Subpopulations of GABAergic neurons containing parvalbumin, calbindin D28k, and cholecystokinin in the rat hippocampus.

The possible coexistence of calbindin D28k with parvalbumin and of calbindin D28k with cholecystokinin was studied in nonpyramidal cells of the rat dorsal hippocampal formation. Neighbouring Vibratome sections were immunostained either for calbindin D28k and parvalbumin or for calbindin D28k and cholecystokinin. The cells, halved during sectioning, were identified in both sections immunostained for different antigens. The coexistence of calbindin D28k and parvalbumin in the same neuron was rare throughout the hippocampal formation with the exception of stratum oriens of the CA1 region, where 9.6% of the parvalbumin-immunoreactive cells also contained calbindin D28k. In stratum radiatum of the CA3 region, calbindin D28k and cholecystokinin coexisted in 12.5% and 21.2% of the calbindin D28k and cholecystokinin-immunoreactive cells, respectively. In other regions of the hippocampal formation, the two markers coexisted in less than 5% of the cells of either type. The present results demonstrate that calbindin D28k-, parvalbumin- and cholecystokinin-containing nonpyramidal cells represent largely nonoverlapping cell populations and may thus be involved in different inhibitory circuits.

Septal GABAergic neurons innervate inhibitory interneurons in the hippocampus of the macaque monkey.

The septohippocampal projection was visualized in three Macaca mulatta monkeys by anterograde transport of Phaseolus vulgaris leucoagglutinin. Following injections of the lectin into the medial septal nucleus, P. vulgaris leucoagglutinin-labelled fibres were found in the hippocampal complex, mainly in stratum oriens of the CA1 subfield, throughout the CA3 subfield, and in the hilus and stratum moleculare of the dentate gyrus. The majority of labelled axons were varicose, and formed multiple contacts with cell bodies and dendrites of calbindin D28k- and parvalbumin-immunoreactive non-pyramidal cells. GABA immunoreactivity of P. vulgaris leucoagglutinin-labelled axons and their postsynaptic targets was investigated by sectioning varicose axon segments for correlated light and electron microscopy, and processing alternate ultrathin sections for postembedding immunogold staining for GABA. All P. vulgaris leucoagglutinin-labelled boutons examined were GABA-immunoreactive and the majority of them formed symmetrical synapses with GABA-immunoreactive cell bodies and dendrites. The results demonstrate that a GABAergic septohippocampal pathway exists in the monkey, and, similar to the rat, terminates on different types of GABAergic neurons, including the parvalbumin- and calbindin D28k-containing non-pyramidal cells.

Reconstituted Newcastle disease virus envelopes as a split vaccine.

Isolated envelope proteins of Newcastle disease virus (NDV) were inserted into the lipid bilayer of artificial vesicles to create a viral envelope-like structure. The structure-containing viral antigens at high density elicited a strong immune response, in contrast to purified viral proteins. The artificial envelopes or immunosomes possessed several advantageous properties when used as vaccines. They elicited a faster response and the immunity lasted longer in animals treated with these vesicles than in controls vaccinated with envelope proteins in combination with traditional adjuvants. A further useful feature of the artificial envelopes is that no inflammatory lesions develop at the site of their injection.

[Ruptured myocardial infarcts].

Forty one cardiac rupture complicating acute myocardial infarction (AMI) were studied in a ten-year period. The anterior AMIs are most likely to be observed with ruptures. The interval from the onset of AMI to the clinical detection of rupture was short (acute period). Some factors such as physical work in the acute period and transmural infarctions strongly infiltrated with leukocytes cause a higher frequency of rupture. Diabetes and hypertension might be predisposing factors.

Dark and photoreactivity of 4'-aminomethyl-4,5',8-trimethylpsoralen with T7 phage.

The dark and photoreactions of 4'-aminomethyl-4,5',8-trimethylpsoralen (AMT) with T7 phage were investigated from biological and structural points of view. The dark reaction leads to the structural destabilization of the double helix of the DNA as is shown by optical melting measurements. The genotoxicity of AMT in the dark is comparable with that of known genotoxic drugs as determined by phage inactivation. The photoreaction with UVA light leads to the formation of mono- and di-adducts depending on the wavelength and dose used. Mono- and di-adducts influence DNA stability differently; biologically both types of adducts are genotoxic as measured by action spectra.

[Peripheral vascular disease of the legs associated with diseases of the coronary and carotid systems].

Authors analysed the frequency of occurrences of coronary and carotid diseases on 213 patients suffering from vasosclerosis in the lower limbs. They deal emphatically with diagnostic criteria of vaso-sclerosis appearing in multilocalisation and with the problem of planned therapy. They call readers' attention that multilocular appearance of the vascular disease often escapes the examining physician's observation or it is not adequately stressed in connection with the treatment to choose.

[Hemorheologic parameters during the 6 months following myocardial infarct].

It is verified that rheological factors play a role in the pathomechanism of ischemic heart disease (IHD) and acute myocardial infarction (AMI). The changes of the rheological parameters (whole blood and plasma viscosity, fibrinogen level, hematocrit) of 17 patients (mean age: 59 years) were studied in the first six months after AMI. The measurements were carried out after the admission, before the discharge from the hospital and at the control examinations after one and six months. A slight increase of hematocrit and whole blood viscosity was found during the six months follow up period which was statistically significant concerning the hematocrit (p less than 0.05). As the part of the control examination exercise stress test was performed. Among the rheological parameters hematocrit and whole blood viscosity of those who showed myocardial ischemia during the test were significantly worse (p less than 0.05) comparing to the non ischemic group. These results underline the role of hemorheological parameters as risk factors in these diseases.

[Multistep prognosis in acute myocardial infarction using the PRIMA method (Pattern Recognition by Independent Multicategory Analysis)].

Authors analysed the possibilities of in-hospital prognosis made of patients with acute myocardial infarction by using information obtained during admission. Eighty eight patients with transmural myocardial infarction (not older than 70 years and the prehospital delay shorter than 24 hours) were analysed. On the basis of the hospital events they were divided into 3 classes: (a) uneventful (16), (b) complicated (55) and (c) lethal (17). The initial 26 data available in the first hour were analysed by PRIMA pattern recognition method adapted to ROSY--80B microcomputer. 3 days later the 3 class distances of the initial PRIMA analysis together with the 21 new data of the remaining 79 patients were analysed again. The third analysis (66 patients) was made between the 4th and 6th days after a successful mobilization. The last analysis (44 patients) was performed just before the discharge of the patients, who were able to carry out the low-level and submaximal ergometric tests. The average effectiveness of the method gradually improved during hospitalization from 80% to 91%. The recognition ability of each class respectively was 57% for (a) uneventful, 82% for (b) complicated, and 95% for (c) lethal initially, and it has improved finally: 82% for uneventful, 94% for complicated cases.

[Hemorrheologic parameters in myocardial infarct during the hospitalization phase].

The changes of the rheological parameters (whole blood and plasma viscosity, fibrinogen level, red cell count, hematocrit) of 22 AMI patients (mean age: 59 years) were studied during the hospital phase of acute myocardial infarction. The measurements were carried out after the admission, after the beginning of the mobilization and before the discharge from the hospital. Looking at the results of all of the patients they could not find any statistically significant change, although in the first phase improving hemorheological parameters were measured and this way the discharge values were similar to the admission ones. In men significantly worse starting values were found than in women and the deterioration of these parameters in their group was also expressed. The deliberate use of diuretics and the administration of high dose nitroglycerin may be one part of the less worsening rheological parameters in comparison with the literature.

Lipid-rich cell thyroid adenoma: histopathology with comparative lipid analysis.

A second case of the unique lipid-rich cell thyroid adenoma is described complemented by detailed lipid analysis. New observations were made. The cytoplasm of the tumour cells contained scattered, aggregated sudanophil crystals; under polarized light the frozen, unstained sections exhibited numerous birefringent lipid crystals; electron microscopy provided further evidence that the clear cell appearance was due to intracellular lipid droplets with scanty glycogen particles. Comparative lipid analysis by thin layer chromatography and high-pressure liquid chromatography (HPLC) revealed quantitative and qualitative differences in lipid composition of tumour cells when compared with goitre cells from normal thyroid gland and subcutane fat. Qualitative differences in triglyceride composition (by HPLC) between tumour cells and subcutaneous fat indicated that the fat accumulation in the follicle cells was not a result of simple storage, but an expression of altered intracellular lipid metabolism.

Serotonergic control of the hippocampus via local inhibitory interneurons.

Information flow and processing in hippocampal neuronal networks is determined by a wide range of inhibitory mechanisms [e.g., feedforward or feedback, gamma-aminobutyrate (GABA) A or B receptor-mediated, perisomatic shunting, or distal dendritic inhibition], each subserving specialized functions. These forms of local inhibition are mediated by morphologically and neurochemically well-defined, mostly GABA-containing, interneurons, which control large populations of principal cells through their extensive axonal arborizations. These neurons can serve as ideal targets for subcortical pathways, such as those originating in the septum or raphe, which exercise a global control over hippocampal activity. This intriguing possibility prompted us to study whether the profound effect of the serotonergic raphe-hippocampal pathway is mediated by inhibitory interneurons or whether a direct diffuse action on the principal cells is dominant. We demonstrate that axons of this pathway form multiple synaptic contacts with hippocampal GABAergic interneurons. Interestingly, the serotonergic afferents selectively innervate the somata and dendritic trees of GABAergic neurons that contain the 28-kDa calcium-binding protein calbindin D28K, but never those that contain another calcium-binding protein, parvalbumin. These results show that the mechanism by which the serotonergic pathway may exert a powerful influence on hippocampal function involves the modulation of local inhibitory circuits. Furthermore, the selectivity in the choice of target GABAergic interneurons suggests a strong functional specialization among inhibitory circuits, as well as among the subcortical input pathways originating in the septum and raphe.

Coincidental papillary carcinoma of the thyroid in two sisters.

A coincidentally occurring papillary cancer of the thyroid in two sisters is discussed. No clinical, biochemical or chromosomal changes were found in these cases which could have shown any inheritable feature. It is concluded that, in contrast with the previous data, the simultaneous manifestation is not a peculiar feature but a fortuitous appearance.

[Experience with the Matti-Russe-Zolczer method in the surgical management of pseudoarthrosis of the navicular bone of the wrist].

Authors performed in the Department of Traumatology of the County Hospital Kaposvár from 1979 to 1985 22 Matti-Russe-Zolczer operations for pseudoarthrosis of the navicular bone. The necessity of the strict observance of the conditions of indication is stressed. The results of operations were assessed in a follow-up. The time elapsed between the operation and control was 2 and 6 years. Good results were achieved after the use of Matti-Russe-Zolczer operation in 22 patients with navicular pseudoarthrosis.

[Salt fluoridation].

The author reports on more than 17 years of trials involving determination of optimum daily requirement of F-, water and salt as well as the F- content of various foods and of urine. On the basis of these physiological data, support is found for salt fluoridation to prevent tooth decay. The results demonstrate that salt fluoridation is not only suitable for community use in automated systems, but is also a universally applicable substance.

[Clinico-pathological observations in myocardial rupture].

In their material of 10 years authors describe 42 myocardial ruptures developed as complication of acute myocardial infarct. The rupture developed in the acute stage of infarct. Rupture of anterior wall infarct is the most frequent. The most decisive clinical factor in development of rupture is the loading in acute stage, from pathologic aspect the strongly infiltrated, transmural infarct is considered the most decisive pathologic factor. Diabetes mellitus and hypertonia can predispose to myocardial rupture.

Pattern recognition in evaluation of haemorheological and haemodynamical measurements in the cardiological diagnostics.

The non-invasive differential diagnosis of ischaemic heart disease (IHD) and acute myocarditis or secondary cardiomyopathy following myocarditis can be difficult on the basis of the complaints, resting and exercise ECG and nuclear cardiological tests. 92 patients (mean age: 46 years) in the first step and 100 patients (mean age: 44 years) in the second step all with heart troubles, were examined. Besides determination of the routine parameters, nuclear haemodynamical and haemorheological measurements were carried out. Then each group of the patients was classified into 4 subgroups: 1) myocardial infarction /n:9/, 2) IHD /52/, 3) myocarditis /28/, 4) chronic cor pulmonale (CCP) /3/ subgroups in the first group and 1) normal /n:20/, 2) IHD /50/, 3) myocarditis /16/, 4) chronic cor pulmonale /14/ subgroups in the second group. The patients were reclassified by our multivariate pattern recognition algorithm (PRIMA). The average effectiveness of our method was over 80%, the recognition abilities for the subgroups (classes) ranged between 71 and 100%. An analysis of the discrimination power of the properties has made it evident that the haemorheological features were more characteristic than the haemodynamic ones in distinguishing the two differential-diagnostically critical groups. Our results show that our multivariate statistical method can be useful for the computer-aided decision in cardiological diagnostics.
