[Interaction of various tetrahydroisoquinoline alkaloids with opiate receptors in the rat hypothalamus and midbrain].

Using the method of competitive displacement of 3H-labelled naloxone, (D-ala2)-met enkephalinamide or (D-ala2)-D-leu-enkephalin by salsolinol, 1-methyl-6-hydroxy-1, 2, 3, 4-tetrahydro-beta-carboline or 1-methyl-6-methoxy-1, 2, 3, 4-tetrahydro-beta-carboline from opiate receptors, it was shown that the alkaloids studied were capable to cause specific interactions with rat hypothalamus and midbrain however, exhibiting distinctly less affinity as compared with morphine or its analogs. The sodium ratios, determined from the effective doses of the tetrahydroisoquinoline alkaloids corresponding to the alkaloid concentrations, which induce 50% displacement of 3H-naloxone from the opiate receptors in the presence or absence of 100 mM NaCl, have been found to be 0.75 for salsolinol and 3.6 for beta-carbolines studied. The data obtained suggest that salsolinol, similar to naloxone, is a "pure" morphine antagonist, whereas the beta-carbolines studied may be classified with the agonist-antagonist type. A considerable decrease in the affinity of mu-type opiate receptors has been found in presence of salsolinol in the incubation medium. The possible mechanisms of pharmacological action of the alkaloids and their relation to development of alcohol dependence and tolerance are discussed.

[Effect of chronic alcohol consumption on activity of enkephalinase A in rat striatum, hypothalamus and midbrain].

A modified procedure is developed for estimation of encephalinase A activity in rough membrane material from rat brain; the accuracy of the procedure was about +/- 5% and sensitivity--0.2 X 10(-15)M of 3H-Tyr-Gly-Gly per min if labelled Leu-encephalin was used as a substrate. Activity of encephalinase A in midbrain and hypothalamus was increased by 10% and in striatum--by 25% after chronic consumption of alcohol within 6 weeks. Rates of specific binding of 3H-Leu-encephalin with opiate receptors in the brain departments studied of the alcohol-treated rats were similar to those of control values. Encephalinase A appears to participate in development of dependence on alcohol or tolerance to it.

[Characterization of clofibrate-stimulated alkaline phosphatase in rat liver].

Hypolipidemic agent clofibrate induced the alkaline phosphatase activity in rat liver but did not affect this activity in kidney, brain, heart and intestine. In liver cells the clofibrate induced alkaline phosphatase was localized in plasma membrane and cytosol and was similar to the enzyme found in normal hepatocytes by its main properties (pH optimum, inactivation temperature, substrate specificity, Km, effect of inhibitors and inducers). The putative mechanism of the alkaline phosphatase induction in liver tissue by means of clofibrate is discussed.

[Superoxide dismutase--radiobiologic significance and possibilities (review)].

Modern data are considered on the importance of superoxide radical in the oxygen effect of ionizing radiation as well as the participation of superoxide dismutase in biological radioresistance. Biological role and some physico-chemical properties of the enzyme are described. Estimation of the superoxide dismutase activity might serve as a test for study of the primary effect of ionizing radiation on cells; at the same time, the enzymatic preparations could be used as radioprotective drugs.

[The effect of reaferon on the opioid systems of chronically alcoholized rats].

Chronic alcoholic intoxication is followed by a fall in 3H-(D-ala-2,D-leu-5)-enkephalin affinity of mu-opioid receptors with their unchanged concentrations in the rat brain cortex, by reductions in the tissue and plasma levels of beta-endorphine and met-enkephalin. A daily administration of reaferon in a dose of 10,000 IU during two fortnights completely restored both the binding affinity of the receptors and the concentrations of the peptides tested: those of beta-endorphine in the adenohypophysis and plasma and those of metenkephalin in the adrenals and plasma.

[Immunoenzyme analysis of ferritin in diagnosing brain tumors].

A procedure was developed to purify ferritin from the human brain tissue. The preparation is a heavy chain of ferritin. The level of ferritin in biological fluids was evaluated using the sandwich solid-phase immunoassay. This fraction of ferritin was found in the cerebrospinal fluid of patients with brain tumors. Content of the ferritin heavy chain in cerebrospinal fluid correlated with the rate of brain tissue malignancy.

[Features of metabolism of the human myocardium during alcoholic intoxication].

Systems of alcohol oxidation, malate-aspartate shunt, content of total and alpha-cholesterols in heart muscle and blood as well as morphometry of aorta were studied. The tissues investigated were obtained post mortem in lethal traumatic injury accompanied by increased content of ethanol in blood. The potentiating effect of alcoholization on atherogenesis was not found.

[Opiate reception in the rat brain: the effect of chronic consumption of alcohol and reaferon].

Binding properties of opiate receptors were studied in brain tissue of alcoholized rats after administration of the interferon-derived drug reaferon using equilibrium radioreceptor assay. Chronic alcohol intoxication was accompanied by a decrease in affinity of opiate mu-receptors for ligands in the animals with alcohol intoxication and ethanol withdrawal. Alterations in parameters of delta-reception and in content of mu-receptors were not found. Daily administration of reaferon at a dose of 10(4) U within 1.5 month did not affect these impairments, observed in rats, studied in intoxication. However, the drug reduced completely the alcohol effect on receptors during the withdrawal period. Possible normalizing effects of reaferon on the state of opiate receptors are discussed.

[Autoantibodies against nerve tissue proteins long after cranio-cerebral injury].

20 samples of serum of patients with remote consequences of brain trauma were tested to detect autoantibodies (a-AB) against brain proteins S-100 and NKP. Essential pathology was not found out by standard solid-phase immunoassay. The original method was used to detect a-AB to the whole complex of antigen determinants. Using this method the elevation of a-AB against S-100 and NKP was shown at 16 cases. The disturbance of idiotype-antiidiotype regulation was revealed at this group of patients. It is suggested that insufficient level of corresponding antiidiotypes creates the conditions for autoimmune aggressor of a-AB against brain tissue.
