[Detection of Chlamydia in inflammation of the cervix and uterus in cows].

Clinical findings in cows of seven farms with a frequent occurrence of birth canal inflammations were matched against results of serological and direct immunofluorescence tests for Chlamydia sp. infection. Normal clinical picture, weak to moderate and serious inflammations were found in 16.1%, 46.7% and 37.2% of the 205 examined cows, respectively. The results of micro CFT were negative in 32.7% and titres 1:5, 1:10, 1:20, and 1: > or = 40 were found in 8.7%, 38.0%, 12.3%, and 8.3%, respectively. Direct immunofluorescence test of cervical swabs, using the Progen Biotechnic GmbH set, was negative in 29.1% and clearly positive in 34.7% of the tested cows. The rest was animals in which sporadic elementary bodies were seen. The significance of the findings is discussed and the necessity to test Chlamydia infections in herds affected by frequent and/or chronic inflammations of genital organs is stressed. Evidence of the transmission of Chlamydia infection by contaminated semen is presented.

[Detection of chlamydia in the intestines and lungs in pigeons and humans].

Attempts were made to detect Chlamydia sp. by direct immunofluorescence (IF) in intestinal and lung tissues collected from wild town pigeons, young reared pigeons, and human corpses. Chlamydia in any organ were detected in 86.6% of the 427 adult pigeons and 27.3% of 16 young pigeons. Positive IF results were in intestinal and lung samples, collected from 427 adult pigeons, in 34.3% and 40.05% of the birds, respectively. Human (n = 100) intestinal and lung tissues were positive in 22% and 20% of the corpses, respectively. Titres 1:20 and higher were demonstrated by CF in 53.86% of the 427 adult pigeons. The highest titre found was 1:5 120.

Antigenicity of Dermatophilus congolensis hemolysin.

The separated cell-free form of hemolytic exosubstance was obtained from five strains of Dermatophilus congolensis. Three strains produced exosubstance with high activity, two strains produced exosubstance with lower intensity of activity. The separated forms exhibited the same hemolytic interactions as the native forms produced by growing strains, namely the antagonism with staphylococcal beta hemolysin and the synergism with staphylococcal delta hemolysin, streptococcal CAMP factor and rhodococcal equi factor. Rabbit sera obtained after intravenous or intraperitoneal application of the separated forms contained precipitation and neutralization antibodies. Cross tests of precipitation and neutralization proved antigen identity of hemolysins of different D. congolensis, strains which makes the serodiagnostics of this species possible.

Hemolytic interactions of Dermatophilus congolensis.

The strains of Dermatophilus congolensis grew on blood agar with washed sheep erythrocytes with marked total hemolysis. In testing for hemolytic interactions they gave a significant synergistic effect of a characteristic shape with Rhodococcus equi and Streptococcus agalactiae, whereas with Staphylococcus aureus producing beta hemolysin and with Staphylococcus aureus producing delta hemolysin a simultaneous synergistic as well as antagonistic effect were observed. First of all a conspicuous inhibition of in the beta hemolysin zone began and then the hemolytic effect of D. congolensis was enhanced. A similar double reaction was also observed with Listeria ivanovii. With delta hemolysin there was an inhibition of the hemolytic effect of D. congolensis and at the same time a synergistic effect could be observed. Also D. congolensis gave a weak synergistic effect with Micrococcus lylae and Listeria monocytogenes, and a further weak antagonistic effect with alpha hemolysin of Staphylococcus aureus, Staphylococcus hyicus, Staphylococcus chromogenes and Micrococcus luteus. No interaction of D. congolensis was established with Corynebacterium pseudotuberculosis.

Experimental dermatophilosis.

The histopathologic features of an experimental dermatophilosis of rabbit skin were described. The strain of Dermatophilus congolensis used for the experimental infection was obtained from the unique isolation and cultivation of this actinomycete from the ulcerous leg lesion of one male patient. Further strains in experiment came from the collections of type cultures. The experimental infection was characterized as an exudative and crustal dermatitis with acanthosis and subcorneal abscesses. Branching hyphae and clusters of coccoid bodies were found mostly in the parakeratotic layer and infrequently in the abscesses. Both the coccoid bodies and the hyphae were Gram-positive. The hyphae were acid-fast but the coccoid bodies were non-acid-fast. The fine structure of hyphae and coccoid bodies, namely of those with flagella, was also described by means of a transmission and scanning electron microscope.

Corneal phaeohyphomycosis caused by Wangiella dermatitidis.

A case is described of a corneal abscess of the left eye of a patient, male, 35 years, from which there was found in pure culture a mycotic micro-organism, Exophiala (Wangiella) dermatitidis, determined also in the native preparation from the pus. The patient is feeble-minded, suffers from von Recklinghausen's disease and has an impairment of the immune system (above all the reduction of T lymphocytes).

[Typing of skin staphylococci using the commercial identification system Staphy-Test].

The authors examined 150 strains of S. aureus and 180 strains of S. epidermidis isolated from different dermatoses. The determination of S. aureus and S. epidermidis was based on common routine tests as plasmacoagulase, production of hemolysin, acid production from mannitol and production of pigment. This determination was verified with the aid of a new identification system STAPHYtest. The STAPHYtest confirmed the previous diagnosis of S. aureus in 60.66% and that of S. epidermidis in 31.66% of strains only. The remaining strains were identified by means of STAPHYtest as S. simulans, S. xylosus, S. hominis, S. haemolyticus, S. warneri, S. auricularis, S. sciuri and Micrococcus sp. The authors discuss the importance of an curate identification of skin staphylococci by means of STAPHYtest for taxonomic purposes and for classification of staphylococci influence on different dermatoses.

A simplified ELISA method for syphilis.

A new serological method for syphilis using a modified ELISA reaction is given. The reaction is performed on polystyrene carriers which have absorbed an ultrasonicated preparation of T. pallidum. The reaction between the antigen and antibody is detected by means of the addition of antihuman IgG swine globulin conjugated with horseradish peroxidase. To make the reaction visible, addition of 5-methyl-9-aminocarbazol in dimethylformamide is necessary. In a positive case of syphilis the carrier becomes a red-brown-violet color; in a negative reaction the white color does not change. The results obtained with this reaction are in absolute concordance with those of FTA-ABS and TPHA.

Further observations of Corynebacterium renale as an indicator organism in the detection of the exfoliation-positive strains of Staphylococcus aureus.

Exfoliatin-positive S. aureus strains of known phenotypes were examined for bacteriocin production using S. aureus 209P, C. pseudodiphtheriticum, or C. renale as indicator strains. Strains of the phenotype Bac+ Tox+ had a positive result when the simultaneous method was used, and produced a wide inhibition of the indicator strains, especially of C. renale, if the deferred method was used. The strains of the phenotype Bac- Tox+ were negative upon examination by the simultaneous method, but they caused narrow inhibition zones of the indicator strains to appear with the deferred method, especially with C. renale. S. aureus strains of diverse origin used as controls were in both methods negative with regard to inhibition of indicator strains. C. renale was found to be very suitable for the detection of bacteriocins of the exfoliatin-positive staphylococci, particularly when using the deferred method.

New possibilities of staphylococcin detection in the exfoliatin-producing strains of staphylococcus aureus.

C. renale was found to be highly sensitive to the effect of bacteriocin produced by the exfoliatin-positive strains of S. aureus. The sensitivity of other indicator organisms used was determined through descending series of C. pseudodiphtheriticum, C. diphtheriae, S. aureus strains Oxford 209P, UT 0017, and Wood 46. Strains of C. pyogenes only produced contradictory results which made any satisfactory interpretation impossible. A modification of the simultaneous bacteriocin-detection method of exfoliatin-positive S. aureus strains, which use suspension of an indicator strain in nutrient agar to obtain lawn homogeneity, has been employed. The use of the modified method together with C. renale to serve as the indicator strain represents a rapid and reliable test suitable for routine diagnostics.
