The presence and quantity of protein A in Staphylococcus aureus 147 isolates from the tonsils of healthy pigs were examined by three methods. Cell-bound protein A was detected in 71 (48%), 104 (71%) and 123 (84%) of 147 isolates by the slide hemagglutination test, microplate hemagglutination test and enzyme-linked immunosorbent assay (ELISA), respectively. Extracellular protein A was not detected in any isolates by the microplate hemagglutination test. When the quantity of cell-bound protein A in the isolates was determined by the ELISA, most of the isolates contained about 0.8 to 2.2 micrograms of protein A/ml in bacterial cell suspensions of a concentration of MacFarland No. 3.
Two hundred eighty-two students at Arizona State University in the U.S. and 339 students at Okayama University in Japan completed a questionnaire on their alcohol use, expectancies of the effects of alcohol on their own and others' moods and behaviors, the desirability of these effects, norms of significant others for levels of alcohol use and the subject's desire to comply with these norms, and reasons for drinking and not drinking alcohol. Although frequencies of current drinkers versus abstainers did not differ between the two samples, the U.S. students began regular alcohol use at a significantly earlier age, currently drank more alcohol, had higher alcohol expectancies for emotional responses, and endorsed more celebratory reasons for drinking than their Japanese counterparts. U.S. students, however, had lower expectancies for flushing and lower perceived norms for drinking. Hierarchical multiple regressions performed using data from the current drinkers indicated that expectancies of disinhibition and especially aggressiveness after alcohol use, alcohol norms, celebratory (but not pathological) reasons for drinking, and reasons for not drinking were more predictive of reported levels of alcohol use among the U.S. students as compared with the Japanese students.
We report a 26 year-old male patient who had floaters and hyperemia in his left eye following uveitis due to gnathostoma that had migrated into the vitreous cavity. Severe iridocyclitis and mild opacity of the vitreous body were observed, together with whitish-yellow subretinal tracks accompanied by dot and blot hemorrhages in the fundus. Slit lamp microscopic examination revealed a worm which writhed in the vitreous cavity. We performed vitrectomy to remove the worm from the anterior vitreous uneventfully, followed by prompt subsidence of the inflammatory signs. The worm was identified as a third instar larva of Gnathostoma doloresi. Eosinophilia and creeping eruption did not appear throughout the follow-up period. The patient was accustomed to eat live roaches and whitebait, as well as sliced raw beef liver.
A 43-year-old female was admitted to our hospital for high blood pressure control. She showed hypokalemia with increased urinary potassium excretion, high plasma aldosterone concentration (33.9ng/dl, normal range; 2-12ng/dl) and reduced plasma renin activity (trace, normal range; 0.83-5ng/ml/hr). Bilateral adrenal tumors were revealed by abdominal computed tomography, and left adrenalectomy and right partial adrenalectomy were performed. Cytochromes p-450 and other enzymes involved in aldosterone synthesis were found predominantly in the tumor portions but not in the zona glomerulosa of the attached adrenals, which histopathologically showed paradoxical hyperplasia. This was a rare case of bilateral aldosterone-producing adrenal adenomas, which we could differentiate from idiopathic hyperaldosteronism by employing immunohistochemical analysis of steroidogenic enzymes.
Although the anatomical results of diabetic vitrectomy have been reported to be rather good when silicone oil is used as a tamponade substitute, the functional results remain unsatisfactory. To investigate the factors influencing the functional results in these cases of severe diabetic retinopathy, the correlation between the surgical results and the overall condition of these patients, including the tamponade substitute, was examined. The vitrectomy results obtained in cases of diabetic retinopathy treated using silicone-oil tamponade were studied retrospectively in comparison with those obtained using SF6 gas tamponade or without any tamponade substitute. The visual acuity of more than 80% of eyes belonging to the groups treated with SF6 gas or without any tamponade improved postoperatively, whereas the rate of improvement in the silicone-oil group was limited to low and the postoperative vision ranged widely, since the silicone was usually used in much more severe cases. Iris rubeosis that occurred after pars plana vitrectomy subsided following secondary silicone-oil injection into the vitreous cavity. Only one disadvantage in using silicone oil was that an additional operation was required to remove it, and the visual acuity of nearly 30% of the eyes deteriorated after its removal. As far as the general condition of these patients is concerned, the visual outcome of subjects who had hypoalbuminemia and anemia due to renal dysfunction was poor as compared with that of patients who did not have these disorders. Blood transfusion did not lead to better function.(ABSTRACT TRUNCATED AT 250 WORDS)

The cDNAs for two DNA binding proteins of BTE, a GC box sequence in the promoter region of the P-450IA1(CYP1A1) gene, have been isolated from a rat liver cDNA library by using the BTE sequence as a binding probe. While one is for the rat equivalent to human Sp1, the other encodes a primary structure of 244 amino acids, a novel DNA binding protein designated BTEB. Both proteins contain a zinc finger domain of Cys-Cys/His-His motif that is repeated three times with sequence similarity of 72% to each other, otherwise they share little or no similarity. The function of BTEB was analysed by transfection of plasmids expressing BTEB and/or Sp1 with appropriate reporter plasmids into a monkey cell line CV-1 and compared with Sp1. BTEB and Sp1 activated the expression of genes with repeated GC box sequences in promoters such as the simian virus 40 early promoter and the human immunodeficiency virus-1 long terminal repeat promoter. In contrast, BTEB repressed the activity of a promoter containing BTE, a single GC box of the CYP1A1 gene that is stimulated by Sp1. When the BTE sequence was repeated five times, however, BTEB turned out to be an activator of the promoter. RNA blot analysis showed that mRNAs for BTEB and Sp1 were expressed in all tissues tested, but their concentrations varied independently in tissues. The former mRNA was rich in the brain, kidney, lung and testis, while the latter was relatively abundant in the thymus and spleen.(ABSTRACT TRUNCATED AT 250 WORDS)
Mouse-human heterohybrids (M X H) were constructed and compared with other cell lines (human or mouse) as parental cells to obtain hybrids secreting human monoclonal antibody (MoAb). One of the M X H lines, HM-5, was far superior to the others and useful for establishing hybrids secreting human MoAb. Using HM-5 as a parental cell line, we have obtained 2 hybrids secreting human anti-tetanus toxoid MoAb with neutralizing activity and a hybrid secreting human anti-hepatitis B virus surface antigen (HBsAg) MoAb which recognizes the a-determinant of HBsAg.
