[Enzymatic characteristics of a peroxisome fraction isolated by the use of hypotonic treatment of subcellular structures].

A peroxisome fraction was isolated from rat liver tissue using hypotonic treatment of mitochondria and lysosomes. In the fraction obtained activity of marker enzymes was studied in various subcellular structures. Content of mitochondria, lysosomes and microsomes in the peroxisome fraction was about 15-20% of the total protein in the fraction. The results of hypotonic treatment of lysosomes and mitochondria are considered. Flotation activity of lysosomes was decreased to 1.16-1.20 g/cm3 after osmotic lysis and elimination of matrix enzymes from these organelles.

[Glutathione: dehydroascorbate-oxidoreductase activity in rat tissues].

Glutathione: dehydroascorbate reductase activity was studied in rat liver, heart, spleen, lungs as well as in Zajdela hepatoma. Correlation between the activities of glutathione reductase and glutathione: dehydroascorbate reductase was observed in all the tissues studied. Glutathione: dehydroascorbate reductase activity was higher in Zajdela hepatoma as compared with the rat liver. The role of glutathione: dehydroascorbate reductase in regulation of antioxidative activity and of cell division is discussed.

[Effect of short-term hypokinesia on the response of the opioid system in the rat].

The content of methionine-enkephalin, leucine-enkephalin and beta-endorphine was measured in various brain compartments (hypophysis, hypothalamus, midbrain, medulla oblongata, striatum), adrenals and plasma of the rats exposed to single and repeated immobilization. The reaction of the opiate systems to immobilization was very distinct in the emotiogenic brain structures (hypothalamus and midbrain) and hypophysis. The content of opiate-like peptides varied as a function of the immobilization time, with the most distinct changes occurring at the 150th minute. After daily immobilization repeated 40 times adaptation to the chronic stress-effect developed.

[Effect of ethanol and clofibrate on the activity of lipid catabolism enzyme systems in rat liver].

Male rats were fed ethanol as a constituent of an isocaloric diet. Ethanol administration caused hypercholesterolemia and accumulation of lipids in the liver. However, it did not alter enzymatic activity of fatty acids and cholesterol catabolism in the organ. Addition of the hypolipemic agent clofibrate to the ethanol-containing diet reduced the adverse effects of ethanol on lipid metabolism due to activation of peroxisomal and mitochondrial enzymes involved in oxidative degradation of lipids in the liver cell.

[Effect of chronic administration of ethanol on the enzyme activity of rat serum, liver and brain].

Effect of chronic ethanol administration on some enzyme activities was studied in plasma membranes, brain homogenate cytoplasmic reticulum and cytosol, liver homogenate and microsomal fractions and blood serum. Ethanol was ingested as a constituent of isocaloric "semiliquid" diet. The investigation was carried out to estimate the diagnostic value of certain enzymes in evaluation of alcohol intoxication. In male rats ethanol caused remarkable hyperlipidemia, accumulation of lipids in liver tissue and elevation of gamma-glutamyl transpeptidase activity in blood serum and brain tissue. In liver tissue moderate induction of glucose-6-phosphatase, NADPH-cytochrome c reductase and alkaline phosphatase was observed. The putative mechanism of elevation of organospecific enzyme activities in blood serum during chronic ethanol consumption is discussed.

[Superoxide radical and superoxide dismutase in the free-radical theory of aging (a review)].

Review is presented on the role of superoxide radicals and other intermediates of single electron reduction of oxygen in aging as well as on geroprotective importance of superoxide dismutase. These factors are considered in relation with age. Significance of direct and indirect methods for estimation of total O2-radicals is analyzed. Possible role of O2-radicals and superoxide dismutase in regulation of cell division and differentiation is also discussed. Life span of cells in peripheric circulation correlates with superoxide dismutase content in these cells and their ability to generate exogenous O2-.
