Effect of dizocilpine maleate on cerebral anoxia and ischemic damage in rodents.

The Protective effects of dizocilpine maleate (DM) against anoxia in mice and ischemic damage in rats of 4-vessel occlusion (4-VO) were studied. DM 0.5 or 1.0 mg.kg-1 ip significantly prolonged the survival time of mice in closed containers. DM 0.5 and 1.0 mg.kg-1 ip 30 min prior to 4-VO obviously accelerated the electroencephalographic recovery, reduced the neuronal loss in the hippocampus, and increased the survival rate after 72-h reperfusion. These effects followed a dose-dependent manner. Our results indicate that selective non-competitive N-methyl-D-aspartate receptor blocker DM protects against anoxic and ischemic cerebral damage.

Hysteroscopic hydrotubation for treatment of tubal blockage.

In order to increase the effectiveness of hydrotubation, a plastic cannula was inserted into the fallopian tube under hysteroscopic control. Fifty women with tubal blockage confirmed by HSG were selected for treatment. Hydrotubation solution consisted of hydrocortisone (20 mg), gentamicin (160,000 IU) and procaine (80 mg) in 20 ml distilled water. Repeat HSG was performed. After three treatments, tubal patency (one or both tubes) was achieved in 20 women (40%), and seven pregnancies resulted within 1-18 months. Hysteroscopic hydrotubation may be an alternative treatment for tubal blockage.

Identification and distribution of tropomyosin isoforms in chicken digestive canal.

Two-dimensional gel electrophoresis of eight digestive organ extracts from 60-day-old chicks was performed. Judging from the similarity of their protein maps, the organs were classified into the following four types: 1) esophagus type, 2) proventriculus type, 3) gizzard type, and 4) intestine type. In four representative organs of these types, the distribution of tropomyosin isoforms was examined, and four high- and five low-Mr-type isoforms in addition to alpha and beta isoforms were detected in the embryonic organs. In the adult organs, however, there were three high- and four low-Mr-type isoforms, which were restricted to the mucous membrane, in addition to alpha and beta isoforms. Immunoblot analysis indicated that the high- and low-Mr-type isoforms in the embryo corresponded with those in the adult mucous membrane, but differences in the number and amount of the isoforms were found between the embryo and the adult mucous membrane.

[Separation of epimers of anisodamine (654-2) with HPLC using chiral additives in the mobile phase].

The separation of epimers of anisodamine were studied with HPLC using a reversed phase ODS C18 column. Several chiral additives were tried in the mobile phases. d-Camphorsulfonic acid and L-(+)-dibutyl tartrate showed good resolution ability for the separation of the isomers, a pair of epimers of anisodamine were well separated; while beta-cyclodextrin appeared to have no selectivity for the epimers.

Effect of intrahippocampal quinolinic acid infusion on the amygdala kindling in rat.

The effect of intrahippocampal infusion of quinolinic acid (Quin), an endogenous excitatory amino acid, was studied on the amygdala kindling. Quin 120 nmol injected intrahippocampally 2 wk prior to the beginning of amygdala kindling significantly not only produced dorsal hippocampal pyramidal and granule cell loss but also decreased the number of stimuli to trigger the stage 5 seizures of amygdala kindling. In kindled rats, intrahippocampal 20 nmol Quin infusion fully inhibited the stage 5 of amygdala-kindled seizures. The inhibitory effect of Quin was antagonized by dl-2-amino-7-phosphonoheptanoic acid, a selective antagonist of N-methyl-D-aspartate (NMDA) type receptors. The results suggest that NMDA-type receptors in the hippocampus may play a role in the control of the seizure threshold in the amygdala.

[Total synthesis of an analogue of schizandrin].

A schizandrin analogue 9, 1, 2, 3, 10, 11, 12-hexamethoxy-6,7-dihydroxy-cis-6,7-dimethyl dibenzocyclooctadiene, is one of the metabolites of schizandrin with comparable anti-convulsion activity. In this paper, compound 9 was synthesized from gallic acid through the following sequence of reactions: methylation, Henry condensation, reduction with Fe-FeCl3-HCl, intermolecular reductive coupling and intramolecular nonphenolic oxidative coupling, totally in seven steps. The intermediate 1,4-diaryl-2,3-dimethyl-2,3-butanediol (7) which is a mixture of erythro and threo isomers was converted by DDQ in TFA into compound 9 and another new compound 10 via a different reaction path.

[Chemical identification of structure of podocarpamide by synthesis].

Podocarpamide, isolated from the bark of Zanthoxylum podocarpum Hemsl., is a new compound with platelet aggregation inhibiting and SGPT level lowering activities. Its structure was inferred to have two possible forms, either I or II, from its spectra. The structure of podocarpamide was identified as I by synthesis.

[Studies on the assimilation of inorganic selenium by yeast].

In the process of assimilation of inorganic selenium by yeast to the organic-selenium, some rules on the relation of the kinds of culture medium, concentration of sodium selenite and methionine to the total selenium and selenomethionine content in the yeast formed have been found; and a new, accurate procedure--modified acid hydrolysis-ion exchange chromatography for determining the content of seleno-amino acid in biological materials has been established.

A simplified technique for the short-term tissue culture of rabbit corneal cells.

A technique for the short-term culture of pure populations of rabbit corneal endothelial and epithelial cells has been developed. Rabbit corneas were placed on concave agarose surfaces, treated briefly with a solution of trypsin and ethylenediamine tetracetic acid, and transferred, either epithelial cell surface or endothelial cell surface down, to microscope slide culture chambers. Within 6 to 12 h the epithelial cells or endothelial cells attached to the slide chamber surface and the cornea was removed, leaving behind a pure population of cells which spread out and grew to fill the surface of the slide chamber. This technique provides a simple and economic means for the reproducible initiation of primary cultures of rabbit corneal epithelial and endothelial cells for us in a variety of experiments.

[The effect of vanillin on the fully amygdala-kindled seizures in the rat].

The anticonvulsive action of vanillin, an antiepileptic agent structurally related to vanillyl alcohol isolated from Gastrodia elata Blume, on the fully amygdala-kindled seizures was investigated. Fully kindled seizures were produced by repeated application of low intensity electrical stimulation to the basolateral amygdala once per day. Fifteen consecutive stimulations were needed to reach the first stage 5 seizures at which animals showed behavioral convulsions and electroencephalographic abnormalities. The stage 5 seizures were suppressed by vanillin ip 1 h before stimulation with the ED50 of 286 mg/kg. Meanwhile, the epileptic afterdischarge duration was significantly shortened. Phenytoin at a nontoxic dose 50 mg/kg ip obviously reduced the stage 5 seizures. The present study provides a very useful experimental model of chronic epilepsy to detect anticonvulsants.

[Comparative study of outer envelope protein and LPS of five strains of leptospires by SDS-PAGE and 2D-PAGE].

We applied SDS-PAGE, 2D-PAGE and Western blot to analyse the outer envelopes protein and LPS of five strains of leptospires. The work would lay foundations for taxonomy, the development of vaccination regimens and the elucidation of pathogenic mechanisms. The outer envelope proteins of leptospires were analyzed by SDS-PAGE and silver staining. We found that the protein profiles of the pathogenic leptospires were basically identical. A comparison of the protein profiles of the pathogenic L. with those exhibited by two nonpathogenic L. indicated that there was no obvious relationship between these organisms and any of the L. interrogans strains examined. The quantity of 21.5 kd protein of strain 017 was greater than that of strain 601 and 156. Approximately 200, 225, 238 distinct polypeptides were detected in the strain 017, 601 and 156 in 2D-PAGE by silver staining respectively. The profiles of 2D-PAGe showed obvious differences in pI. The pI of strain 017, 601 and 156 were mainly 6.68-7.4, 6.55-6.9, 5.85-7.1 respectively. The 21.5 kd protein of strain 017 was made up of six polypeptides. Our immunoblots revealed that McAb (LB1) reacted with a 41 kd antigen, which was common to the three virulent leptospires tested. SDS-PAGE profiles of silver stained outer envelope LPS of pathogenic L. differed greatly from those of the nonpathogenic L. There was a distinct differences between strain Patoc I and 3055. Our studies showed that each of the five strains of leptospires possessed characteristic outer envelope LPS, which may be used to identify the genus, species and serovars of a strain of L.
