[Genetic aspects of the effect of Vi-antigen deposition in the human body. I. Hereditary cause of the effect of Vi-antigen deposition on erythrocytes].

Investigation of the minimal and the optimal capasity of the Vi-antigen sorption in erythrocytes was carried out in partners of 11 pairs of monozygotic and 8 pairs of dizygotic twins. It is shown that the capacity of erythrocytes to sorb Vi-antigen in minimal concentrations (Smin) is under relatively rigid genetic control (G = 0.84), while the capacity of its sorption in optimal doses (Sopt) is to a much greater extent subjected to paratypic effects (G = 0.40). It is assumed that Smin is controlled by the system of the "main" gene, while Sopt is determined multifactorially, which affords the possibilities of creating adequate conditions for the vaccination of a concrete individual.

[Genotypical characteristics of the immunological Vi-antibody maturation depending on the effect of Vi-antigen deposit in the body].

The authors carried out a comparative analysis of the changes in the antibody synthesis and their immunological maturation in the immunization with various doses of the Vi-antigen of linear rats differing by the capacity to deposit this antigen. August rats with a high capacity to deposit the Vi-antigen (in comparison with Wistar rats in which this capacity was less) were characterized by an increased level of highly avid antibodies. The interlinear differences were found in the character of the changes in the antibody synthesis depending on the immunizing dose of the antigen. Genotypical peculiarities of the immunological maturation of the antibodies were demonstrated. The authors suppose that the described phenomenon was genetically determined and referred to the factors determining the individual immunological reactivity. In the choice of the optimal immunizing doses of the antigens the authors recommend complex determination of the antibody level and the degree of their avidity.

[Immunoglobulins and their specific activity in cholera].

The authors studied the quantitative content of nonspecific immunoglobulins and antibodies of the IgG-, IgA-, and IgM-classes to the O-antigen of the cholera vibrio. There proved to be no marked dynamics of the quantitative immunoglobulins indices of various classes established at periods from the 3rd and the 18th day from the beginning of the disease in persons who sustained cholera. In persons who sustained cholera in the nonendemic focus of the disease serum antibodies were represented chiefly by the IgM-antibodies. The formation of IgA-antibodies in the persons examined differed from the regularities detected for the IgG- and IgM-classes and were characterized by rapidity and short duration. No correlation was found between the immunoglobulin levels of various classes and the serum antibody levels in case of cholera affection.

[A study of the avidity of Vi antibodies in typhoid fever patients and chronic carriers].

Avidity of Vi-antibodies was studied in the patients suffering from typhoid fever and chronic carriers in the indirect hemagglutination test (IHAT) by means of titration with erythrocytic diagnostic agents sensitized with various doses of the Vi-antigen. The minimal sensitizing dose still adequate for detection of the antibodies in the titre of 1 : 20 was accepted as the avidity index. By the avidity sign the antibody heterogeneity index was calculated as the ratio of the optimal sensitizing dose to the minimal sensitizing one. In comparison with the patients suffering from typhoid fever of at the acute period of the disease in chronic carriers Vi-antibodies were more avid, this being conditioned chiefly by the highly avid IgG-antibodies. During the acute period of the disease the avidity of Vi-antibodies elevated only against the antibodies of the IgG-class. An increase of heterogeneity by the same sign was noted with increase of the degree of the serum antibody avidity.

[Peculiarities of immunological response to the administration of Vi-antigen in patients suffering from typhoid fever and in experimental animals].

Vi-antigen parenterally administered into the organism could be revealed on erythrocytes with the aid of inhibition of erythrocyte sedimentation reaction with immune Vi-serum. Patients with typhoid fever displayed positive results in 53.8% of cases. There were differences in the incidence of detection of the Vi-antigen in August and Wistar rats conditioned by theigenotype and due to the different capacity of their erythrocytes to sorb this antigen. Peculiarities of immunological response to the Vi-antigen administration depended on its sorption by erythrocytes in the organism and were the same in patients with typhoid fever and in the linear animals. A decreased capacity of erythrocyte to sorb Vi-antigen induced its more rapid elimination from the organism and caused a weak production of specific antibodies; this should be taken into consideration in immunotherapy of typhoid fever.

[Avidity of specific antibodies in the sera of cholera patients and vibrio carriers].

Avidity of specific antibodies was studied in the sera of patients suffering from cholera and vibrio-carriers by the rate of antibody binding (in the bacterial agglutination reaction) and by the completeness of their binding with O-antigen (in the passive hemagglutination test). There were revealed statistically significant higher indices of antibody avidity contained in the blood sera of the vibrio carriers, in comparison with the antibodies in the blood sera of patients with clinically manifest forms of cholera. There proved to be an increase in the antibody avidity in the dynamics of the infectious process. It is supposed that the degree of avidity of anticholera antibodies in examination of whole sera depended on the ratio of the specific macro- and microglobulins in them.

Agglutination Tests↗

[Use of the thrombocyte aggregation test for detecting immune complexes circulating in the serum of cholera patients].

It was shown that the blood sera of patients suffering from cholera and of persons who had sustained the disease contained specific antibodies in titres below the diagnostic level (according to the indirect hemagglutination test) caused platelet aggregation, most pronounced during the first week of the infectious process. In the presence of a specific cholera antigen the phenomenon of platelet aggregation was considerably enhanced; the indices reached the maximum when the experiment was conducted with the blood sera of those who had sustained the disease.
