Induction of ovulation with pulsatile subcutaneous administration of human menopausal gonadotropin in anovulatory infertile women.

Pulsatile administration of human menopausal gonadotropin (hMG) via the subcutaneous route was evaluated in 15 patients with various ovulatory disorders. Administration of hMG was started at a dose of 4.6875 IU (75 IU/day) or 9.375 IU (150 IU/day) per pulse every 90 minutes. Ovulation was observed in 26 (92.9%) of 28 treatment cycles, and two singleton pregnancies were confirmed. Ovarian hyperstimulation was observed in 1 to 26 ovulatory cycles; however, no other side effects were observed during treatment. A regimen of 75 IU/day resulted in a significant increase (P less than 0.0001) of the total dose and prolongation of the treatment period for induction of ovulation, as compared with that of 150 IU/day. Shortened luteal phases occurred in ovulatory cycles induced by pulsatile subcutaneous treatment. Human chorionic gonadotropin administration given every other day until the midluteal phase significantly prolonged the duration of the luteal phase (P less than 0.05). This treatment in patients with the polycystic ovary syndrome was followed by a normalization of luteinizing hormone/follicle-stimulating hormone ratio and resulted in a successful induction of ovulation in 8 to 10 cycles. The present data demonstrated that pulsatile subcutaneous administration of hMG was effective in inducing follicular maturation and ovulation in patients with various types of anovulatory infertility.

Inhibition of in vitro growth of Plasmodium falciparum by a brief exposure to the cationic rhodamine dyes.

The effects of eight permeant fluorescent dyes on the in vitro growth of Plasmodium falciparum was investigated. First, P. falciparum-infected human erythrocytes were synchronized with D-sorbitol and treated with the cationic fluorescent dye rhodamine 123 at 37 degrees C for 30 minutes, and the growth of the treated parasites monitored by examining daily parasitaemias. Rhodamine 123 inhibited the parasite growth at more than 5 microM, the 50% effective concentration being 6 microM. Ring forms and trophozoites were more susceptible to the dye than schizonts. The development of dye-treated ring forms and trophozoites to schizonts was greatly inhibited, and so few new ring forms were produced. In contrast, the dye-treated schizonts produced a large number of new ring forms, though to a slightly lesser extent than untreated schizonts. The rhodamine 123-induced growth inhibition was partially reversed by treating the dye-pre-exposed infected erythrocytes with the proton ionophore carbonyl-cyanide m-chlorophenylhydrazone, which dissipates transmembrane proton gradients. A survey of seven other fluorescent dyes demonstrated that the cationic dyes, including rhodamine 123, rhodamine 6G, rhodamine 6G perchlorate and rhodamine 3B perchlorate, exerted the growth inhibitory effect, whereas the neutral dyes rhodamine B, rhodamine 110, and rhodamine 19 perchlorate, and the anionic dye fluorescein, did not. Fluorescent microscopy revealed that P. falciparum accumulated the cationic dyes but not the neutral and anionic dyes. These results indicate that the cationic rhodamine dyes, which accumulated in the parasite, inhibit the growth of P. falciparum.

Structural differences of microtubule associated proteins from brain probed by tryptic peptide mapping.

Microtubules were purified from porcine brain by two cycles of temperature-dependent assembly and disassembly, then microtubule associated proteins, MAP-1, MAP-2, and tau, were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Two-dimensional tryptic peptide maps of radioiodinated polypeptides were compared with each other by means of mixed sample experiments, and the following results were obtained. Subspecies of MAP-1 (355-345 and 325 kDa) showed about 33% homology in the tryptic peptide maps. Structural homology of MAP-1 and MAP-2 was very low; only 3 out of 40 peptide spots of MAP-2 were identical with those of MAP-1-C. Subspecies of tau proteins (65 and 60 kDa) were very closely related. Structural similarity between MAP-2 and tau was very low. MAP-1 from porcine brain and rat brain showed very high structural homology.

Renal oncocytoma.

Up to date only five cases of renal oncocytomas have been reported in Japan. We describe two cases of renal oncocytomas. Case 1: A 63-year-old woman, who had no urological symptoms, was pointed out of a cystic mass in the left kidney by chance. A resected kidney had a cystic tumor, which was well encapsulated and measured 6 X 7 X 8 cm. The tumor was unilocular and thick-walled cyst filled with bloody fluid. Histologically, the tumor was composed of oncocytes with eosinophilic granular cytoplasms that were packed with mitochondria. Case 2: A 70-year-old women died of malignant lymphoma and an autopsy was performed. The tumor was found in the right kidney by chance and measured 1 cm in diameter. The cut-surface of the tumor was uniform and tan-brown in color. Histologically, the tumor was composed of cells same as Case 1. We discuss the incidence of this tumor and the morphological characteristics of our cases.

Growth of Plasmodium falciparum in sodium-enriched human erythrocytes.

Transport of nutrients into animal cells is driven by transmembrane gradient of Na+ across the plasma membrane. The protozoan malaria parasite, Plasmodium falciparum, however, grows within the host human erythrocytes, in which the cytoplasmic concentration of Na+ is maintained low by the membrane Na+, K+-ATPase. Our experiments show that human erythrocytes enriched with Na+ by treatment with ouabain (an inhibitor of the ATPase) will support the growth of P. falciparum in culture.

Possible cytoskeletal association of 69,000- and 68,000-dalton heat shock proteins and structural relations among heat shock proteins in murine mastocytoma cells.

When murine mastocytoma cells (FMA 1) were heat shocked (42 degrees C for 4 h), nine heat shock proteins (HSPs) were detected by two-dimensional gel electrophoresis. Their apparent molecular weights were 100, 85, 69, 68, 32, 30, and 23 kDa (3 of 23 kDa). The structural homology of 4, 69, 68, 32, and 30 kDa, was demonstrated by two-dimensional tryptic peptide mapping. The 69- and 68-kDa HSPs were purified and rabbit antisera against these HSPs were prepared. A small fraction (less than 10%) of the 69- and 68-kDa HSPs were copurified with the microtubules and were present in the Triton X-100/KCl cytoskeletal fraction as shown by immunoblotting with the antiserum and by peptide mapping. Our results are consistent with the hypothesis of a cytoskeletal role for HSPs.

[Clinical evaluation of cefmenoxime in internal medicine, with special reference to infection associated with hematological disorders].

Clinical evaluation of cefmenoxime (CMX, Bestcall) was performed against infections associated with hematological, respiratory tract and other disorders. Clinical effectiveness of CMX against severe infections with hematological disorders including sepsis, pneumonia, pyelitis and so on was 74.4% for good responses and against the respiratory tract infections, 96.2% for good responses was obtained. Neither objective or subjective side effects nor extreme abnormalities in laboratory tests were observed in these patients. It can be concluded, therefore, that CMX is one of the most useful drugs against infectious diseases associated with hematological disorders, respiratory tract and other disorders.

[Clinical evaluation of sulpiride against nausea and vomiting during cancer chemotherapy compared with domperidone--envelope method].

Sulpiride, which has been utilized as an antipeptic ulcer and antidepressant in Japan, is one of the dopamine antagonists, and is considered to have anti vomiting activity. We designed a comparative controlled study of Sulpiride and Domperidone, against nausea and vomiting during cancer chemotherapy. A total of 69 cases were entered into this study, 34 cases being treated with Sulpiride at a dose of 100 mg i.v. or i.m. (S group), and 35 cases being treated with Domperidone at a dose of 10 mg i.v. (D group), before and after chemotherapy. Three cases from the S group and 4 cases from the D group were excluded from this study because of differences in the administration schedule. There was no difference in patient characteristics between the two groups, such as age, sex, original disease and antitumor agents. According to our criteria 27 of 31 cases in the S group showed effectiveness of the agent against nausea and vomiting (87%), while in the D group, 24 cases showed effectiveness (77%). There was no statistical difference between them, and no side effects were found in either group.

Characterization of a Mr = 56,000 polypeptide associated with 10S DNA polymerase alpha purified from calf thymus using monoclonal antibody.

Existence of a Mr = 56,000 polypeptide associated with 10S DNA polymerase alpha was shown by production of a monoclonal anti-calf thymus 10S DNA polymerase alpha antibody secreted from a hybridoma line named 3H1. The antibody bound three polypeptides with Mr = 180,000, 56,000 and 32,000 in hydroxylapatite fraction of 10S DNA polymerase alpha by immunoblot. The antibody co-precipitated the polypeptides with the large polypeptide (Mr = 150,000-140,000) of 10S DNA polymerase alpha with the aid of second antibody. Among three polypeptides, the Mr = 56,000 polypeptide was co-purified with DNA polymerase alpha through DNA-cellulose chromatography and repeated sucrose rate-zonal centrifugations. The Mr = 56,000 polypeptide was still associated with 10S DNA polymerase alpha after second sucrose rate-zonal centrifugation, but the amount of it was reduced. The polypeptide was banded at pH 7.2-8.0 and displayed microheterogeneity in respect of isoelectric point by isoelectrofocusing with 7 M urea, and showed weak DNA-binding property after blotting onto a nitrocellulose. The antibody against the polypeptide precipitated DNA polymerase alpha from human, rat, and mouse, and Mr = 56,000 and 32,000 polypeptides were detected in these DNA polymerase alpha fractions by immunoblot. These results suggest that the polypeptide with Mr = 56,000 may take part in the DNA polymerase reaction.

Identification and characterization of a variant of the third component of complement (C3) in rainbow trout (Salmo gairdneri) serum.

A rainbow trout serum protein that is cross-reactive with the third complement component of rainbow trout (C3-1) was purified to homogeneity and its structural and functional properties compared with those of C3-1. This protein (termed C3-related protein: C3-2) bears a close structural resemblance to C3-1, although C3-2 apparently shows no hemolytic activity. Like C3-1, C3-2 consists of two disulfide-linked polypeptide chains (128,000 alpha and 72,000 beta) and retains the unique thiol ester site in the alpha-chain. C3-2 shares some antigenicity with C3-1, but it also displays distinctive antigenic determinants of its own. Comparison of tryptic peptide maps revealed that about 20% of the peptides was specific to either C3-1 or C3-2, and about 80% of the peptides were common to both proteins. Amino acid compositions of the alpha- and beta-chains of C3-2 were similar to those of C3-1. Furthermore, amino acid sequence analysis of the NH2 termini of the alpha- and beta-chains of C3-2 revealed a high degree of homology with those of C3-1, 24 of 26 residues in the alpha-chain and all 20 in the beta-chain of C3-2 were identical with those found in C3-1. Both C3-1 and C3-2 were detected in all the adult rainbow trout tested and in first generation offspring randomly bred from them.

Visualization of the mitochondria of Toxoplasma gondii-infected mouse fibroblasts by the cationic permeant fluorescent dye rhodamine 123.

The mitochondria of living mouse fibroblasts infected with Toxoplasma gondii were monitored with the cationic permeant fluorescent dye rhodamine 123. Fluorescence microscopy revealed that host cell mitochondria accumulated at the cytoplasmic surface of parasitophorous vacuoles and increased the dye uptake in the periparasitophorous vacuole as T. gondii multiplied.

Localization of 350K molecular weight and related proteins in both the cytoskeleton and nuclear flecks that increase during G1 phase.

Monoclonal and polyclonal antibodies were raised against the highest molecular weight microtubule-associated protein (MAP-1) isolated from brain. Immunoblotting with the antibodies revealed the presence of cross-reactive protein of 350K or less on whole cells, isolated nuclei and cellular microtubules. Two-dimensional peptide maps showed substantial homology of immunoprecipitated cellular proteins of 350K, 80K and 51K with a 25K fragment of brain MAP-1. On antibody staining, immunofluorescence was seen on a cytoplasmic network, the mitotic spindle, the centrosome, and intranuclear flecks. The antibody causing immunofluorescence in all these sites was absorbed most effectively with slices of blotted membrane which contained the 350K protein. These results suggest that the cross-reactive molecules in diverse sites belong to the family of the 350K protein. The number of nuclear flecks and the amount of bound radioactivity of 125I-antibody almost doubled during G1 phase.

Isolation and characteristics of a leukemia-growth-promoting factor from calf thymus.

Leukemia-growth-promoting factor (LGPF) as we previously reported stimulates the growth of a murine leukemia subline (L17R) extensively. LGPF was isolated and 10(4) fold purified from calf thymuses by a combination of ammonium sulfate precipitation, Sephadex G-100 chromatography, hydroxylapatite chromatography, and Mono S-fast protein liquid chromatography (Mono S-FPLC). The mol. wt of LGPF was estimated to be approximately 25,000 a.m.u. from the elution pattern of Sephadex G-100 chromatography. The activity had high affinity for Mono S beads which are cation exchangers. Mono S fractions of LGPF are effective at a low concentration of 5 ng/ml. The activity was inactivated by heat (56 degrees C, 30 min), 1 mg/ml trypsin (37 degrees C, 1h), and 50 mM dithiothreitol (20 degrees C, 1h). The growth L17R leukemia cells are not only stimulated by LGPF, but also by pituitary and brain fibroblast growth factor (FGF). These data strongly suggest that LGPF is a heat sensitive cationic protein(s) acting as a member of FGF family.

Demonstration of a gradient in inhibin activity, estrogen, progesterone, and delta 4-androstenedione in follicular fluid, ovarian vein blood, and peripheral blood of normal women.

Ovarian vein serum from 3 subjects during the late follicular phase of the menstrual cycle had detectable inhibin activity, whereas ovarian vein serum of 12 other subjects during the early follicular phase and luteal phase had no detectable inhibin activity in a rat anterior pituitary cell culture assay. Subjects having detectable inhibin activity (102 +/- 47 U/100 microliters) had 1257 +/- 582 U/100 microliters inhibin activity in FF, whereas subjects having no detectable inhibin activity had FF levels of 711 +/- 203 U/100 microliters of inhibin activity. Estrogen levels of FF and ovarian vein serum of the group having detectable inhibin activity in ovarian vein serum were 282 +/- 239 ng/ml and 4.8 +/- 1.77 ng/ml, respectively. The estrogen content of FF and ovarian vein blood of the group having nondetectable inhibin activity in ovarian vein blood was 127 +/0 45 ng/ml and 3.03 +/- 0.6 ng/ml, respectively.
