Partially purified samples of thymidine phosphorylase were obtained from four preparations of human uterine leiomyomas and uteri using the method of Yoshimura et al. (1990), Biochim. Biophys. Acta 1034, 107-113. Among the studied twelve pyrimidine derivatives, 5-bromouracil, 5-nitrouracil, 5-fluorouracil, 6-aminouracil, 4, 6-dihydroxy-5-nitropyrimidine are competitive inhibitors, while allyloxymethylthymine is an uncompetitive inhibitor of thymidine phosphorylase activity, 6-benzyl-2-thiouracil inhibits the activity of the enzyme in a mixed way. The most potent inhibitor of the thymidine phosphorylase activity is 5-bromouracil and uracil the weakest one. Stronger inhibition of these compounds on the activity of thymidine phosphorylase was found in uterine leiomyomas than in uteri.
The differences between cis- and trans-diamminedichloroplatinum II (DDP) in forming DNA-protein cross-links in isolated human lymphocytes were investigated. Both cis- and trans-DDP can induce DNA-protein cross-links. We show that cis-DDP forms complexes between DNA and proteins faster than trans-DDP. This results from an increase in the quantity of DNA and platinum together with an increase in drug concentration. Under the same conditions trans-DDP causes a decrease in DNA-forming complexes with proteins. After a 12 h incubation of lymphocytes we observe a similar level of DNA in DNA-protein cross-links induced by DDP isomers, but more platinum appears in complexes induced by trans-DDP. The results obtained demonstrate that the antitumor drug - cis-DDP and the clinically ineffective trans-DDP induce links between DNA and proteins in a different manner. We suggest that the therapeutic activity of cis-DDP can in part arise from rapidly forming DNA-protein complexes which can destroy the most important cellular processes, such as replication and transcription.
In 22 patients with chronic cor pulmonale syndrome plasma renin activity (PRA) was determined at rest and after stimulation by means of intravenous injection of furosemide. In 50% of cases PRA was raised exceeding 250 ng/100 ml/3 hours. The systemic arterial pressure in these patients was normal. The degree of PRA increase after stimulation was lower than in healthy subjects.
The prevalence of the markers of blood groups ABO, MN, Kell, Hp, Gm1 was determined in 723 persons belonging to three age groups: from 61 to 91 years, and was compared with a similar study in a population of adults aged 20 to 60 years. It was found that with respect to MN markers the proportion of NN homozygotes was significantly increased, mainly in females aged 71-80 years. In the haptoglobin system a considerable increase was noted in the prevalence of Hp 1-1 phenotype and the appearance of ahaptoglobinaemia. Increased prevalence of the Gm1 marker was observed only in females aged 71-80 years. On the other hand, no significant differences were noted in the frequency of the phenotypes of the ABO and Kell blood groups. These preliminary results confirm the stability of the serological polymorphism in the ABO and Kell blood groups, and in the remaining blood groups studied a natural selection may be supposed to exist. Characteristic changes in the distribution of the frequency of the determined genetic markers encourage the continuation of such studies.
