[Preparation of mouse antiserum against isologous aggregated immunoglobulins and its effect on rosette forming cells].

A method of obtaining antisera against isologous aggregated mouse immunoglobulins is described. This serum designated MAAS blocked in vitro the antigen-binding receptors of the immune rosette-forming cells. MAAS was injected to mice immunized with SRBC. In comparision with the immunized mice given normal isologous serum rosette-forming B-cells were absent in the spleen of mice given MAAS at the peak of isologous response. But the antibody-forming cell count was not decreased under the influence of MAAS.

[Antigen-dependent induction of a non-specific humoral factor blocking rosette-forming cells in experiments in vitro and in vivo].

Isologous serum of CBA mice immunized with rabbit immunoglobulins (ARIS) contained a factor capable of inactivating rosette-forming splenocytes (RFC) in vitro from the same strain of mice immunized with SRBC. When mouse SRBC immunization was carried out against the background of ARIS injection the court of RFC to SRBC at the peak of immune response was 30% of that of mice injected with SRBC and normal isologous serum. A decrease of RFC count was the result of disappearance of the theta-negative RFC. Passive ARIS immunization failed to influence the antigen-induced proliferation of the antibody-forming cells and the synthesis of antibodies against SRBC.

[Molecular genotyping of Mycobacterium tuberculosis strains isolated from patients in the Samara region by the restriction DNA fragment length polymorphism].

The typing of 106 M. tuberculosis (MBT) strains isolated from patients in the Samara region by the restriction DNA fragment length polymorphism (RFLP) IS6110 revealed that most of the strains (71.7%) belonged to the W family, 5 MBT strains (4.7%) belonged to the AI family, one culture was the mixture of two strains, AI and W. In addition, 24 MBT strains (22.6%) classified with other genotypes were detected. The analysis of the sensitivity of the MBT strains to rifampicin and isoniazid, with the method of absolute concentrations and by point mutations, demonstrated that 29 MBT strains (27.3%) were sensitive to rifampicin and isoniazid and 56 MBT strains (52.9%) were resistant to rifampicin and isoniazid simultaneously. Among the MBT strains of different RFLP families, strains both sensitive and resistant to these two preparations could be detected, but strains with multiple drug resistance prevailed in the W family (61.8%).

[The identification of the antigenic determinants of Mycobacterium leprae passed through laboratory animals by using monoclonal antibodies].

The antigenic preparations of M. leprae isolated from the biopsy leproma material of leprosy patients, M. leprae obtained by passage through laboratory animals (armadillos, rats) and M. lepraemurium have been identified by indirect solid-phase enzyme immunoassay. Experiments made with the use of the set of monoclonal antibodies (McAb) to different M. leprae antigenic determinants, supplied by the WHO Bank of Monoclonal Antibodies (IMMLEP), have demonstrated the presence of common antigenic determinants in 65-kD protein of M. leprae obtained by passage through armadillos and rats and M. lepraemurium, as well as in 12-kD protein of M. leprae isolated from armadillos and rats and M. leprae isolated from the biotic material of human lepromas. The passage of M. leprae through experimental animals leads to the appearance of a new antigenic determinant in their structure, which is recognized by McAb 111E4.

[Improving of methods of production and screening of monoclonal antibodies to Salmonella antigens].

Methods for the production of and screening for monoclonal antibodies (MAB) to Salmonella antigens have been optimized. Schemes for immunization of animals have been developed for various Salmonella antigens. Prolonged (up to 6 months) immunization is needed to obtain antibodies in high titers to 0-antigen, short (up to 1 month) to obtain antigens to K-antigen. Affinity purification is desirable if MAB to bacterial antigens from ascitic fluid are used. Study of the kinetics of MAB binding to antigens helps appreciably accelerate enzyme immunoassay. Microplates manufactured by Medpolimer National Research Institute (St. Petersburg) may be recommended as the solid phase. Glutaraldehyde treatment of the plates to improve their adsorption capacity is not recommended.

[The practical use of monoclonal antibodies to Mycobacterium tuberculosis].

The article deals with the practical use of monoclonal antibodies to M. tuberculosis. As shown in this work, the use of monoclonal antibodies permits the determination of antibodies to M. tuberculosis and mycobacterial antigens in the spinal fluid of patients with tuberculous meningitis.

[Specific antisera to mycobacteria].

Antisera to several Mycobacterium species (M. bovis, M. avium, M. kansasii and M. smegmatis), capable of reacting with homologous antigen diluted not below 1:16000 in the ELISA test, have been obtained. These antisera have been found to cross-react with other Mycobacterium species. Specific antisera to M. bovis, M. avium, M. kansasii and M. smegmatis have been obtained by the multiple exhaustion method on Mycobacterium antigens giving the maximum cross reaction and used for the isolation of antibodies to these Mycobacterium species. In ELISA exhausted antisera produce reactions in the dilution 1:500. Antisera (antibodies) to M. bovis react with the determinant of the antigen with a molecular weight of 25-26 kD, antisera (antibodies) to M. kansasii react with the determinant of the antigen with a molecular weight of 72 kD, those to M. smegmatis react with the determinant of the antigen with a molecular weight of 38 kD and those to M. avium, with the determinant of the antigen with a molecular weight of 16-18 kD.
