Preliminary study on the effect of the selected calmodulin antagonists on the skin.

The aim of the study was to make a selection of the calmodulin antagonists, which could be useful in topical treatment of psoriasis. Four drugs were assessed: chlorpromazine, trifluoperazine, miconazole and ketoconazole. They were applied on the skin of guinea pigs, twice a day for the period of two weeks. Biopsy specimens from these sites were being taken for light microscopy, histoenzymatic examination and for evaluation of the proliferation activity of the epidermis (the AgNOR stain, after Ploton). A decrease in reaction activities for lactate dehydrogenase and succinic dehydrogenase as well as in the proliferation activity of the epidermis, could suggest an inhibitory effect of chlorpromazine and miconazole on the cell cycle and keratinization process.

Study of lithogenicity of bile among patients suffering from cholecystolithiasis and among patients not suffering from this disease.

On the basis of biochemical and morphological research, the authors decided to attempt to determine the degree of lithogenicity of the bile among patients suffering from cholecystolithiasis (100 patients) and those not suffering from this disease (31 patients). Having analyzed the obtained data, it can be concluded that the degree of lithogenicity of bile is influenced by the concentration of cholesterol, bile acids and phospholipids as well as the damage of the liver and of the wall of the gallbladder as well as its kinetics. Lithogenic bile was noticed in 84% of the patients of group I and in 54% in group II. However, the degree of lithogenicity was significantly higher in patients suffering from cholecystolithiasis. According to the authors, the degree of lithogenicity is the deciding factor in the creation of bile stones.

Cor pulmonale in experimental lung emphysema. I. The morphometric evaluation of pulmonary and myocardium changes.

Selected morphometric parameters of the lungs and heart of rats with experimental emphysema caused by a proteolytic enzyme--papain have been presented. The morphometric evaluation was carried out after the 1st, 3rd and 6th months following an intratracheal infusion of papain solution. It was found that administration of the papain caused emphysematous changes in the lungs, which intensity measured morphometricly increased until the third month, and then became stable. The first evidence of the right ventricle hypertrophy appeared already in the third month of the experiment. The evident hypertrophy of the muscle of both ventricles and of the interventricular septum was observed after six months of the experiment. Morphologic changes developing in both ventricles suggest an existence of a common pathogenetic factor affecting simultaneously the myocardium of right and left heart ventricles.

[Comparison of healing characteristics of DALLON-standard and DALLON-double velour vascular prostheses].

By means of histological method and the scanning and transmission electron microscope healing processes of two types of Polish prostheses: DALLON-standard and DALLON-double velour were compared. The experiments were carried out on 36 dogs. The two types of prostheses mentioned were implanted in abdominal aorta. The grafts were removed 1, 4 and 12 months after implantation. The performed studies showed that although the sequence and nature of morphological changes leading to the reconstruction of the vascular wall in both types of grafts were similar, the development of the neointima as well as the formation of the neo-adventitia firmly accreted with surrounding tissues were considerably faster in double velour DALLON prostheses. Unlike in standard DALLON prostheses, the neointima of double velour DALLON grafts was rich in cells, had elastic fibres. Already after 12 months double velour DALLON grafts were completely covered by endothelial cells, which proves better reconstruction of the vascular wall. Rapid endothelial cells coverage of the lumen surface prevents thrombosis. Morphological differences observed in endothelial cells covering the neointima of standard grafts and double velour graft suggest the existance of various sources of endothelialization in both types of grafts.

Chondroprotective action of salmon calcitonin in experimental arthropathies.

To assess whether calcitonin exerts an influence on cartilage, three models of arthropathies in rabbits--representing three different modes of cartilage destruction--were used: (1) corticosteroid administration (endocrinological disturbances model); (2) meniscectomy (mechanical stress model); and (3) immobilization of the hind leg (nutritional disorder model). After 12 weeks of methylprednisolone (MP) administration, the rabbit femur heads displayed cartilage erosions, marked decrease of glycosaminoglycans (GAG) content, and narrowing of joint spaces. Elevation of serum uronic acid, activity of alkaline phosphatase, and calmodulin content was evident. All these changes were minimal--close to normal--in the group treated for 12 weeks with MP + salmon calcitonin (sCT). Partial meniscectomy and hind leg immobilization caused statistically significant loss of GAG from the cartilage and narrowing of the knee joint space during the same experimental period, 12 weeks. In both these models the groups of rabbits treated simultaneously with sCT showed only insignificantly smaller joint spaces and GAG content. These results support our hypothesis of a chondroprotective property of calcitonin. However, the mechanism through which calcitonin influences joint cartilage remains unknown. A direct effect of calcitonin on cultivated chondrocytes, as well as the role of calmodulin, beta-endorphins, calcium, and interleukin-1 in the process are discussed.

[Morphological studies of the effects of protein-free diet and branched-chain amino acids (aminosteril-hepa (Frasenius)) on experimental thioacetamide-induced liver damage].

The studies were performed in two groups of rats in which experimental hepatic encephalopathy was induced with tioacetamide++. One group of rats on protein-free diet was given intraperitoneally a mixture of amino-acids (Aminosteril-Hepa), the other one was given proteins in the diet in a dose 1 g per kg of body weight. Histological and histochemical studies revealed that free-protein diet with a mixture of aliphatic polybranched amino-acids (Aminosteril-Hepa) inhibited hepatotoxic thioacetamide+ action on the rat liver.

[Morphological picture of the lateral cruciate ligament of the rabbit knee after its intra-articular transposition].

The aim of the study was to evaluate morphological changes occurring in an experimentally reconstructed anterior cruciate ligament. Experimental studies concerned 20 New Zealand rabbits. We showed the phasic character of morphological changes within the intra-articularly transposed ligament. The phase of transplant revascularization and maturation of collagen fibers were considered the most significant.

[Effect of differentiated diet therapy on the clinical results and biochemical indicators in chronic hepatic encephalopathy].

Effects of differentiated diet with reduced proteins or diet without proteins with simultaneous use of lactulose or the preparation enriched with aliphatic amino acids with aminosteril-hepa chain on the clinical results and the ammonia, phenols, alpha-amino nitrogen, tyrosine, phenylalanine and tryptophan++ concentrations in the serum have been studied. It has been demonstrated that limitations of proteins in the diet or diet without proteins with the use of lactulose or amino acids mixtures above does not influence significantly the clinical amelioration or biochemical indices of encephalopathy or coma during the liver cirrhosis course. Homogenates of the liver and brain of patients who died due to encephalopathy and liver cirrhosis showed high contents of ammonia, tyrosine, phenylalanine and tryptophan++.

[Bilateral parathyroid adenoma].

A patient with multiple parathyroid adenomas is reported. Their size was considerable, and the course of primary hyperparathyroidism was stormy. A rapid regression of symptoms followed surgical treatment. The rarity of multiple parathyroid adenomas of large size is stressed.

[Studies of the effect of a protein-free diet and branched-chain amino acids (preparation Aminosteril-Hepa) on selected biochemical indicators in chronic experimental hepatic encephalopathy].

The latest observations suggest that a decreased level of serum branched-chain amino acids plays a role in the pathogenesis of liver encephalopathy. In this study we analysed the behaviour of the serum concentrations of: ammonia, phenols, alpha-amino nitrogen, tyrosine, phenylalanine and tryptophan in rats with thioacetamide-induced liver encephalopathy. The first group of rats was on protein-free diet and Aminosteril-Hepa (Fresenius FRG)--an amino acids solution with predominance of branched-chain amino acids. The second group of rats received a diet with 1 g/kg b.w. of protein daily. The levels of biochemical indices were analysed 0, 3, 5 and 7 days after liver encephalopathy development. The results of our study indicate a favourable influence of infusion a branched-chain amino acids solution (Aminosteril-Hepa) on the biochemical indices, particularly phenols and alpha-amino nitrogen in experimental liver encephalopathy.

[Ultrastructure of the new endothelium in polyester aortic prosthesis].

The development of a new intima in polyester prostheses Dallon implanted into the abdominal part of dog's aorta was tested in the scanning and transmission electron microscope. It was proved that during the first month of the experiment morphoetic elements of blood appeared on the graft surface and later within 36 months smooth muscle cells, collagen fibres and small amounts of elasticin could be found. The process of endothelialization began in the 4 month and after 12 months the graft was totally covered by endothelium. The picture in the SEM especially in the central part differed from the picture of normal arterial endothelium.

The effect of repeated BCG vaccine administration associated with surgery upon the growth of lung metastases in rats with transplantable Morris 5123 hepatoma.

Eighty four male Buffalo rats bearing transplantable Morris 5123 hepatoma were used for the experiment. Half of the animals were given into the tumor BCG vaccine (final dose 2.2 X 10(6) living mycobacteria), the other 42 rats after BCG underwent surgical removal of the hind leg together with the tumor. Though the immunotherapy reduced the size of lung metastases of the tumor it did not, however diminish the survival rate of the animals. Multiple BCG doses associated with the surgical procedure prevented dissemination of the tumor cells and prolonged the survival of rats up to the 100th day of the follow-up.
