Localization of uridine monophosphate synthase (UMPS) gene to river buffalo chromosomes by FISH.

Uridine monophosphate synthase plays an important role in pyrimidine synthesis, converting orotic acid to uridine 5' monophosphate. In cattle, UMPS deficiency is inherited as a monogenic autosomal recessive trait. While heterozygous carriers are phenotypically normal, homozygotes are lethal in utero (Shanks et al. 1992). UMPS has been mapped to human chromosome 3q13 (Qumsiyeh et al. 1989), sheep chromosome 1q (Burkin et al. 1993) and cattle chromosome 1q31 (Ryan et al. 1994). In the present study we used a cattle genomic probe to localize UMPS to river buffalo chromosomes by fluorescence in situ hybridization.

Chromosomal localization of the lysozyme gene cluster in river buffalo (Bubalus bubalis L.).

Lysozyme (LYZ) is an antibacterial enzyme which allows the digestion of bacteria present in tears and saliva. In the true stomach of ruminants LYZ breaks open the bacteria of the foregut, which are subsequently digested by typical mammalian digestive enzymes, allowing the incorporation of nutrients from the bacteria. Southern analysis with a single exon from a cow lysozyme gene revealed that there are about 10 genes in ruminants (Irwin & Wilson 1989), while pig and primates have a single lysozyme gene (Swanson et al. 1991) and camels have two (Irwin et al. 1992). The higher number of LYZ genes in ruminants is believed to be the result of gene duplication associated with the evolution of foregut fermentation (Irwin et al. 1992). Recently, the genomic organization of the lysozyme gene family has been determined in domestic cattle, and, using a cocktail of genomic clones, the lysozyme gene cluster (LYZ/) was assigned to chromosome (Chr) 5, band 23 by fluorescence in situ hybridization (FISH) (Gallagher et al. 1993). In our continued effort to test the genetic homology of conserved chromosome banding regions between cattle and river buffalo, and to extend the river buffalo physical gene map, we have mapped the LYZ/ by FISH and R-banding.

Cloning and sequence analysis of A cDNA encoding bovine cytosolic aspartate aminotransferase.

1. Complementary DNA encoding cytosolic aspartate aminotransferase was isolated from an adult bovine heart library. 2. The amino acid sequence deduced for the protein (412 amino acids) is extremely similar (> 94% identity) to that of porcine cytosolic aspartate aminotransferase but interesting differences were noticed comparing the position of cysteine residues.

Acetohydroxy acid synthase and threonine deaminase activities, and the biosynthesis of isoleucine-leucine-valine in Streptococcus bovis.

Acetohydroxy acid synthase (AHAS) and threonine deaminase (TD) activities were found in Streptococcus bovis and shown to be involved in the biosynthesis of the branched chain amino acids isoleucine, leucine and valine. Apparent lack of repression of AHAS synthesis by the end-products and reduced sensitivity of S. bovis growth to analogues of the branched chain amino acids suggested that secretion of isoleucine, leucine and valine in the growth medium may be a consequence of the regulatory features of AHAS. A glycyl-leucine-resistant mutant with reduced TD activity secreted a reduced amount of isoleucine and an increased amount of valine, which might be a result of the reduced rate of synthesis of the isoleucine precursor alpha-ketobutyrate and of a consequent preferential carbon flow through the valine branch of the pathway.

A new centric fusion translocation in cattle, rob(16;18).

A Barrosã bull (Portugal) has been found to carry a new Robertsonian translocation involving chromosomes 16 and 18 of standard cattle karyotype, as demonstrated by GBG- and RBG-banding techniques. C-banding patterns revealed the dicentric nature of this translocation. A comparison between normal cattle chromosome 16 and the q-arms of translocation chromosome and river buffalo chromosome 5 revealed the same G- and R-banding patterns, with only exception of a pericentromeric G-positive band which has been lost in river buffalo 5q and conserved in normal cattle chromosome 16 and rob(16;18) q-arms.

Sister chromatid exchange in the goat (Capra hircus).

Lymphocyte cells from 30 Maltese and Syrian derivative goats (13 males and 17 females) reared in southern Italy underwent sister chromatid exchange (SCE) test. For the 902 cells we studied, the SCE-mean values were 6.6 +/- 3.0 per cell for both breeds. The SCE-frequency did not follow a Poisson distribution. The simultaneous visualization of SCEs and G-bands in the lightly stained chromatid allowed the study of SCE-distribution in chromosomes 1 and X. The number of SCEs in chromosome 1 was significantly higher (P > 0.001) than expected from relative chromosome length. No statistical differences between the numbers of SCEs in the active (early-replicating) and inactive (late-replicating) X-chromosomes of female cells were found.

Identification and characterization of the proBA operon of Streptococcus bovis.

A genomic DNA library of the rumen bacterium Streptococcus bovis was constructed in Escherichia coli, and recombinant plasmids able to complement proA and proB mutations of the host were found. Southern hybridization and restriction analysis showed that a 3.5-kb fragment of S. bovis DNA contained two genes, organized in an operon and coding for enzymes functionally similar to the glutamyl phosphate reductase-glutamyl kinase enzyme complex that in E. coli catalyzes the first step of proline biosynthesis. Complementation of the E. coli mutations was observed with the fragment inserted in both orientations, which suggested that the S. bovis proBA operon was transcribed from its own promoter. Genetic and biochemical data suggested that the proline biosynthetic pathway of S. bovis is similar to the one previously characterized for E. coli.

[Pollution due to automobile traffic in the region of Presidi Cardarelli, Santobono e Presidio Multizonale di Prevenzione della USL 40 di Na di Napoli].

A control of air pollution has been performed in the Hospital Area of Naples during a period of two years, from 1988 to 1990. The concentration of inert suspended dust, lead and the respirable fraction of both components has been determined to obtain information about air pollutants produced by vehicular traffic which is very high in the observed zone.

Identification of nucleolus organizer chromosomes in sheep (Ovis aries L.) by sequential GBG/Ag-NOR and RBG/Ag-NOR techniques.

Seven Gentile di Puglia sheep (Ovis aries L., 2n = 54) reared in southern Italy were cytogenetically investigated to reveal which chromosomes carry nucleolus organizer regions (NORs). The use of sequential GBG/Ag-NOR and RBG/Ag-NOR techniques in prometaphase preparations revealed NOR-localization at the telomeres of chromosomes 1p, 2q, 3q, 6 and 25 on the basis of the standard R-banded karyotype. This procedure also permitted the characterization of G- and R-banding patterns of acrocentrics 6 and 25 which are often confused with the closest pairs in sheep and related species karyotypes. Indirect comparisons among nucleolus organizer chromosomes (NOCs) of sheep, goat, cattle and river buffalo, when using a common R-banding nomenclature, revealed that (a) sheep and goat have the same five homologous NOCs, (b) sheep and cattle have three homologous NOCs (c) sheep and river buffalo have two NOCs in common, and (d) the four species have only one homologous NOC in common.

[Surveillance of the circulation of arbovirus of medical interest in the region of eastern Senegal].

A study about the circulation of arboviruses of medical interest in southeastern Senegal was conducted from 1988 to 1991, during and around the periods of transmission. Specific IgM antibodies were detected by ELISA test in human sera, as a sign of a recent infection within 2 to 5 months. The comparison of the serological IgM results from human surveys in different villages, and the isolations of arboviruses from mosquitoes during the same period of time permitted a rapid and global evaluation of the circulation of these viruses. A low level of yellow fever virus activity was detected both in humans and mosquitoes in 1988 to 1990. A dengue 2 epizootic occurred in 1989-1990. Dengue 2 virus was isolated from humans and mosquitoes in 1990. Some dengue 2 outbreak may occur in the upcoming years. A Zika virus epizootic outbreak was observed each year. A human strain was isolated in 1990. The other flaviviruses (West-Nile, Kedougou, Wesselsbron), Chikungunya virus, Rift Valley Fever virus and Crimean-Congo Hemorrhagic Fever virus did not seem to present a major public health concern in southeastern Senegal.

[Analytical evaluation of significant chemical and physico-chemical parameters in water for human consumption].

A quality control of drinking water has been performed in hospitals and schools located in the Vomero-Area of Naples. Some chemical pollutants (iron, manganese, ammonia nitrogen, nitrite, nitrate, fluoride) as well as chemical and physical parameters (hardness, pH, electric conductivity, chloride, cations) have been periodically analyzed from Autumn 1991 to Summer 1992.

Isolation of mouse Lyt2- mutant cells using magnetic particles.

A procedure is described for the selection of mouse Lyt2- mutant cells. The procedure is based upon repeated cycles of selection with rat monoclonal antibodies to the Lyt2 antigen and magnetic particles coated with goat anti-mouse IgG. Stable Lyt2- mutant clones were derived from cells previously mutagenized with X rays and, at a lower frequency, also from non-mutagenized cells. The procedure can be used to rescue Lyt2- cells when the ratio of Lyt2+:Lyt2- cells is 10(4):1.

Evolution of ruminant hemoglobins. Thermodynamic divergence of ox and buffalo hemoglobins.

The ligand-binding properties of hemoglobins from two homozygote phenotypes (AA and BB) of water buffalo (Bubalus bubalis) have been characterized by equilibrium and kinetic techniques. In the case of the BB phenotype, the two constituent hemoglobins have been purified and separately analysed. Buffalo hemoglobins display the reduced sensitivity to organic phosphates characteristic of ruminant hemoglobins, their physiological effector probably being the chloride ion. In contrast to the other known hemoglobins from ruminants, all the hemoglobins from the water buffalo display a significant temperature sensitivity, the delta H for oxygen binding in the presence of physiological effectors approaching that of human hemoglobin (delta H = -30.5 kJ/mol O2). This discrepancy with the other ruminant hemoglobins (e.g. ox, delta H = -10.4 kJ/mol O2), whose primary structure is very similar to that of buffalo, hemoglobins might be correlated to the different habitat and phylogenetic history of the two subfamilies (Bos and Bubalus) of Bovidae.

Detection of a sperm-coating antigen in the semen of Bubalus bubalis.

1. Surface antigens of B. bubalis spermatozoa were solubilized by Triton X-100 and EDTA; the sperm extract was used to raise antibodies in rabbits. 2. Two major polypeptides, immunoprecipitated from the seminal plasma by the antibodies against the sperm extract, exhibited the same electrophoretic mobilities of two immunorelated sperm surface antigens. 3. The two polypeptides were isolated from the seminal plasma, by a multi-step chromatographic procedure, and found subunits of a single protein (MW 30,000), called SP 30. 4. The SP 30 protein bound in vitro to the postacrosomal region of homologous spermatozoa from cauda epididymis. 5. The localization of the sperm-coating antigen on the cell surface is compatible with a role in the fertilization process.

A new Robertsonian translocation in cattle, rob(15;25).

A new Robertsonian translocation, rob(15;25), was discovered in a Portuguese Barrosa cow. The animal (2n = 59,XX) was found by G- and R-banding to be a heterozygous carrier of a centric fusion translocation involving chromosomes 15 and 25. C-banding revealed the dicentric nature of this new centric fusion. Comparison of this new translocation with the well-known Robertsonian translocation rob(1;29), which is often found in the same breed, confirmed that two different chromosomes (25 and 29) were involved in the short arms of these two Robertsonian translocations.

Evidence for the presence of two different beta-globin chains in the hemoglobin of the river buffalo (Bubalus bubalis L.).

1. Hemoglobin (Hb) of the river buffalo (Bubalus bubalis L.) was studied by employing isoelectric focusing (IEF) in the 6.7-7.7 pH range and by IEF in ultra-narrow immobilized pH gradient (IPG) 7.1-7.5. 2. Three Hb BB phenotypes were identified which were characterized by sets of two or four Hbs with different isoelectric points. 3. These type were called BB, BsBs and BBs, the Bs phenotype showing Hbs with slightly slower mobility. 4. Analysis of constituent globin chains by acid urea polyacrylamide gel electrophoresis in the presence of Triton X-100, provided clear evidence of a novel polymorphism at the beta-globin level. 5. Titration curves of beta-globins from heterozygous Hb BBs indicated a single curve thus suggesting the absence of a net charge in all the pH field. 6. A neutral-to-neutral amino acid replacement probably differentiates the two beta-globins.

Sister chromatid exchange in chromosomes of cattle from three different breeds reared under similar conditions.

A homogeneous group (same sex, age and environmental conditions) of 35 Italian cattle of the Podolian, Friesian, and Romagna breeds was investigated concerning the spontaneous incidence of sister chromatid exchange (SCE). The mean values of SCEs/cell were 7.9 +/- 3.4, 7.1 +/- 3.3, and 7.3 +/- 3.2 in the Podolian, the Friesian, and the Romagna breeds, respectively, with significant differences between the Podolian and the Friesian breeds. Simultaneous disclosure of SCEs and fluorescent G-bands in the lighter chromatid made possible the identification of chromosome 1, in addition to the biarmed X and Y chromosomes. The inter-chromosomal SCE-distribution revealed a nonrandom pattern due to significantly increased values of SCEs in chromosomes 1 and X, particularly in the Romagna breed.
