[Collagen type IV of the basal membrane of the epithelium of the skin in certain forms of pathology].

Using immunofluorescence, attenuation or complete disappearance of type IV collagen antiserum reaction were shown in the epidermal basal membrane zone of SLE patients. Some positive material was revealed in the intercellular spaces of the epidermis. There is an inverse correlation between the amount of immune complexes in the dermo-epidermal region and the intensity of anticollagen serum reaction in the basal membrane zone. This can be explained by the toxic effect of immune complexes on collagen-synthesizing cells and the disturbance of this protein fixation in the basal membrane followed by its sequestration from the affected regions. Complex therapy including hemoperfusion restores normal collagen location in the epidermal basal membrane.

[Immunomorphological (light and electron microscopic) diagnosis of human squamous cell cancer using monoclonal antibodies].

It is shown with fluorescent and peroxidase labels for antibodies that the antigen to which the monoclonal antibody A-6/2 is directed, is a marker for cells of the basal stratified epithelium (basal-cell antigen). This antigen persists in squamous cell carcinomas of various sites and degrees of differentiation, but does not occur in tumors of other origins. With electron immunocytochemistry, the antigen was found to be located in tonofilaments and desmosomes of human squamous cell carcinoma. The A-6/2 monoclonal and basal-cell antigen may be used when developing immunomorphologic (light- or electronmicroscopic) procedures for histogenetic diagnosis of these carcinomas.

[Monoclonal antibodies against group A streptococcal polysaccharide reacting with basal-cell antigen of tumors histogenetically related to epidermal tissues].

Immunofluorescence of tumour tissues histogenetically related to tissues containing cross-reacting squamous epithelial basal-cell antigen (BCAg) was performed using monoclonal antibodies (McAb) A6/I-D to squamous epithelial basal-cell antigen cross-reacting with group A staphylococcal polysaccharide. BCAg was found in tumour cells arising from surface tissues of the epidermal type (basal-cell cancer, squamous-cell cancer of the skin, esophagus, cervix uteri and other organs) and was not found in glandular tumours (adenocarcinoma of the stomach, intestine, mammary glands). The results obtained indicate that McAb A6/I-D against cross-reacting BCAg may be used while characterizing the majority of normal epidermal tissues and tumours arising from tissues containing cross-reacting BCAg.

[Comparative immunochemical and immunocytochemical study of 3 soluble granulocytic proteins in blood cells and serum].

Using immunofluorescence, it has been shown that leukocyte thermostable alpha-glycoprotein (LTAG) and leukocyte beta-globulin (LBG) are, like lactoferrin, components of polynuclear leukocytes. These proteins were found in the perinuclear zone, cytoplasm membrane and extracellularly. Serum LTAG concentration increases in immune inflammatory diseases. In severe forms of diabetes and glomerulonephritis there is a rise in LBG concentration. The biological role of LTAG and LBG is unknown.

[Reaction of loose connective tissue fibroblasts after poly-4-vinylpyridine administration].

Mice were injected with poly-4-vinylpyridine at a dose known to be effective for immunogenesis stimulation. Within 24 hours after the injection fibroblasts reacted by dendrit production, cytoplasm vacuolization and clasmocytosis. 2-3 days after the polymer injection the fibroblast cytoplasm was restored. The immunofluorescence employed rabbit serum natural antibodies to fibroblast antigens. The reaction observed may be used for the estimation of biological polymer activity.

[Effect of hemosorption on the dynamics of immunopathologic changes in the skin of patients with diseases accompanying the immune complex syndrome].

In the majority of diseases with the immune complexes (IC) syndrome, elimination of IC from the body is markedly disturbed. The authors carried out a study with the aid of the direct immunofluorsecence technique. The use of hemosorption in the treatment of patients with grave patterns of systemic lupus erythematosus, rheumatoid arthritis, psoriasis, systemic vasculitis and similar illnesses in conjunction with conventional therapy (corticosteroids, cytostatics) leads to sequestration of the depositions of immune complexes from the skin. As early as after the first hemosorption one can observe the displacement of IC from the derma to the epidermis, advance of IC towards the stratum corneum. After the third session in hemosorption IC granules appear on the body surface, in the form of warty formulations which are removed afterwards together with skin scales. It is assumed that hemosorption recovers the secretory function of the skin, aimed at IC elimination.

[Localization of basal cell antigen in the epithelial tissues of animals and humans detected by means of natural antibodies].

By means of the immunofluorescent method using rabbit serum that contains natural antibodies against the basal cell antigen of epidermis, the distribution of the antigen has been demonstrated in cells of the basal layer of all types of the stratified epithelium. The reaction is also noted in cytoplasm of the epithelial cells in the thymus and the tracheal mucous membrane. This demonstrates their histogenic affinity to stratified epithelii. The antigen studied is not species-specific, since it is revealed in the stratified epithelium of all species examined (human being, mouse, rat, guinea pig, rabbit). It is possible to use the basal cell antigen as a marker for immunomorphological reveal of epithelial cells in the thymus in the process of its physiological and pathological involution.

[Antibodies induced by group A Streptococcus reacting with epidermis elements in patients with basalomas].

Microbial mass of cultured streptococci treated with pepsin as an immunosorbent was used for isolation of an immunoglobulin factor which specifically reacted with elements of basal membrane of epidermis of basal cell cancer patients in an immunofluorescence test. The reaction took place in skin bioptates obtained both from the zone of tumor growth and distant regions. Antigens isolated from the basal membrane of skin epithelium of healthy subjects did not react with immunoglobulin factor.

[Receptors for lactoferrin on human thymic lymphocytes. The stimulation of expression as affected by adenosine, theophylline and a supernatant of thymic lymphocytes].

It was established by indirect immunofluorescence with the use of antibodies to human lactoferrin that thymic lymphocytes bear lactoferrin receptors in the point structures on the cell surface. The ability of thymic lymphocytes to express lactoferrin receptors depends on the cAMP concentration in the cell, inasmuch the treatment of lymphocytes with adenosine and theophylline increases the number of cells bearing lactoferrin receptors. Supernatant of thymic lymphocytes is also capable of stimulating expression of lactoferrin receptors. It is assumed that these treatments can be used for increasing the number of lymphocytes with lactoferrin receptors with the purpose of separation and study of the function of this subpopulation in health and in different pathological conditions.

[Elective detection of thymus epithelium and assessment of the degree of functional and pathological involution of the organ in mice].

With the help of antibodies-containing serum reacting with the thymus reticulum epithelial cells components by immunofluorescence method the thymus parenchimal tissue in mice with functional and pathological involution has been detected electively. In spite of large thymus changes in hairless mice with the mutation of gene hrrhy in 14-th chromosome of B10.R109/Y animals the basal cells antigen in epithelial reticulum has been preserved. It permits to estimate thymus involution level of this organ. Low lymphocyte content in the thymus of C57BL/6 mice is accompanied by total decamouflage of the epithelium. The functional thymus involution of pregnant mice is characterized by the luminescence of large number of epithelial cells, the restoration of organ after the delivery--by their few number. The elective detection of thymus epithelium many serve as additional test for the estimation of functional and pathological involution level.

[Immunological changes in the thymus epithelial tissue of mice during accidental involution].

Using sera containing antibodies to antigens of epithelial reticular cells it is possible to detect by immunofluorescence early stages of organ accidental involution and to trace parenchyma demasking that results from lymphocyte migration upon injection of prednisolone, azathioprin or their combinations. Prior to the drug administration only certain short thin processes of epithelial cells masked by a great number of lymphoid elements are detectable in the cortical zone of the thymus. Injection of 10-40 mg/kg prednisolone, 100-200 mg/kg azathioprin or their combinations causes stable different-degree demasking of the epithelial reticulum seen during immunofluorescence as a network of fluorescent processes. Administration of drug combinations also causes marked attenuation of the reaction, probably resulting from dystrophic changes in thymic epithelial cells.

[Localization of glycoconjugates in the tissue structures of man and animals].

By means of concanavalin A (Con A) labelled with fluorescein isotiocyonate in cryostate sections of some human and animal organs (skin, heart, thymus) localization of clycoconjugates containing hexoses and reacting with this lectin has been studied. Glycoconjugates are revealed along the periphery of all cellular elements, in Z-lines and in intercalated disks of the myocardial muscle fibers, in the connective tissue ground substance and also in that of the basal membranes of epithelium, endothelium, muscle fibers (sarcolemma), in cytoplasm of epithelioreticulocytes of the thymus. In fibroblasts of the L-cells culture, glycoconjugates are also localized in cytoplasm. When N-acetylglucosamin is added to the labelled Con A solution, the ability of lectin to react with tissue components is inhibited.

[Fibronectin in the structures of total preparations of loose connective tissue].

A comparative study of fibronectin localization in the cells and paraplastic structures of total film loose connective tissue preparations obtained from different animal species (mice, rats, guinea-pigs, golden hamsters) made it possible to reveal some regularities. In mice and rats, protein was found in fibroblast cytoplasm (or on its surface) in the form of dots and dotted lines. In golden hamsters, it was found in the form of larger round loci, whereas in guinea-pigs, fibronectin was diffused. In glycokalyx, fibronectin was detectable on the surface of reticular fibres in the form of ununiform accumulations. The greatest amount of protein was found in the cross-zones of reticular fibres, where fibroblasts actively synthesizing fibronectin are located.
