Transmission characteristics of a Fabry-Perot etalon-microtoroid resonator coupled system.

The transmission spectra of a Fabry-Perot etalon coupled to a microtoroid resonator are studied theoretically and experimentally. The resonance line shapes depend strongly on the resonance wavelength detuning and coupling strength between the two resonators. A wide variety of line shapes, ranging from a single to triple peaks, symmetric to asymmetric Fano-like peaks, and notches were predicted and observed experimentally. The capability to modify the spectral line shapes by tuning the coupling between or losses of two resonators may find applications in optical filtering, switching, sensing, and dispersion engineering.

[The effects of 20(R)-Rg3 on lung carcinoma A549 cell line and endogenous VEGF secreted by tumor cells].

OBJECTIVE: To assess the effect of 20(R)-Rg3 on the inhibition of angiogenesis of lung cancer and explore its role in the change of endogenous VEGF secreted by A549 tumor cells themselves. METHODS: A549 tumor cells were cultured with different concentrations of 20(R)-Rg3(10(-7) mol/L, 10(-6) mol/L, 10(-5) mol/L and 10(-4), mol/L), the VEGF contents were detected by ELASA method after 24 h, 48 h and 72 h. Cellular proliferation was detected by MTT method, and apoptosis was detected with flow cytometry. RESULTS: It was found that Rg3 had no significant inhibitory effect on the proliferation of A549 cells. The apoptosis rate was 29.8% after the cells being interfered with Rg3 at 3 x 10(-5) mol/L for 120 h. The levels of VEGF in the groups of Rg3 at 10(-5) mol/L and 10(-4) mol/L were lower than that in the contrast group (P < 0.05). The level of VEGF in the group of 10(-4) mol/L was lower than that in the group of 10(-7) mol/L and group of 10(-6) mol/L, and it was lower after 72 h of interference than that after 24 h and 48 h of interfernce. CONCLUSION: With the action of 20 (R)-Rg3, the level of VEGF secreted by A549 tumor cells themselves declined; the apoptosis of A549 tumor cells increased, especially at higher concentration and longer action time of Rg3. This might be one of the antiangiogenesis mechanisms of 20(R)-Rg3.

[A multicentre study on the pathogenic agents in 665 adult patients with community-acquired pneumonia in cities of China].

OBJECTIVE: To investigate the pathogenic causes of community-acquired pneumonia (CAP) in adult patients in China, the relation of previous antibiotic use and the Pneumonia Patient Outcome Research Team (PORT) classification to microbial etiology, and the prevalence of drug resistance of common CAP bacteria. METHODS: A prospective study was performed on 665 consecutive adult patients with CAP at 12 centers in 7 Chinese cities during one year. The etiology of pneumonia was considered if one of the following criteria was met: (1) valid sputum sample yielding one or more predominant strains; (2) blood cultures yielding a bacterial pathogen; (3) seroconversion, a > or = 4-fold increase or decrease titers of antibodies to Mycoplasma pneumoniae, Chlamydia pneumoniae and Legionella pneumophila. Minimum inhibitory concentration (MIC) of respiratory tract isolates was determined using the agar dilution method. RESULTS: Pathogens were identified in 324/610 patients (53.1%) with valid serum samples and sputum cultures as follows: Mycoplasma pneumoniae (126, 20.7%), Streptococcus pneumoniae (63, 10.3%), Haemophilus influenzae (56, 9.2%), Chlamydia pneumoniae (40, 6.6%), Klebsiella pneumoniae (37, 6.1%), Legionella pneumophila (31, 5.1%), Staphylococcus aureus (23, 3.8%), Escherichia coli (10, 1.6%), Moraxella catarrhalis (8, 1.3%), Pseudomonas aeruginosa (6, 1.0%). Of 195 patients with a bacterial pathogen, an atypical pathogen was identified in 62 (10.2%) cases. The non-susceptibility rate of Streptococcus pneumoniae to penicillin, azithromycin, and moxifloxacin was 20.3%, 75.4% and 4.3% respectively. CONCLUSIONS: Atypical pathogens have important role in CAP, with Mycoplasma pneumoniae being the most common pathogen, and mixed infection of atypical pathogens with bacteria was found in 10.2% of the cases. Streptococcus pneumoniae and Haemophilus influenzae remain the most important bacteria for CAP. More than 75.0% of Streptococcus pneumoniae was resistant to macrolides and 20.3% was resistant to penicillin.

[Studies on the chemical constituents from stem of Chirita longgangensis var. Hongyao].

OBJECTIVE: To isolate and elucidate the constituents from stem of Chirita longgangensis var. hongyao. METHOD: The constituents were extracted with methanol and isolated by chromatography on silica gel, Sephadex LH-20 and ODS. The structures were determined by NMR and MS spectral analysis. RESULT: Seven compounds were identified as 2-hydroxy-7-methyl-9, 10-anthraquinone (1), 2-methyl-9, 10-anthraquinone (tectoquinone) (2), ursolic acid (3), vanillic acid (4), beta-sitosteryl-D-glucoside-6'-palmitate (5), beta-sitosterol (6), daucosterol (7), respectively. CONCLUSION: Compounds 1, 2, 3 and 5 were isolated for the first time from the family Gesneriaceae, compounds 4 and 6 were isolated for the first time from the genus Chirita, and compound 7 was isolated for the frist time from Chirita longgangensis var.

Promoter-specific expression of the imprinted IGF2 gene in cattle (Bos taurus).

The domestic cattle (Bos taurus) has been a good animal model for embryo biotechnologies, such as in vitro fertilization and nuclear transfer. However, animals produced from these technologies often suffer from large-calf syndrome, suggesting fetal growth disregulation. The product of the insulin-like growth factor 2 (IGF2) gene is one of the most important fetal mitogens known to date. A detailed analysis of age-, tissue-, and allele-specific expression of IGF2 has not been performed in the bovine mainly because the majority of the bovine sequence has been unavailable. In the present study, we obtained virtually the entire sequence of the bovine IGF2 cDNA, identified expressed single-nucleotide polymorphisms (SNPs) in both exons 3 and 10, and determined the age-, tissue-, and promoter-specific expression of bovine IGF2 in fetal, calf, and adult tissues. We found that, similar to the human and mouse, bovine IGF2 is subjected to extensive transcriptional regulation through multiple promoters, alternative splicing and polyadenylation, as well as genetic imprinting. However, major differences were found in the regulation of the bovine IGF2 in nearly all aspects of age-, tissue-, promoter-, and allele-specific expression of IGF2, and the promoter-specific loss of imprinting from every other species studied, including cattle's close relatives, the sheep and the pig. The data presented here are of important reference value to cattle produced from embryo biotechnologies.

Temporal behavior of radiation-pressure-induced vibrations of an optical microcavity phonon mode.

We analyze experimentally and theoretically mechanical oscillation within an optical cavity stimulated by the pressure of circulating optical radiation. The resulting radio frequency cavity vibrations (phonon mode) cause modulation of the incident, continuous-wave (cw) input pump beam. Furthermore, with increasing cw pump power, an evolution from sinusoidal modulation to random oscillations is observed in the pump power coupled from the resonator. The temporal evolution with pump power is studied, and agreement was found with theory. In addition to applications in quantum optomechanics, the present work suggests that radiation-pressure-induced effects can establish a practical limit for the miniaturization of optical silica microcavities.

Expression of imprinted genes is aberrant in deceased newborn cloned calves and relatively normal in surviving adult clones.

Cattle are the species used most frequently for the development of assisted reproductive technologies, such as nuclear transfer. Cattle cloning can be performed by a large number of laboratories around the world, and the efficiency of nuclear transfer in cattle is the highest among all species in which successful cloning has been achieved. However, an understanding of the expression of imprinted genes in this important species is lacking. In the present study, real time reverse transcription polymerase chain reaction (RT-PCR) was utilized to quantify the expression of the bovine Igf2, Igf2r, and H19 genes in eight major organs (brain, bladder, heart, kidney, liver, lung, spleen, and thymus) of somatic cell cloned calves that died shortly after birth, in three tissues (skin, muscle, and liver) of healthy clones that survived to adulthood, and in corresponding tissues of control animals from natural reproduction. We found that, deceased bovine cloned calves exhibited abnormal expression of all three genes studied in various organs. Large variations in the expression levels of imprinted genes were also seen among these clones, which were produced from the same genetic donor. In surviving adult clones, however, the expression of these imprinted genes was largely normal, except for the expression of the Igf2 gene in muscle, which was highly variable. Our data showed disruptions of expression of imprinted genes in bovine clones, which is possibly due to incomplete reprogramming of donor cell nuclei during nuclear transfer, and these abnormalities may be associated with the high neonatal mortality in cloned animals; clones that survived to adulthood, however, are not only physically healthy but also relatively normal at the molecular level of those three imprinted genes.

Conceptual framework of knowledge management for ethical decision-making support in neonatal intensive care.

This research is built on the belief that artificial intelligence estimations need to be integrated into clinical social context to create value for health-care decisions. In sophisticated neonatal intensive care units (NICUs), decisions to continue or discontinue aggressive treatment are an integral part of clinical practice. High-quality evidence supports clinical decision-making, and a decision-aid tool based on specific outcome information for individual NICU patients will provide significant support for parents and caregivers in making difficult "ethical" treatment decisions. In our approach, information on a newborn patient's likely outcomes is integrated with the physician's interpretation and parents' perspectives into codified knowledge. Context-sensitive content adaptation delivers personalized and customized information to a variety of users, from physicians to parents. The system provides structuralized knowledge translation and exchange between all participants in the decision, facilitating collaborative decision-making that involves parents at every stage on whether to initiate, continue, limit, or terminate intensive care for their infant.

[The effect of 20(R)-ginsenoside Rg3 on the differential expression of cell signaling genes and other related genes in human lung adenocarcinoma cell line A549].

OBJECTIVE: To study the effect of 20(R)-ginsenoside Rg3 [20(R)-Rg3] on the differential expression of cell signaling genes and other related genes in human lung adenocarcinoma cell line A549. METHODS: The cell line A549 was cultured with 20(R)-Rg3 (10(-6) mol/L) for 72 h, RNA was extracted, and the differential expression of cell signaling genes and other related genes were detected with DNA microarray. RESULTS: A total of 3 490 genes were detected, and the expression of 24 genes were changed after the addition of 20(R)-Rg3. Two cell signaling genes & transducin genes were up-regulated. Two cystoskeleton & locomotion genes were up-regulated. Three proto-oncogene & anti-oncogene genes were down-regulated. Two of the translation & synthetical genes were down-regulated, and 1 of them was up-regulated. One DNA recombination gene and one metabolism gene were down-regulated. CONCLUSION: 20(R)-Rg3 showed significant effect on the differential expression of cell signaling genes and other related genes in human lung cell line A549.

[Clinical significance of detection of thrombus precursor protein in severe sepsis].

OBJECTIVE: To evaluate the significance of the changes in plasma thrombus precursor protein (TPP) in severe sepsis. METHODS: Enzyme linked immunoadsorbent assay (ELISA) was used in the determination of plasma TPP in 22 patients with severe sepsis group. Prothrombin time (PT), activated partial thromboplastin time(APTT), fibrin(Fib), D-Dimer were also determined and the values were compared with those obtained from 10 patients with infection and 8 healthy normal controls. At the same time, scores of sepsis related organ failure assessment(SOFA), simplified acute physiology score (SAPSII), Marshall criteria were made respectively in patients with severe sepsis on 1,3,5 days after admission to the ICU. Analysis of correlation between TPP and scores was done. RESULTS: (1)The concentration of TPP and positive rate of D-Dimer in severe sepsis were obviously higher than that in the ordinary infection group and normal control group (all P<0.05). But there were no differences in levels of PT, APTT, and Fib among three groups. (2)The concentration of TPP rose continuously in nonsurvivors due to severe sepsis, and it was positively correlated with scores of SOFA, SAPSII, Marshall criteria. CONCLUSION: TPP levels showed a higher specificity and sensitivity in detecting hypercoagulability state in severe than D-Dimer, PT, APTT, Fib assay. It can be used as a diagnostic and prognostic parameter for early hypercoagulability states and outcome of severe sepsis.

[Chemical constituents of the rhizome of Matteuccia struthiopteris].

AIM: To study the chemical constituents of the rhizome of Matteuccia struthiopteris (L.) The constituents were separated and purified by column chromatography with normal Todaro. METHODS: phase silica gel and Sephadex LH-20. Their structures were identified on the basis of physical and spectral data (MS, NMR, HMBC and HMQC). RESULTS: Four compounds were isolated and identified as 1-O-beta-D-gl ucopyranosyl-(2S,3R,4E, 8Z) -2-N-(2'-hydroxydocosanoyl) eicosasphinga-4,8-dienine (1), 1-O-beta-D-galactosyl-(6-->1)-alpha-D-galactosyl-2,3-O-dihexadecanoyl-glycerol (2), succinic acid (3), D-mannitol (4). CONCLUSION: Compounds 1 and 2 are new compounds. Compounds 3 and 4 were isolated from this plant for the first time.

RESULT: Five phenylethanoid glycosides were identified as 3, 4-dihydroxyphenyl alcohol-beta-D-glucopyranoside (1), 3, 4-dihydroxyphenyl alcohol-6-O-caffeoyl--D-glucopyranoside (calceolarioside B) (2), 3, 4-dihydroxyphenyl alcohol-beta-D-glucopyranosyl-(1 --> 3)-6-O-caffeoyl-beta-D-glucopyranoside (plantainoside D) (3), 3, 4-dihydroxyphenyl alcohol-beta-D-glucopyranosyl-(1 --> 3)-4-O-caffeoyl-beta-D-glucopyranoside (plantamajoside) (4), 3, 4-dihydroxyphenyl alcohol-beta-D-glucopyranosyl-(1 --> 3)-6-O-feruloyl-beta-D-glucopyranoside (scroside E) (5), respectively. CONCLUSION: Compounds 3 and 5 were isolated for the first time from the family Gesneriaceae, and compounds 1, 2 and 4 were isolated for the first time from the genus Chirita.

Genomic imprinting of H19 in naturally reproduced and cloned cattle.

Animals produced from assisted reproductive technologies suffer from developmental abnormalities and early fetal death at a higher frequency than that observed in those produced by natural breeding. These symptoms are reminiscent of imprinting disruptions in the human and mouse, suggesting the possibility of perturbations in the expression of imprinted genes such as biallelic expression or silencing. H19 is one of the imprinted genes first identified in mice and humans, but its sequence and imprinting status have not been determined in cattle. In the present study, we obtained the majority of the bovine H19 gene sequence (approximately 2311 base pairs), identified a single nucleotide polymorphism (SNP) in exon 5 and determined the frequencies of different alleles containing the SNP. Our analysis demonstrated that, in cattle produced by natural breeding, H19 was indeed imprinted as shown by either predominant or exclusive expression of the maternal allele. We also analyzed the imprinting pattern of H19 in organs of four animals produced by somatic cell nuclear transfer that died shortly after birth or had developed abnormalities that necessitated immediate killing at birth. Three out of four cloned animals showed biallelic expression of H19, supporting our hypothesis that imprinting disruption is present in cloned animals that suffered from developmental abnormalities at birth. Examination of the expression of H19 in the offspring of a cloned animal produced by artificial insemination showed that the imprinting pattern in this animal was indistinguishable from those of control animals, suggesting that either imprinting disruptions in cloned animals are corrected through natural reproduction or that they are not present in healthy cloned animals capable of undergoing natural reproduction.

Distinct LIM domains of Hic-5/ARA55 are required for nuclear matrix targeting and glucocorticoid receptor binding and coactivation.

Hydrogen peroxide-inducible clone-5 (Hic-5), belongs to the group III LIM domain protein family and contains four carboxyl-terminal LIM domains (LIM1-LIM4). In addition to its role in focal adhesion signaling, Hic-5 acts in the nucleus as a coactivator for some steroid hormone receptors such as the glucocorticoid receptor (GR) and androgen receptor (AR). Based upon its effect on AR transactivation, Hic-5 has also been designated as ARA55. Here, we report mapping studies of Hic-5/ARA55 functional domains and establish that LIM3 and LIM4 are necessary for maximal effects on GR transactivation. However, results from yeast two-hybrid assays demonstrated that these two LIM domains together, while necessary, are not sufficient to interact with the tau2 transactivation domain of GR. LIM4 also functions as a nuclear matrix targeting sequence (NMTS) for Hic-5/ARA55, as it is both necessary and sufficient to target a heterologous protein to the nuclear matrix. Thus, as suggested from previous analysis of LIM domain-containing proteins, separate but highly related LIM domains serve distinct functions.

Replica-molded high-Q polymer microresonators.

Ultrahigh-Q microtoroids on a chip are applied as replication masters to demonstrate replica-molded high-Q microresonator arrays. Replica Q factors are nearly material loss limited, affirming the integrity of the replication process, and are as high as 5 x 10(6), or nearly a factor of 40 greater than previous polymer-based devices. Because the molding process is nondestructive, both the master and the molds can be reused. Additionally, by using a novel optical polymer (Vicast), we demonstrate storage of high-Q microresonators in the mold for weeks, providing a method to preserve the whispering-gallery Q factor.

Determination of ortho-phenylphenol residues in lemon rind by high-performance liquid chromatography with electrochemical detection using a microbore column.

A simple and highly sensitive method has been developed for determining ortho-phenylphenol (OPP) in lemon rind by high-performance liquid chromatography with electrochemical detection using a microbore column (microHPLC-ECD). Based on the voltammetric behavior of OPP, microHPLC-ECD was established using a CAPCELL PAK C-18 UG 120 microbore ODS column, 17 mM acetic acid-sodium acetate buffer (pH 4.0)/acetonitrile (60/40, v/v) as a mobile phase and an applied potential at +0.9 V vs. Ag/AgCl. The current peak height was found to be linearly related to the amount of OPP injected from 3.4 pg to 1.7 ng (r > 0.999). The detection limit (S/N = 3) was 3.4 pg (20 fmol), which was 100 times greater in terms of sensitivity when compared to conventional HPLC with UV detection. Standard OPP at 0.425 ng was detected with a relative standard deviation (RSD) of 1.9% (n = 10). The OPP contents in several lemon samples were determined by the present method. The recoveries of OPP from lemon rind exceeded 98% with an RSD (n = 5) of less than 3.01%.

[Studies on chemical constituents in rhizome of Matteuccia struthiopteris].

OBJECTIVE: To study the chemical constituents in rhizome of Matteuccia struthiopteris. METHOD: The compounds were isolated by normal phase silica gel chromatography. The structures were identified by physical and spectral data. RESULT: Six compounds were isolated and identified as woodwardic acid (1), ergost-6,22-diene-3beta,5alpha,8alpha-triol (2), apigenin (3), riboflavin (4), 4-O-beta-D-glucopyranosyl-p-coumaric acid (5), 4-O-beta-D-glucopyranosylcaffeic acid (6). CONCLUSION: All the compounds were obtained from this plant for the first time.
