Chilling of somatic cells unnecessary for Sendai virus-induced heterokaryon formation.

Incubation of cell suspensions or monolayers at 4 degrees C for 20 min after addition of beta-propiolactone-inactivated Sendai virus did not enhance heterokaryon formation as compared to parallel cell cultures incubated at 37 degrees C immediately after the addition of Sendai virus. These findings show that the routine chilling of somatic cells in fusion experiments can be omitted.

Influence of some cytostatics on leucocyte function.

Phagocytic activity, nitroblue tetrazolium reduction and candidacidal capacity have been studied in a group of normal persons and in three patients with acute leukaemia. The studies were performed in vitro in whole blood to which either a solvent only, or daunorubicin or vincristine had been added. The nuclei of all kinds of leucocytes showed characteristic changes in daunorubicin samples, resembling a beginning necrobiosis. In spite of this, these cells had an unchanged or even an increased phagocyting capacity. Vincristine reduced the phagocyting activity in most cases. The NBT test showed no significant change. Killing function of leucocytes, represented by their candidacidal capacity, was not affected by the cytostatics.

Chemotherapy of advanced malignant lymphomas comparative evaluation of results with single agent and combination therapy.

86 patients with advanced malignant lymphomas (stage III and IV) were treated either with Vinblastine given as a single agent (49 cases) or with combination chemotherapy MOPP or COP (37 cases). In both, Hodgkin's disease and non-Hodgkin's lymphomas, induction of remission, relaps-free survival and prognostic significance of initial clinical stage were evaluated, in relation to the therapeutic modality. In patients with Hodgkin's disease, combination chemotherapy was found to produce significantly more durable remissions, when compared with single agent therapy. However, difference in the response rate, which was found to be more effective following the combination chemotherapy, was not statistically significant. In patients with non-Hodgkin's lymphomas overall lower response was recorded, regardless of the therapy applied. In all patients, better response rate and longer lasting remissions correlated with initial stage III.

Phagocytic activity of leukaemic blasts.

Phagocytic activity of leukaemic blasts in 20 adults and 16 children suffering from acute leukaemia was studied in vitro by the use of ferrioxidsaccharate. Most frequently phagocytosing blasts were encountered in myelomonocytic leukaemias. The morphological character of positive blasts resembled often that of monocytoid cells. In lymphoblastic and reticular leukaemias phagocytic ability of blasts was an exceptional finding.

Trichlortriethylamine (TS-160 Spofa) in combination chemotherapy of malignant lymphomas.

Thirteen patients with disseminated malignant lymphomas were treated with combination chemotherapy which included trichlortriethyl amine (TS-160), vincristine (Oncovin), procarbazine hydrochloride (Natulan) and prednisone. All patients but two received some form of chemotherapy and/or radiotherapy prior to being started on four-drug regimen. Good response to the combination chemotherapy was recorded in 84% of patients, while complete remission was achieved in 23% of cases. Drug toxicity has not proved to be a major problem in the combination used. Nausea and vomiting were the side effects the most frequently observed.
