Receptors for estrogens and progesterone in the porcine cervix.

In vitro binding and exchange methods were used to determine the levels of estradiol and progesterone receptors in cytosolic and nuclear fractions of cells obtained from the porcine cervix at different stages of the estrous cycle. The concentration of estradiol cytosolic receptors was about 4500 sites/cell during the luteal phase and increased to a maximum of approximately 7600 sites/cell on day 1 of the cycle, decreasing to a level of 2700 sites/cell on days 3-4. The estradiol nuclear receptor level increased between the end of the luteal phase and the onset of heat from 300 to 1200 sites/cell. No reduction in the number of nuclear sites was seen between day 1 and 3-4. The level of the progesterone cytosolic receptor and its cycle profile was very similar to that of the estradiol receptor. The nuclear receptor, however, reached its lowest level of 760 sites/cell on day 1 of the cycle, increased to a value of 4700 sites on days 3-4 and showed a steady level of about 1000 sites/cell during the luteal phase. The data obtained agree with present theories on the endocrine mechanisms regulating receptor levels in the uterus. Furthermore, these data support a concept in which the constriction of the cervix occurring in response to increased concentrations of circulating estradiol is mediated via steroid receptors.

Blood levels of 15-keto-13, 14-dihydroprostaglandin F(2alpha) during the postpartum period in primiparous cows.

Plasma levels of the PGF(2alpha) metabolite 15-keto-13, 14-dihydroprostaglandin F(2alpha) were determined postpartum in 51 primiparous Black and White Lowland cows. The highest geometric mean was 1702 pmol/l on day 3 and lowest 190 pmol/l on day 21. It should be noted that variation between animals in the concentration of the metabolite is high. For instance, on days 4, 10, 16 and 22, concentration of metabolite ranged from 775-2500, 209-2450, 45-851 and 30-398 pmol/l, respectively. The duration of the massive postpartum release of PGF(2alpha) could be determined in only 29 cows. Significant correlations were found between the duration of elevated PGF(2alpha) metabolite levels and the time required for completion of uterine involution (r = -0.41, P < 0.05) and between the duration of increased PGF(2alpha) metabolite levels and the interval from parturition to occurrence of the first ovulation followed by a normal luteal phase length (r = -0.37, P < 0.05). The occurrence of the first ovulation followed by a normal luteal phase length in the 29 cows was positively correlated with the time needed for completion of uterine involution (r = 0.54, P < 0.01).

Solubility of copper and zinc containing proteins before and after 2-mercaptoethanol treatment of liver samples from normal and from chronically copper poisoned sheep.

The distribution between cytosol and pellet of Cu- and Zn-containing proteins was studied in livers from normal sheep and sheep with chronic copper poisoning (CCP). It was found that the relative amount of soluble Cu- and Zn-containing proteins was lower in liver homogenates from CCP sheep than in homogenates from normal sheep. Treatment of homogenate from the livers of 2-mercaptoethanol (ME) increased the solubility of Cu- and Zn-containing proteins, especially in CCP sheep. Oxidation of the liver homogenates from normal sheep with air oxygen gave a decrease in the relative amount of soluble CU-containing proteins and an increase in Zn-containing proteins present in the cytosol. It is proposed that oxidation in vivo of Cu and/or the metal-binding protein causes the observed decrease in soluble CU- and Zn-containing proteins seen in CCP sheep.

Erythrocyte glutathione peroxidase activity in acne vulgaris and the effect of selenium and vitamin E treatment.

The glutathione-peroxidase (GSH-Px) activity in erythrocytes was determined in 42 men with severe acne and 47 women with acne--26 of a moderate degree and 21 severe. The male acne patients had significantly lower GSH-Px levels than the controls. The women with acne did not differ significantly from the controls in this respect when patients and controls using oral contraceptives were excluded. Both the female controls and the women with acne using oral contraceptives had significantly higher GSH-Px values than the corresponding groups not using the pill. The pubertal acne girls had the same high GHSH-Px activity as women on oral contraceptives. In an open trial 29 patients were given 0.2 mg of selenium (as Na2Se O3) + 10 mg of tocopheryl succinate for their acne twice daily for 6-12 weeks. A good result was obtained, especially in patients with pustular acne and low GSH-Px activity, and the beneficial effect was usually paralleled by a slow rise of the GSH-Px activity. Some 6-8 weeks after withdrawal of the treatment the GSH-Px values had returned to the pretreatment levels.

Endocrine profiles and embryo quality in the PMSG-PGF2alpha treated cow.

Plasma progesterone and LH concentrations around estrus were determined for both PMSG treated (experimental animals) and non-treated (control animals) dairy cows and heifers of the Holstein Friesian and Jersey breeds, and these hormone profiles were related to the embryo quality. Most experimental animals experienced an increase in progesterone concentrations following PMSG treatment and an abrupt decrease to values below 3 nmol/l after PG injection. The mean (+/-SE) intervals from prostaglandin treatment to estrus were 46.9+/-1.8 h and 64.5+/-4.8 h for experimental and control animals, respectively. At the onset of heat the progesterone concentration in experimental animals with optimal embryo quality (group I) was significantly lower (p<0.01) than in experimental animals which yielded unfertilized eggs (group II) (1.2+/-0.1 versus 3.9+/-0.8 nmol/l) and significantly higher than the level in the control group (0.6+/-0.1 nmol/l). Following estrus the progesterone profiles in all 3 groups were studied and the length of the superovulatory cycle was measured to 26.0+/-4.8 days. The preovulatory LH surge occurred sooner after prostaglandin injection in experimental (41 h) than in control animals (65 h). The LH surge in group I occurred within a narrow range and reached a higher average level than group II (24.2+/-2.2 ng/ml and 16.3+/-3.7 ng/ml, respectively). The control group attained an even higher LH surge (31.8+/-8.8 ng/ml) than did the experimental animals. The data presented in this experiment indicate that plasma levels of progesterone and LH in PMSG-PGF2alpha treated animals are related to embryo or egg quality.

Plasma progesterone levels from oestrus through day 7 after A.I. in heifers carrying embryos with normal or deviating morphology.

Twenty-five dairy heifers, 11 repeat breeder and 14 virgin heifers, were used. Blood samples were taken twice a day (9 a.m. and 3 p.m.) from day of A.I. to 7 days later when embryo collection was made. One series of blood samples were taken from 16 heifers and two and three, respectively, from 6 and 3 heifers. Analysis of progesterone was performed by radioimmunoassay. The embryos were classified in 3 groups: normal blastocysts, morphological deviators, and degenerated embryos. Analysis of variance and a discriminant analysis was performed. The discriminant analysis was made to predict embryonic morphology, using daily progesterone values as predictors. No significant relationship between embryonic morphology and progesterone levels was found. The discriminant analysis showed that 64.9 % of progesterone curves were correctly classified, with individual groups ranging from 50 % (degenerated) to 76.5 % (normal embryos). It was concluded that abnormal embryonic development occurs with normal lateal function and that normal bovine embryo does not exhibit luteotrophic activity 7 days after mating.

Post-partum release of prostaglandin F(2alpha) and uterine involution in the cow.

Peripheral plasma levels of the main blood plasma metabolite of PGF(2alpha) (15-keto-13,14-dihydro-PGF(2alpha)) and progesterone were investigated during the immediate, post-partum period in 59 normally calving cows. Uterine involution was monitored by weekly rectal palpations. The levels of the prostaglandin metabolite were high at parturition and remained thereafter elevated for periods varying up to 7-23 days. Uterine involution was completed during periods ranging from 16-53 days. According to the clinical findings, the animals were divided into three groups. Group A comprises 46 animals which had an uncomplicated, puerperal period. A significant (p<0.001) correlation between the duration of elevated prostaglandin levels and the time for completed uterine involution (Y=29.6 - 1.3 (X - 13.5)) was found for these animals. Group B animals (n=8) had periods of varying length with uterine discharge during the first 30 days post-partum. When compared to group A animals, the animals in group B had comparatively longer periods of prostaglandin release and also longer periods for completion of uterine involution. Group C animals (n=5) at times had palpable, thin-walled, cystlike structures in the ovaries during the first 30 days post-partum. In this group of animals, the periods of high prostaglandin levels, as well as for the completion of uterine involution, were similar to those for the animals in group A. Progesterone levels remained low during the immediate post-partum period and in no case were elevated levels found until the prostaglandin release had ceased.

Absorption of topically applied hydrocortisone from the eye of the rhesus monkey.

Blood samples were collected from anaesthetized monkeys during one h a) after topical application of an eyedrop of hydrocortisone acetate suspension, b) after topical application of a drop of the vehicle alone, and c) after no application at all. Radioimmunoassay (RIA) showed a rapid and similar increase of hydrocortisone plasma levels in all 3 kinds of experiments. RIA revealed that in monkeys the anaesthetic procedure alone was sufficient to cause a considerable increase of endogenous hydrocortisone levels overshadowing any systemic absorption of topical hydrocortisone. Using tritiated hydrocortisone instead a rapid increase of systemic absorption after topical application was found. A plasma concentration of 1% of the dose was found one min after topical instillation increasing to a maximum of 6-7% at 30 min. The half time of the show phase of elimination was about 19 h.

Blood glutathione-peroxidase levels in skin diseases: effect of selenium and vitamin E treatment.

Blood glutathione-peroxidase (GSH-Px) was determined in 61 healthy subjects and 506 patients with various skin disorders. Depressed levels were observed in patients with psoriasis, eczema, atopic dermatitis, vasculitis, mycosis fungoides and dermatitis herpetiformis. Low values of GSH-Px were also found in some patients with pemphigoid, acne conglobata, polymyositis, rheumatoid arthritis, scleroderma and systemic lupus erythematodes. Vegetarian diet, malnutrition and alcohol abuse could possibly account for the low values in some patients. Fifty patients with low GSH-Px levels were treated with tablets containing 0.2 mg selenium as Na2SeO3 and 10 mg tocopheryl succinate. The GSH-Px levels increased slowly within 6-8 weeks of treatment. The clinical effect was encouraging and calls for controlled studies.
