Electrophoretic and chromatographic evidence for allelic polymorphisms in the river buffalo alpha-globin gene complex.

Isoelectric focusing in the ultranarrow immobilized (7.1-7.5) pH gradient (IPG) of hemoglobin and high-performances liquid chromatography (HPLC) of globin chains were used to investigate Hb polymorphism in Italian river buffalo. Six different phenotypes, each characterized by two or four different Hbs, were detected by IPG, whereas two different II alpha-globin chains were separated from two different I alpha-chains by HPLC. Two alpha-chains (I alpha 1 and II alpha 3), and Hbs with similar mobilities (Hb1 and Hb3), were associated with the AA Hb phenotype: two alpha-chains (I alpha 2 and II alpha 4), and Hbs with different mobilities (Hb2 and Hb4), were associated with the BB phenotype: two sets of doublet Hbs were associated with the AB phenotype, thus suggesting allelic polymorphisms at the two alpha loci. An allele at the beta locus is responsible for increasing to as many as eight the number of different Hbs, thus further complicating the notable Hb polymorphism of the river buffalo.

[Varicocele and pregnancy. Comparison of treated and untreated patients].

A study was made of 209 cases of sterility due to male varicocele out of a series of 1730 infertile couples. Corrective surgery was performed in 38 cases with 8 subsequent pregnancies (21.5%). A total of 31 pregnancies were achieved in the 171 cases which did not undergo surgical treatment (18.7%).

[The role of Papanicolaou test in gynecologic practice].

The paper analyses the importance of Papanicolaou's test in gynecological diagnostics and reports an interesting checklist drawn up by the "Istituto Superiore di Sanità" with which all women should be familiar in order to diagnose cervical cancer at an early stage.

Time-resolved fluoroimmunoassay of progesterone in milk.

A direct, solid-phase, time-resolved fluoroimmunoassay for progesterone in cow's and goat's milk, using europium-chelate-protein A as a label, is described. The coefficients of correlation with the results by RIA were 0.987 and 0.989.

The high resolution G- and R-banding pattern in chromosomes of river buffalo (Bubalus bubalis L.).

High resolution G- and R-banding patterns in chromosomes of river buffalo (Bubalus bubalis L.) were obtained by using early (G-bands) and late (R-bands) BrdU-incorporation in synchronized cell cultures. To better characterize the river buffalo chromosomes, GTG-, GBG-, and RBG-techniques were used. The total number of bands achieved were 490 (207 G-positive, 207 R-positive, 45 variable, and 31 centromeric regions). Only one common G- and R-banding nomenclature was reported. The number, position and intensity of G bands were highly similar by the structural GTG and the replicating GBG-techniques. However, the replicating G- and R-bands appeared to be more distinct and reproducible than the structural G-bands. Some changes in chromosome nomenclature (chromosomes 1p, 2p, 5p, and 21) were made when referred to the cattle homologues.

[Respiratory distress syndrome in premature births].

The paper reports the results of a study of respiratory distress syndrome in premature birth, one of the most common and important forms of respiratory disorders affecting neonates during the first days of life. One hundred and fifty-six premature births from 1 January 1986 to 30 June 1988 were included in the study. From an analysis of results it emerges that there is a lower incidence of respiratory distress syndrome in both vaginal deliveries and cases treated with Ambroxol.

[Clinical study of the use of a new tri-phasic pill].

The paper reports the results of a study to assess the contraceptive efficacy of a new triphasic pill (Trinovum - Cilag AG). The pill was administered to 50 women aged between 18 and 41 years for 12 months, giving a total of 600 menstrual cycles. Data confirm the value of this pill which ensures excellent contraceptive efficacy and is well tolerated by patients with very few adverse effects.

Evaluation of time-resolved fluoroimmunoassay with Eu-labelled protein-A for serum progesterone.

Progesterone has been assayed in several serum samples by a time-resolved fluoroimmunoassay (TR-FIA). The solid phase was 11 alpha-hydroxyprogesterone hemisuccinate bound covalently to ovalbumin and adsorbed on wells of polystyrene. The assay was based on competitive reaction of solid phase-bound hormone and samples with specific antibody labelled in situ with protein-A prelabelled with europium. The bound Eu was dissociated from the solid phase by an enhancement solution and measured by time-resolved fluorometry. The coefficient of correlation between TR-FIA and RIA was 0.97.

Water buffalo (Bubalus bubalis) hemoglobins: an electrophoretic and chromatographic study.

1. Hemoglobins from three phenotypes of Italian water buffalo (Bubalus bubalis), named AA, AB and BB, were selected by starch gel electrophoresis at alkaline pH and analyzed using polyacrylamide gel isoelectric focusing and subsequent analysis of titration curves to reveal differences between two types of hemoglobin identified as Hb fast and Hb slow. 2. Globins from Hb fast and Hb slow were purified by fast protein liquid chromatography (FPLC). Electrophoretic differences were found in the respective alpha-chains using polyacrylamide gel disc-electrophoresis at acid pH, polyacrylamide gel isoelectric focusing and by subsequently analyzing titration curves. 3. The results suggest that the alpha chains of Hb fast and Hb slow, called I alpha and II alpha, respectively, differ in at least two aminoacid residues. Subsequently, these amino acids were identified as lysine and cysteine.

High-resolution studies on late-replicating segments (G + C bands) in mammalian chromosomes.

High resolution of late-replicating segments (G + C bands) in chromosomes of human, cattle and water buffalo was obtained by using 5-bromodeoxyuridine incorporation early in the cell cycle, simultaneous with methotrexate treatment combined with reduced colcemid treatment and addition of ethidium bromide, which increased the proportion of prometaphase cells. Giemsa counterstaining, following fluorescence microscopy observation and treatment with 2 x SSC, improved the resolution of the banding patterns, particularly in the pericentromeric regions. Acrocentric bovine and water buffalo chromosomes, which were seen to be C-positive by fluorescence microscopy observation and C-negative after counterstaining, showed the presence of subcentromeric G-positive bands within the heterochromatic blocks of several chromosomes.

[Purification of waste water from olive-oil pressing plants using hydrophytes. Preliminary study on the use of Eichornia crassipes].

The use of Eichornia crassipes has been tested in order to depurate the waste water from olive oil factories. The control of some parameter variations during the treatment, such as COD, total azote, dissolved oxygen and pH, has given encouraging results about the depurative efficacy of employed hydrophytae. Particularly indicative is, in this regard, COD rejection of about 60%.

[Identification of the root of Punica granatum in galenic preparations using TLC].

The alkaloids present in the root of "Punica granatum" have been extracted by two different methods: extraction by Soxlet and extraction by steam distillation. Then the extracts have been compared by TLC chromatography using different solvents and specific chromogen reagents. The presence of pseudo-pelletierine has been confirmed in both the extractive solution by reaction with conc. K2Cr2O7. The above results explains the toxic activity of the unsuitable galenic preparations.

Bilateral testicular injury from external trauma.

Among 79 patients with testicular injury as a result of external trauma 5 had bilateral injury. Such injury occurred in only 1 of 66 patients (1.5 per cent) with blunt trauma versus 4 of 13 (31 per cent) with penetrating trauma. Bilateral ruptures were managed by bilateral partial orchiectomy in 2 cases, and orchiectomy with contralateral débridement and suturing in 1. Of 2 patients with unilateral rupture and contralateral contusion the contusions were treated by evacuation of the hematoma and the ruptures were managed by orchiectomy in 1 and partial orchiectomy in the other. Four patients were available for followup. One patient was hormonally anorchid but he obtained normal erections with testosterone cypionate, 1 had normal serum testosterone concentrations but borderline normal semen analysis, and 1 had reduced erections and borderline normal semen. The remaining patient had normal erections and no semen analysis was performed. The nature of the injury rather than the type of treatment appears to be the primary determinant of the outcome in these cases.
