Affinity chromatography of human serum proteins using immobilized lectin from Allomyrina dichotoma.

The beta-D-galactoside-specific lectin from Allomyrina dichotoma reacts with serum proteins which contain the corresponding carbohydrate moieties. By affinity chromatography of human serum using the insolubilized lectin coupled to Sepharose, it is possible to fractionate human serum proteins in 2 groups: those which react with the lectin (alpha 1-acid glycoprotein, haptoglobin, etc.) and those which do not (albumin, Gc-globulin, etc.). IgG is the only serum protein that can be found in both groups.

A new alpha 2HS-glycoprotein typing by isoelectric focusing.

Isoelectric focusing (pH 4.0-5.0) of serum alpha 2HS-glycoprotein on polyacrylamide gels has been found to be a useful tool in population genetics and forensic science. Using this method, we isolated three common types, alpha 2HS 1-1, alpha 2HS 2-1 and alpha 2HS 2-2, and showed that alpha 2HS types are determined by two autosomal codominant alleles, alpha 2HS1 and alpha 2HS2. The method is simple, fast and easy to perform. Results of typing for the two alleles, alpha 2HS1 and alpha 2HS2, are described for a Japanese population sample (n = 1003).

A new alpha 2HS-glycoprotein allele (AHS*5) in two Japanese families.

Serum specimens of two unrelated Japanese males had a new variant of the alpha 2HS-glycoprotein phenotypes. They had unusual bands designated AHS 5. Family studies indicated that the new variant phenotypes were determined by a new allele, AHS*5, in combination with a common allele AHS*1 or AHS*2, and that the new allele had an autosomal codominant inheritance with other AHS alleles. The frequency of the new alpha 2HS-glycoprotein allele, AHS*5, is 0.0005.

An experimental study of the identification of the person of origin of a bloodstain by crossed immunoelectrophoresis.

Crossed immunoelectrophoretic (CIEP) patterns were examined for the purpose of individualization of bloodstains. For comparison the areas of peaks were measured and ratios of the areas of corresponding peaks were calculated. To assess the dispersion of the ratios, the coefficient of variation (CV) was calculated as CV = 100 X standard deviation/mean of ratios. With 1-month-old bloodstains there were significant differences in the distributions of CV between the groups of the same and those of different origin. These results indicate that CIEP is useful as a means of identifying the origin of bloodstains.

Purification and characterization of a lectin from the beetle, Allomyrina dichotoma.

A lectin was purified from the hemolymph of Allomyrina dichotoma larvae by affinity chromatography on acid-treated Sepharose 4B. The purified lectin showed two protein bands on polyacrylamide gel electrophoresis. These two lectin bands (allo A-I and -II) were separated by DEAE-Cellulofine column chromatography. By gel filtration on Sephadex G-100, the molecular weights of allo A-I and -II were estimated to be 65,000 and 66,500, respectively. On the other hand, by SDS-polyacrylamide gel electrophoresis after cross-linking of subunits with glutaraldehyde, they are estimated to be 38,000 and 39,000, respectively. On SDS-polyacrylamide gel electrophoresis, it was proved that both allo A-I and -II lectin consisted of two subunits, respectively. The molecular weights were 17,500 and 20,000 for allo A-I, and 19,000 and 20,000 for allo A-II. The isoelectric points of allo A-I and -II were estimated to be 6.4 and 5.9, respectively. On double immunodiffusion, allo A-I and -II gave single precipitin lines, which fused completely with each other, against the antibody to crude allo A. The hemagglutinating activity of allo A-I and -II was inhibited only by beta-linked D-galactose such as lactose and lactulose.

Classification of alpha 2-HS-glycoprotein (alpha 2HS) types by isoelectric focusing.

A sample of 300 sera from unrelated individuals from Northern Japan was examined by isoelectric focusing on polyacrylamide gels. Three common types, alpha 2HS 1-1, 2-1, and 2-2 were differentiated. The frequencies of the alpha 2HS alleles in our sample were found to be: alpha 2HS1 = 0.7250 and alpha 2HS2 = 0.2750. Analysis of 16 parents with 21 children did not show deviations from the expected mode of inheritance.

[Postmortem permeation of myoglobin into the blood].

Myoglobin in heart and femoral blood of human cadavers and of experimentally killed dogs was examined immunologically. Myoglobin was found in blood of most human cadavers more or less without relation to sudden or delayed death. In general, more myoglobin was found in the heart blood than in the femoral blood of cadavers within 1 day after death. The appearance of myoglobin in blood seemed to be related to the length between death and blood taking. In experimentally killed dogs, myoglobin was found in heart blood already 1-2 h and in femoral blood 6-8 h after death. These results reveal that postmortem myoglobin permeation into the blood occurs with in 1-8 h after death and suggest that by examination of myoglobin in blood stains a differentiation between ante- and postmortem blood is highly possible.

Experimental model of sudden cardiac death from unexplained cause.

To find a possible explanation for sudden cardiac death from unexplained cause, an experimental model was made with rabbits. The rabbits were fed an atherogenic diet for 2 and 4 months without precordial x-irradiation. Though the coronary sclerosis in the hearts of these rabbits was only slight or not detectable, the ventricular premature contraction threshold (VPCT) and the ventricular fibrillation threshold (VFT) decreased by about 50% and 60% of those in the control animals, respectively. The investigation of isozyme patterns of LDH, GOT, MDH and CPK in the hearts revealed that the changes of these isozyme patterns were almost identical with the results from the human hearts of sudden cardiac death from unexplained cause. From these results, the rabbits fed an atherogenic diet without precordial x-irradiation can be an experimental model of sudden cardiac death from unexplained cause.

Creatine phosphokinase and malate dehydrogenase isozymes in heart muscle of sudden death cases.

CPK and MDK isozyme patterns of the heart muscle in cases of sudden death were studied to investigate the relationship between their changes and the causes of death. In most of death cases from violence, asphyxia, natural disease and poisoning, the percentage of CPK-MB was about 23--34% of the total CPK, and that of MDHm was about 35--50% of the total MDH. In a few death cases of coronary sclerosis and hypertrophy of the heart, the percentages of CPK-MB and MDHm decreased. The percentages of CPK-MB and MDHm in neonates were less than those in adults and they were about 7--15% and 25--34% of the total CPK and MDH, respectively. In more than half of the cases of both cardiac death from unknown cause and cot death, the average percentages of CPK-MB and MDHm decreased significantly. The findings of CPK and MDH isozyme patterns in cases of cardiac death from unknown cause and cot death suggest that there are some similar changes in many other kinds of heart enzymes in these cases.

Studies on anti-B specificity and heterogeneity of some invertebrate hemagglutinins.

Hemolymph from Charybdis japonica and Lymantria dispar, and saline extract from Eunice kobiensis agglutinated human group B erythrocytes specifically. Multiple agglutinins were identified, and independent agglutinins were separated from hemolymph of Charybdis japonica (anti-Bcj+anti-Xcj) and Lymantria dispar (anti-BLD+RLD) by absorption experiments. By the agglutination and agglutination-inhibition experiments with simple sugars, the specificities of the anti-B agglutinins in these invertebrates were found to differ from each other.
