 6 and Isot.PP. Nylon 6 degraded in the eye and Isot.PP degraded in the corneal tissue. But no biodegradation was observed in Isot.PP, PMMA, PVDF and Pt-Ir in the eye. In our investigations, biodegradation has some relationship to age (over 50 years old), duration (within 7.5 years) and inflammation of the eye, but ultimately biodegradation will depend on the individual situation. Degradation occurs most easily in the curved portion of Nylon 6.

Clinical investigations on the pharmacology of azosemide (SK-110) in comparison with furosemide in healthy volunteers.

In this study, 60 mg of Azosemide and 40 mg of Furosemide were each administered to 2 volunteer persons. The daily urine volume after administration of both drugs was almost the same. Azosemide didn't show such rapid diuretic effect as was seen in Furosemide, but Azosemide proved to have a mild diuretic and protracted effect as compared with Furosemide. The influence on a daily urinary and fecal excretion of electrolyte and the decrease of serum electrolytes proved to be the same in administration of either drug. However, in the case of Furosemide, most of the electrolytes were excreted within 3 hours after administration, while Azosemide showed mild and prolonged excretion. The effect of both drugs on serum uric acid was almost the same showing a slightly rising trend.

Anterior pars plana vitrectomy for phakic malignant glaucoma.

Phakic malignant glaucoma was seen in five eyes of four patients: it occurred in four eyes after trabeculectomy and in one eye after peripheral iridectomy. They were treated by anterior pars plana vitrectomy combined with intracapsular lens extraction through a corneal incision. In all eyes the anterior chamber regained its normal depth, and the intraocular pressure could be controlled by medical treatment in three eyes. The remaining two eyes required further filtering procedure and subsequently the intraocular pressure was controlled by medical treatment. No serious complications were encountered throughout the postoperative follow-up period of 7-26 months.

Integration of bovine leukemia virus DNA in the genomes of bovine lymphosarcoma cells.

Integration of bovine leukemia virus (BLV) in the genomes of infected cells was investigated in cattle with enzootic bovine leukosis (EBL) and sporadic bovine leukosis (SBL). Southern blot hybridization of BLV cDNA to Eco RI and Xba I restriction fragments of EBL tumor DNAs revealed that: 1) one to four or more copies of proviral DNA were integrated per genome; 2) the restriction pattern of the integrated proviral DNA was the same in two or three different tumors from the same animals; and 3) different patterns were observed among tumors from four different animals. These findings suggest the monoclonal origin of different tumors in an individual animal and the existence of multiple chromosomal integration sites of BLV provirus. DNAs from several SBL tumors were also analyzed with the same restriction enzymes, but with both representative and cDNA3'-enriched's of BLV RNA. No hybridization bands reactive with representative BLV cDNA could be detected, while several bands appeared to hybridize with cDNA3'-enriched.
