Induction of the Nrf2-driven antioxidant response confers neuroprotection during mitochondrial stress in vivo.

NF-E2 related factor (Nrf2) controls a pleiotropic cellular defense, where multiple antioxidant/detoxification pathways are up-regulated in unison. Although small molecule inducers of Nrf2 activity have been reported to protect neurons in vitro, whether similar pathways can be accessed in vivo is not known. We have investigated whether in vivo toxicity of the mitochondrial complex II inhibitor 3-nitropropionic acid (3-NP) can be attenuated by constitutive and inducible Nrf2 activity. The absence of Nrf2 function in Nrf2(-/-) mice resulted in 3-NP hypersensitivity that became apparent with time and increasing dose, causing motor deficits and striatal lesions on a more rapid time scale than identically treated Nrf2(+/+) and Nrf2(+/-) controls. Striatal succinate dehydrogenase activity, the target of 3-NP, was inhibited to the same extent in all genotypes by a single acute dose of 3-NP, suggesting that brain concentrations of 3-NP were similar. Dietary supplementation with the Nrf2 inducer tert-butylhydroquinone attenuated 3-NP toxicity in Nrf2(+/-) mice, but not Nrf2(-/-), confirming the Nrf2-specific action of the inducer in vivo. Increased Nrf2 activity alone was sufficient to protect animals from 3-NP toxicity because intrastriatal adenovirus-mediated Nrf2 overexpression significantly reduced lesion size compared with green fluorescent protein overexpressing controls. In cultured astrocytes, 3-NP was found to increase Nrf2 activity leading to antioxidant response element-dependent gene expression providing a potential mechanism for the increased sensitivity of Nrf2(-/-) animals to 3-NP toxicity in vivo. We conclude that Nrf2 may underlie a feedback system limiting oxidative load during chronic metabolic stress.

Autosomal dominant neurohypophyseal diabetes insipidus with linkage to chromosome 20p13 but without mutations in the AVP-NPII gene.

CONTEXT: Autosomal dominant neurohypophyseal diabetes insipidus (ADNDI) has been known as a rare disorder transmitted as an autosomal dominant trait, characterized by polyuria and polydipsia, and caused by deficient neurosecretion of arginine vasopressin precursor (AVP-NPII). We reported an ADNDI family with linkage to chromosome 20p13 but without mutations in the AVP-NPII gene. OBJECTIVE: The objective of this study was to identify the corresponding locus responsible for ADNDI in a family without AVP-NP II gene mutations. SUBJECTS AND METHODS: Two families with ADNDI were diagnosed by water deprivation test. The AVP-NPII gene was amplified by PCR and sequenced. A genomewide scan was performed in one family using 400 microsatellite markers covering 22 autosomes. RESULTS: A 3-bp deletion (1827-1829delAGG) of AVP-NPII gene was identified in the affected individuals in one family. Although no mutations could be detected in the coding, the promoter, and intronic regions of AVP-NPII gene in the other family, a maximum LOD score of 1.202999 (theta = 0.00) was obtained at marker D20S889 by genomewide scan, and a 7-cM interval on chromosome 20p13 was defined by fine mapping with markers D20S199-D20S849. Furthermore, the intragenic region that regulates AVP-NPII and oxytocin expression as an enhancer element and the UBCE7IP5 gene that participates in prohormone degradation were sequenced. No alterations could be detected either. CONCLUSION: The corresponding locus responsible for ADNDI is possibly heterogeneous regarding the slightly different clinical features in these two families.

Fabrication of superhydrophobic surfaces by self-assembly and their water-adhesion properties.

The self-assembled films of methyloctyldimethoxysilane (MODMS) and fluorooctylmethyldimethoxysilane (FODMS) were prepared on silicon surfaces and evaluated with AFM, water contact angle measurement, and X-ray photoelectron spectroscopy. Superhydrophobic surfaces were obtained by cooperation of MODMS and FODMS self-assembly with surface roughening. The results showed that preparing closely packed self-assembled films and fabricating surface nanometer-scale and micrometer-scale binary roughness can achieve superhydrophobic films with a water contact angle larger than 156 degrees. The difference between solution deposition and chemical vapor deposition is also investigated. Moreover, superhydrophobic surfaces created with MODMS and FODMS show the different water-adhesion effects, which could have great significance on liquid microtransport in microfluid devices.

Field emission properties of large-area nanowires of organic charge-transfer complexes.

In this contribution, large-area organic charge-transfer complex (AgTCNQ and CuTCNQ) nanowires were synthesized by organic vapor-solid-phase reaction at mild experimental conditions. These nanowires were facilitated on the surface of Cu and Ag foils or different kinds of substrates coated with a layer of silver and copper on a large scale. The excellent field emission properties were observed in the as-grown AgTCNQ and CuTCNQ nanowires. They should have great potential in vacuum device applications.

Bioinspired surfaces with special wettability.

Biomimetic research indicates that many phenomena regarding wettability in nature, such as the self-cleaning effect on a lotus leaf and cicada wing, the anisotropic dewetting behavior on a rice leaf, and striking superhydrophobic force provided by a water strider's leg, are all related to the unique micro- and nanostructures on the surfaces. It gives us much inspiration to realize special wettability on functional surfaces through the cooperation between the chemical composition and the surface micro- and nanostructures, which may bring great advantages in a wide variety of applications in daily life, industry, and agriculture. This Account reviews recent progress in these aspects.

Induction of mucosal and systemic immune response by single-dose oral immunization with biodegradable microparticles containing DNA encoding HBsAg.

The purpose of this work was to assess the ability of plasmid DNA encoding hepatitis B virus (HBV) HBsAg encapsulated in poly(DL-lactide-co-glycolic acid) (PLGA) microparticles to induce local and systemic HBsAg-specific immunity following a single dose of oral immunization. RT-PCR analysis demonstrated prolonged transcription of plasmid DNA, consistent with the sustained expression and presentation of target antigen observed by confocal laser scanning microscopy, in gut-associated lymphocyte tissue (GALT) from mice immunized orally with plasmid DNA encapsulated into PLGA microparticles. Oral administration of PLGA-DNA microparticles induced a long-lasting and stable antigen-specific antibody response, both serum total antibody and intestinal IgA, in BALB/c mice. Mice immunized orally exhibited antigen-specific gamma interferon production and cytotoxic T lymphocyte responses in spleen and GALT after restimulation in vitro with HBsAg or tumour cells stably expressing HBsAg. In contrast, naked DNA vaccines given by intramuscular injection induced only systemic cellular and humoral responses to HBsAg, which were much lower than the responses elicited by oral DNA encapsulated in PLGA microparticles at equivalent doses. The results are encouraging with regard to obtaining good compliance and vaccination coverage with candidate plasmid DNA vaccines, especially in developing countries.

Effects of CpG-ODN on gene expression in formation of foam cells.

AIM: To investigate the effects of CpG-oligodeoxynucleotide (CpG-ODN) on the formation of macrophage foam cells and related gene expression. METHODS: A gene expression profile was examined by microarray techniques, and mRNA expression was detected by reverse transcriptase polymerase chain reaction (RT-PCR). The cholesterol and cholesteryl ester contents of cells were determined by high performance liquid chromatography. RESULTS: CD36, LPL, and Fcgamma2b, which were related to lipid metabolism and the formation of macrophage foam cells, were upregulated after CpG-ODN stimulation. The mRNA expression related to the formation of foam cells was confirmed by semiquantitative RT-PCR. Moreover, histochemical analysis confirmed that lipid deposits inside cells increased after CpG-ODN treatment. However, using flow cytometry, we found that CpG-ODN had no effect on the expression of membrane receptors. CONCLUSION: CpG-ODN up-regulated the expression of genes in macrophage foam cell formation.

[Effect of nucleolin down-regulation on the proliferation and apoptosis in C2C12 cells].

OBJECTIVE: To clarify the effect of nucleolin on the proliferation and apoptosis in C2C12 cells. METHODS: After inhibiting the expression of nucleolin using antisense oligonucleotides, the cellular proliferation was determined by MTT, and the apoptosis was detected by flow cytometry (FCM) assays and DNA ladder assays. RESULTS: After being transfected with antisense oligonucleotides for 24 hours, Western blotting showed that the expression of nucleolin was repressed significantly. In cells treated with antisense oligonucleotides, the cellular proliferation was obviously inhibited; the apoptotic cell increased significantly; and the "DNA ladder" was clearly observed. But the sense and random oligonucleotides had no effect on the cellular proliferation and apoptosis. CONCLUSION: The down-regulation of nucleolin can inhibit the cellular proliferation and initiate the apoptosis in C2C12cells.

Biophysics: water-repellent legs of water striders.

Water striders (Gerris remigis) have remarkable non-wetting legs that enable them to stand effortlessly and move quickly on water, a feature believed to be due to a surface-tension effect caused by secreted wax. We show here, however, that it is the special hierarchical structure of the legs, which are covered by large numbers of oriented tiny hairs (microsetae) with fine nanogrooves, that is more important in inducing this water resistance.

Polymer-controlled synthesis of Fe3O4 single-crystal nanorods.

In this study, we describe a simple approach to preparing single-crystal Fe3O4 nanorods in the presence of poly(vinylpyrrolidone) (PVP). The morphologies of the nanorods are characterized by transmission electron microscopy (TEM), which indicates that these nanorods are formed by nucleation and growth process in situ in aqueous solution. A superconducting quantum interference device (SQUID) magnetometer characteristic of the as-synthesized Fe3O4 nanorods shows superparamagnetic properties.
