Solid phase ELISA for serum ferritin.

We describe a solid-phase enzyme immunoassay for human serum ferritin, with the use of polyethylene glycol to shorten the reaction. The amount of bound enzyme-antibody conjugate used as a second site antibody is proportional to the ferritin titre in the assay. This method is fast, and the reagents are stable for many months at 4 degrees C. The lowest detectable concentration was 5 micrograms/l. Interassay coefficients of variation were 9.3 and 12.2%, respectively, at ferritin concentrations of 100 micrograms/l and 200 micrograms/l. The data also show that polyethylene glycol accelerates antigen-antibody complex formation in the solid phase. An assay may be completed in 5 h.

A solid-phase enzyme immunoassay for serum ferritin.

We describe an enzyme immunoassay for human serum ferritin in which antibody adsorbed on polystyrene tubes is used. Adsorbed gamma-globulins against human ferritin were first allowed to react with ferritin and a second antiferritin antibody, labeled with alkaline phosphatase, was added. The amount of bound enzyme/antibody conjugate was proportional to the ferritin titer in the assay. This method offers stable reagents that can be kept for many months at 4 degrees C. The average values for ferritin in normal men and women were, respectively, 58 and 43 microgram/liter. The lowest detectable concentration was 5 microgram/liter.

Elevated CEA levels in polyvinyl chloride workers.

Plasma CEA titer was measured in 200 polyvinyl chloride workers. A positive test was obtained in 48.3% of this high risk population as compared to 9.2% for a normal healthy population.

[Elevated CEA levels in a group of polyvinyl workers (author's transl)].

Carcinoembryonic antigen was assayed in plasma from a population of polyvinyl chloride workers from the same industry. We have found 56% of elevated levels in a group of workers as compared to 9.2% for a normal population including smokers and non-smokers.
