Comparative genomic hybridisation for the analysis of chromosomal imbalances in solid tumours and haematological malignancies.

Comparative genomic hybridisation (CGH) is based on a two-colour, competitive fluorescence in situ hybridisation of differentially labelled tumour and reference DNA to normal metaphase chromosomes. This new technology has made a great impact in molecular tumour pathology due to its possible application to archival specimens and the ability to create copy number karyotypes throughout the whole genome from very small amounts of DNA. If chromosomal imbalances can be correlated with a etiological and clinical features of tumours, CGH could be able to provide new prognostic and diagnostic criteria. CGH findings further provide starting points for the molecular genetic characterisation of altered chromosomal regions harbouring yet unidentified genes involved in tumorigenesis and tumour progression. An overview of the results of published CGH studies on solid tumours and haematological malignancies is presented. Methodological limitations of the CGH technology are reported, as well as future developments which will improve its use in routine analysis.

[Patellar osteochondrosis dissecans--results of surgical therapy].

Osteochondritis dissecans of the patella is a rare condition. 15 patients who had been treated by surgery were evaluated retrospectively by clinical examination, standard radiographs of the patellofemoral joint and using the Lysholm-Score. The results of treatment were classified as good, sometimes excellent. The worst results were found in centrally located lesions or in chondromalacia changes occurred at the time of surgery.

Intravenous angiography using scanned projection radiography: preliminary investigation of a new method.

The use of digital subtraction techniques combined with fluoroscopy has rekindled interest in arteriography using intranvenous injections of contrast media. A new method is proposed for intravenous angiography in which an x-ray source and xenon detector array from a computed tomographic (CT) scanner are used to scan a region of interest to produce projection image. In order to provide adequate visualization of small concentrations of iodine in blood vessels, a subtraction scheme is used to remove the contribution from overlapping soft tissue and bone. Initial experiments with a temporal subtraction algorithm on phantoms have demonstrated the ability to image simulated blood vessels of 1.7-mm diameter containing dilute diatrizoate with an iodine concentration of 3.7 mg/cc, at an exposure of less than 100 mR. Vascular structures 5-8 mm in diameter have been imaged in dogs with iodine concentrations of less than 37 mg/cc using temporal subtraction. Principal advantages of the method over other film or fluoroscopic subtraction techniques are: 1) wide dynamic range an low noise of the (CT) detectors, providing excellent iodine sensitivity; 2) high scatter rejection; and 3) efficient utilization of x-ray dose.

Do urea/ammonium lactate combinations achieve better skin protection and hydration than either component alone?

Twenty subjects with healthy skin were treated with the following formulations for two weeks: drug-free W/O vehicle, 5% ammonium lactate (CAS 52003-58-4) in W/O vehicle, 5% urea (CAS 57-13-6) in W/O vehicle, 3% ammonium lactate + 3% urea in W/O vehicle, 5% ammonium lactate + 5% urea in W/O vehicle. These formulations were applied in randomized order to 6 test areas on the forearms; one area was left untreated. Repetitive washings were additionally performed in the second treatment week. TEWL, stratum corneum water content (corneometry-determined electrical capacitance), and cutaneous blood flow (laser Doppler) were measured at baseline, day 7, and day 14. 5% urea and both ammonium lactate/urea combinations produced significant stratum corneum hydration and improved stratum corneum barrier function; there were no significant differences between these three treatments.
