Whole-body insulin resistance in the absence of obesity in FVB mice with overexpression of Dgat1 in adipose tissue.

Insulin resistance is often associated with obesity. We tested whether augmentation of triglyceride synthesis in adipose tissue by transgenic overexpression of the diacylglycerol aclytransferase-1 (Dgat1) gene causes obesity and/or alters insulin sensitivity. Male FVB mice expressing the aP2-Dgat1 had threefold more Dgat1 mRNA and twofold greater DGAT activity levels in adipose tissue. After 30 weeks of age, these mice had hyperglycemia, hyperinsulinemia, and glucose intolerance on a high-fat diet but were not more obese than wild-type littermates. Compared with control littermates, Dgat1 transgenic mice were both insulin and leptin resistant and had markedly elevated plasma free fatty acid levels. Adipocytes from Dgat1 transgenic mice displayed increased basal and isoproterenol-stimulated lipolysis rates and decreased gene expression for fatty acid uptake. Muscle triglyceride content was unaffected, but liver mass and triglyceride content were increased by 20 and 300%, respectively. Hepatic insulin signaling was suppressed, as evidenced by decreased phosphorylation of insulin receptor-beta (Tyr(1,131)/Tyr(1,146)) and protein kinase B (Ser473). Gene expression data suggest that the gluconeogenic enzymes, glucose-6-phosphatase and phosphoenolpyruvate carboxykinase, were upregulated. Thus, adipose overexpression of Dgat1 gene in FVB mice leads to diet-inducible insulin resistance, which is secondary to redistribution of fat from adipose tissue to the liver in the absence of obesity.

[Effects of Danshen Decoction on experimental gastric ulcer in rats and mice].

OBJECTIVE: To investigate the preventive and therapeutic effects of Danshen Decoction on gastric ulcer in experimental animal models. METHODS: The model of water immersion restraint stress ulcer in mice, and the models of acetic acid impaired gastric ulcer and pyloric ligation gastric ulcer in rats were established. The total gastric acid, the activity of pepsin and the amount of gastric wall binding mucus were detected. RESULTS: Danshen Decoction reduced the acute gastric mucosal lesion. The effect of 5.0 or 10.0 g/kg group was obviously better than that of the normal saline control group (P<0.05 and P<0.01 respectively). Danshen Decoction accelerated the healing of ulcer induced by acetic acid significantly, and increased the content of gastric wall binding mucus (P<0.01); and it also inhibited the formation of gastric ulcer induced by pyloric ligation, while having no influence on the secretion of gastric acid and the activity of pepsin. CONCLUSION: Danshen Decoction has a significant anti-ulcerative effect, which may be related to improving the gastric mucosal defensive function.

[Junctional epithelialization of dental implants in animals].

OBJECTIVE: To detect the histological changes and the expression of proliferating cell nuclear antigen (PCNA) in junctional epithelialization of dental implants. METHODS: The junctional epithelium was detected by HE staining and the PCNA expression was observed by immunohistochemical staining on the st, 3rd, 6th, 9th, 12th, 15th, 20th and 30th day after implantation in Beagle dogs. RESULTS: The junctional epithelium began to form and PCNA was expressed positively 12 days after implantation. The epithelial hyperplasia tended to stabilization and the expression of PCNA weakened drastically 15 days after implantation. The epithelial attachment showed to be similar to that of natural teeth 20 days later. CONCLUSION: The gingiva epithelium can attach to titanium implants forming junctional epithelium which is similar to the natural teeth.

Attitudes and predictive factors for live kidney donation in British Columbia. A comparison of recipients and wait-list patients.

INTRODUCTION: Live donor kidney transplantation (LDKT) is both medically and economically superior to cadaver kidney transplantation in the treatment of patients with chronic renal failure. Unfortunately, fewer than 50% of patients on the transplant waiting list have a relative or friend who contacts the transplant program about possible donation. We hypothesized that both the potential recipient and potential donor have identifiable and modifiable characteristics that contribute to the likelihood of a live donor transplant. MATERIALS AND METHODS: Specifically-designed and validated questionnaires addressing personal characteristics, knowledge and beliefs about LDKT were mailed to patients who had previously received a LDKT (N = 163) and patients on the cadaver transplant waiting list (N = 251). Response rates were 81% and 67% respectively. RESULTS: There were significant differences between groups in age, ethnicity, marital status, hours worked per week, annual income, and time on the waiting list. Significant differences were found between groups in both knowledge and beliefs about live donor kidney transplantation. All wait-list patients could identify at least one family member (mean = 7 potential donors per wait-list patient) who might serve as a live kidney donor but less than 13% of these potential donors have actually undergone an evaluation. CONCLUSIONS: In British Columbia, an enormous pool of potential live kidney donors exists for patients who are currently waiting for a cadaver kidney transplant. Educational strategies designed for wait-list patients may correct knowledge deficits and alter unfavorable beliefs about LDKT which, in turn, may increase their willingness to seek and accept an offer of live kidney donation.

[Advances on study of organophosphate poisoning prevented by Huperzine A].

Currently, it is a concerned project in medicine to study the effective methods for treatment of organophosphate (OP) poisoning. Huperzine A is a potent, reversible red cells acetylcholinesterases (AChE) inhibitor, which shows high specificity for AChE preserving scavenger capacity of plasma butyrylcholinesterase (BuChE) for OP agents, and cross the blood-brain barrier smoothly preventing the AChE in CNS, so the CNS damages induced by the acute OP poisoning were prevented. These review summarizes the results of experimental in animals, prevention mechanism, and prevention value of HupA against OP poison.

[Effect of shenfu injection on brain natriuretic polypeptide and aminoterminal peptide of precollagen type III in patients with acute myocardial infarction during intervention treatment].

OBJECTIVE: To investigate the effect of Shenfu injection (SFI) on indexes of heart function and myocardial fibrosis in patients with acute myocardial infarction (AMI) treated by percutaneous coronary intervention (PCI). METHODS: Ninety-three AMI patients treated by PCI were randomly divided into two groups, 47 in the treatment group (treated by PCI plus 40 ml SFI intravenous injection, once a day for 7 days) and 46 in the control group (treated by PCI only). Levels of brain natriuretic poly peptide (BNP) and aminoterminal peptide of precollagen type III (NP III) were measured at different time points, i.e. immediately after admission (T1), and at 24th hour (T2) and 7th day (T3) after AMI onset in the two groups. RESULTS: The proportion of TIMI 3 grade of front blood flow in AMI related vessels after PCI was insignificantly different in the two groups. The concentration of BNP of T2 was significantly higher than that of T1 in the two groups (P < 0.01), and it was higher in the control group than that in the treatment group (P < 0.01), while at T3, it was insignificantly different in the treatment group to that of T1 (P > 0.05), but in the control group, it was still significantly higher than that in the treatment group and that of T1 (P < 0.01). The levels of NP III the two groups were similar at T1 and T2 (P > 0.05). At T3, it showed an increase in the treatment group, but with no significant difference (P > 0.05) as compared with that at T1, but in the control group did markedly increase and showed significant difference as compared with that of T1 and that in the treatment group at T3 (P < 0.05). CONCLUSION: Early applying of SFI can protect myocardium from ischemia/reperfusion injury after AMI, ameliorate the degree of injury, improve heart function of patients and prevent myocardial fibrosis.

[Influence of wen-yang herbs on hemodynamics in liver fibrotic rats].

OBJECTIVES: To observe the influence of wen-yang herbs on the hemodynamics in liver fibrotic rats. METHODS: Wistar rats with liver fibrosis, induced by carbon tetrachloride and alcohol, were randomly divided into a treatment group and a control group. The treatment group was administered wen-yang herbs and the control group saline. At the end of the experiment, the hemodynamic markers of the liver and the mesentery, the liver function and hydroxyproline content of liver tissues between the two groups were compared. Blood volume of the livers and hydroxyproline content of liver tissues were also determined. RESULTS: Blood volume of the liver and mesentery (P < 0.01) and blood flow velocity of small vein of mesentery (P < 0.05) of the treatment group were distinctly higher than the control group. The hydroxyproline content (P < 0.01) of the treatment group was remarkably reduced and liver function was improved. CONCLUSION: Wen-yang herbs can activate microcirculation of the liver and mesentery, decrease the deposit of collagen in the liver and improve liver function.

[Studies on formation of aggregates from denatured lysozymes upon renaturing with size exclusion chromatography].

In order to study the renaturation mechanism of denatured protein in denaturant solution, the renaturation and separation process for three kinds of lysozyme molecules, which were separately denatured by urea and guanidine hydrochloride in the presence of reducing agents, was studied by size exclusion chromatography. When initial lysozyme concentration in denaturant solution was more than 10 g/L, the denatured lysozyme molecules were renatured and isolated in a size exclusion chromatographic column. A refolded lysozyme intermediate, a bi-molecular aggregate, was found. This result was confirmed by non-reducing sodium dodecyl sulfate-polyacrylamide gel electrohoresis (SDS-PAGE) analysis of renatured lysozyme molecules with the dilution method. Compared with the dilution method, the amount of the bi-molecular aggregate found by size exclusion chromatography was far less than that found in the dilution method. This result shows that the process for the renaturing of denatured lysozyme molecules in solution can be well described with three-state model in the presence of reducing agents.

[Immune response enhanced by genes encoding IFN-alpha 8 and T alpha 1 co-inoculated with HBV DNA vaccine].

OBJECTIVES: To evaluate if the humoral immune response of hepatitis B DNA vaccine pVAX1-S2S could be enhanced by Talpha1 and/or IFNa expression plasmid co-inoculated. METHODS: The following mammalian expression recombinant plasmids were constructed: the plasmid pVAX1-S2S expressing hepatitis B surface antigen S2S, the plasmid pVAX1-T/I co-expressing thymosin a and IFNalpha, the plasmid pVAX1-I/S2S co-expressing IFNalpha and S2S. These plasmids were inoculated intramuscularly into several BALB/c mice groups in different combinations. In the co-immunization group 1 (pVAX1-I/S2S), each mouse was inoculated with 100 microg of pVAX1-I/S2S; in the co-immunization group 2 (pVAX1-S2S) each mouse was co-inoculated with pVAX1-S2S and 50 microg of pVAX1-TI; in the control group each mouse was inoculated with 100 microg of pVAX1-S2S. All the immunizations were boosted at 2 and 4 week intervals; then the serum samples were collected to detect the anti-HBs and anti-preS2 strengths. RESULTS: 3, 5 and 8 weeks after the first inoculation, the positive rates of anti-HBs were 12.5%, 12.5%, 62.5% respectively in the co-immunization group 1 and 25%, 50%, 50% in the co-immunization group 2, while those in the control group were 0, 25%, 37.5%. The titers of anti-preS2 in co-immunization group 2 was 5 times higher than those in the other two groups. CONCLUSION: The data shows that Talpha1 and/or IFNalpha expression plasmid co-inoculated with pVAX1-S2S might act as an adjuvant to enhance the humoral immune response induced by pVAX1-S2S.

[TNF-alpha mRNA expression in lung cancer cell lines induced by ionizing radiation].

OBJECTIVE: The aim of this study is to investigate the release of TNF-alpha mRNA in two lung cancer cell lines in vitro and the regulation of TNF-alpha mRNA expression by ionizing radiation. METHODS: Two lung cancer cell lines (A549 and NCI-H596) were investigated for their TNF-alpha mRNA expression before and after exposure to different irradiation doses (2, 5, 10, 20, 30 and 40 Gy) and at different time intervals (1, 3, 6, 12, 24, 48 and 72 hours after irradiation). The TNF-alpha mRNA expression was quantified by fluorescence-based real-time quantitative RT-PCR. Colony formation assays were performed after irradiation with a dose of 2, 4, 6, and 8 Gy to determine the clonogenic survival. RESULTS: Dependent on the dose given, irradiation was found to cause increasing induction of TNF-alpha mRNA expression of NCI-H596 cells, reaching maximal level after 40 Gy irradiation, which was 83 times higher than that of normal controls. On the other hand, dependent on the time after irradiation, TNF-alpha mRNA expression of NCI-H596 and A549 cells was increased, reaching maximal level at 6h for NCI-H596 cells, which was 568 times higher than that of normal control cells. TNF-alpha mRNA expression of A549 cells was increased to maximum at 1 h after irradiation and was 136 times higher than that of control cells. Colony formation efficiency (number of colonies divided by the number of inoculated cells) of unirradiated control A549 and NCI-H596 cells was 0.37-0.45 and 0.12-0.24, respectively. The survival fraction (SF) of A549 cells was 47.3% +/- 9.0% at 2 Gy, 18.0% +/- 3.0% at 4 Gy, 6.0% +/- 2.0% at 6 Gy, 1.4% +/- 0.3% at 8 Gy. The SF of NCI-H596 cells was 55.2% +/- 51.0% at 2 Gy, 15.9% +/- 9.2% at 4 Gy, 3.5% +/- 1.7% at 6 Gy; 0.9% +/- 0.6% at 8 Gy. The curves of TNF-alpha expression of the two tumor cell lines were nearly identical, therefore the radiosensitivity of these cell lines was similar. Statistically there was no significant difference for D(0) and D(q) (P > 0.05). CONCLUSION: The two lung cancer cell lines studied express TNF-alpha following irradiation in a time- and irradiation dose-dependent manner. Radiation-induced TNF-alpha production of tumor cells may be of paramount importance not only for tumor behaviour, but also in respect to potential damage to normal tissues and the clinical status of the host.

[Temperature changes of patients with severe acute respiratory syndrome: a comparative study].

OBJECTIVE: To explore the patterns of temperature changes of patients with the severe acute respiratory syndrome (SARS) and the effect of glucocorticoid hormone on the temperature of these patients. METHODS: The clinical data of 94 SARS cases treated during the outbreak of SARS in South China in 2003 were collected for a retrospective review. According to different treatment regimens, the patients were divided into hormone group (n=35) and non-hormone group (n=59). The control groups consisted of 65 patients with interstitial pneumonia, 78 with bacterial pneumonia and 57 with upper respiratory tract infection. The changes in body temperature were compared between the SARS patients and those with other respiratory diseases and the effect of glucocorticoid hormone on controlling body temperature of the SARS patients was explored. RESULTS: The body temperature of patients with the 4 diseases all exhibited obvious reduction 7 days after hospitalization (P<0.001) with only subsequent mild fluctuation within the basically normal range. At each time point of measurement, the body temperature of SARS patients was significantly higher than that of patients with other diseases (P<0.03), with a fluctuation of 0.2 to 0.5 degrees C; and following a pattern of variation similar to those of the other diseases. Of the 4 time points of daily measurement, namely 6, 10, 14 and 18 o'clock, the temperature measured at 14 o'clock was significantly higher than those at the other 3 time points (P<0.001). Hormone therapy did not significantly affect the temperature of SARS patients (P=0.180), who had longer duration of high fever. CONCLUSION: SARS patients have higher body temperature and longer duration of high fever. Hormone therapy may not produce significant effect in controlling the temperature of SARS patients.

[Urinary S100B protein and lactate/creatinine ratio measurements: a tool for the early identification of neonatal hypoxic-ischemic encephalopathy].

OBJECTIVE: To investigate the value of urinary S100B protein and lactate/creatinine ratio determination in early identification of neonatal hypoxic-ischemic encephalopathy (HIE). METHODS: The levels of urinary S100B protein and urinary lactate/creatinine ratio were detected in 58 full-term newborn infants with HIE on the first, second and third day after birth. The severity of clinical manifestations, including the degree of encephalopathy, was assessed within 7 days after birth. Twenty five normal neonates were enrolled into the control groups. RESULTS: (1) The urinary S100B level of HIE neonates was significantly higher in samples collected throughout the monitoring period than those of the normal control groups (all P < 0.001). The urinary lactate/creatinine ratio of the HIE neonates was also significantly higher than that of normal control groups within the first day (P < 0.001). (2) A significantly positive correlation was found between the level of urinary S100B protein within three days and the urinary lactate/creatinine ratio within the first day and between the level of urinary S100B protein within three days and clinical degree (P < 0.05). (3) When S100B concentration was 0.47 microg/L and urinary lactate/creatinine ratio was 0.55, the sensitivity and specificity of detecting the third day urinary S100B alone, were respectively 90.4%, 91.9%. Detecting it associated with the first day urinary lactate/creatinine ratio could increase the sensitivity and specificity (respectively 98.8% and 97.4%) for predicting development of HIE. CONCLUSION: On the basis of clinical manifestations of asphyxic neonatals, detecting the level of urinary S100B within three days and the first day urinary lactate/creatinine ratio may be of important value in early diagnosis and grading of HIE.

[Regulatory effects of inhaled steroids on expression of matrix metalloproteinase-9 and its inhibitor in asthmatic rats].

OBJECTIVE: To investigate the role of matrix metalloproteinase-9 (MMP-9) and its tissue inhibitor (TIMP-1) in the pathogenesis of bronchial asthma and assess the effect of steroid treatment on MMP-9 and TIMP-1 levels. Matrix metalloproteinases are a family of zinc and calcium-dependent endopeptidases. Many MMPs such as MMP-1, MMP-2, MMP-3 are associated with asthma, in which MMP-9 is the key factor in asthma. Tissue inhibitor-1 of metalloproteinases is a specific inhibitor of MMP-9; the MMP-9 and TIMP-1 imbalance could lead to airway inflammation and remodeling in lung disease such as asthma. METHODS: Forty Wistar rats were divided into 4 groups randomly: control, asthma model 7 days (7-day group), asthma model 21 days (21-day group) and steroid treatment groups. Asthma model of rats were established by ovalbumin (OVA) sensitization and challenge with mist inhalation. The expression of MMP-9 and TIMP-1 in lung tissues was detected by immunocytochemistry, RT-PCR and Western blotting. RESULTS: (1) By observing the changes of action, tracing respiratory curves, detecting level of serum IgE level and observing the lung tissues sections, the authors demonstrated that the rat asthmatic models were successfully established. (2) The lung tissue sections of the asthma groups stained with hematoxiline and eosin (HE) showed many inflammatory cell infiltrations around the bronchioli and accompanying arterioles, hyperplasia of caliciform cells, broken bronchial mucous membrane and thickening of submucosal layer. The hyperplasia of airway smooth muscle and basement membrane were more significant in asthma model 21-day group than that in 7-day group. These changes were improved after treatment. (3) The expression of MMP-9 in rat's lung tissues: the expression was 2.71 +/- 0.37 in 7-day group, 1.76 +/- 0.27 in 21-day group, 0.88 +/- 0.18 in the treatment group and 0.52 +/- 0.10 in the control group (F = 151.52, P < 0.01). The expression of TIMP-1 in rat's lung tissues was 1.13 +/- 0.19 in the 7-day group, 1.55 +/- 0.24 in 21-day group, 0.77 +/- 0.15 in the treatment group and 0.47 +/- 0.08 in the control group (F = 69.46, P < 0.01). (4) The results of immunocytochemistry and protein expression were consistent with those of RT-PCR. CONCLUSION: The protein and mRNA expression level of MMP-9 and TIMP-1 was high in asthmatic rat's lung tissues. Down-regulation of the expression of MMP-9 and TIMP-1 by steroids may be one of the mechanisms by which airway inflammation and remodeling are inhibited in asthma.

[The expression of protein kinase C and the regulatory effect of nerve growth factor in the lung and the visceral sensory afferent system of asthmatic guinea pigs].

OBJECTIVE: To explore the expression of protein kinase C (PKC) and the regulatory effect of nerve growth factor (NGF) in the lung and the visceral sensory afferent system (C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord) of asthmatic guinea pigs. METHODS: Forty guinea pigs were divided to four groups, a saline control group (group A, n = 8), a provocation alone control group (group B, n = 8), an asthmatic group (group C, n = 12) and an anti-NGF group (group D, n = 12). The alterations of PKC immunoreactivity were investigated by means of immunohistochemistry in the C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord of all groups. The expressions of NGF and PKC were investigated by Western blot in the lung, C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord of all groups. The results were analyzed by the Luzex-F real time image analysis system and the gel imaging analysis system respectively. RESULTS: (1) Immunohistochemistry results: The absorbency (A) values of PKC were 0.102 +/- 0.009, 0.113 +/- 0.009, 0.106 +/- 0.005 and 0.116 +/- 0.007 in the C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord of group A and group B respectively. There was no significant difference between group A and group B (P > 0.05). The A values of PKC were 0.215 +/- 0.014 and 0.176 +/- 0.010 respectively in the C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord of group C, which were significantly different compared with group A (P < 0.01). The A values of PKC were 0.140 +/- 0.008 and 0.130 +/- 0.011 respectively in the C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord of group D, which were significantly different compared with group C (P < 0.01). (2) Western blot results: Compared with group A (the relative A values were 0.51 +/- 0.02, 0.43 +/- 0.01 and 0.92 +/- 0.02 respectively) and group B, the expression of PKC (the relative A values were 1.51 +/- 0.01, 1.40 +/- 0.03 and 2.22 +/- 0.02 respectively) increased markedly in the lung, C(7)-T(5) spinal ganglia and the corresponding posterior horn of the spinal cord of group C; however, the expression of PKC of group D (the relative A values were 0.80 +/- 0.03, 0.83 +/- 0.01 and 1.12 +/- 0.02 respectively) decreased markedly in comparison with group C. CONCLUSION: The present results indicate that PKC might be involved in the pathogenesis of bronchial asthma, and NGF can upregulate the expression of PKC.

A novel mutation of estrogen receptor gene detected in girls with precocious puberty.

Female precocious puberty is caused by premature activation of the hypothalamic-pituitary-gonadal axis, exposure to exogenous sex steroid hormones, and the presence of endogenous sex steroids caused by various factors. Estrogen is the final key factor to start onset of puberty. However,in some cases of precocious puberty in girls estrogen elevation could not be detected. The raised sensitivity of estrogen receptor, which may caused by ESR1 mutation or polymorphism, has been frequently mentioned for interpreting the etiology of sporadic low estrogen type cases. But no case evidence has been found in clinical practice. For the purpose of screening possible mutations in estrogen receptor gene, leukocyte genomic DNA were collected from 16 girls with precocious puberty of sporadic low estrogen, and exons of ESR1 were amplified and analysized using PCR-SSCP/silver staining method. A single strand conformation change in exon 8 was found in one of the patients (No. 14). The suspected fragment were cloned to a T vector and sequenced for analysis. Sequencing of these clones revealed that this conformation change is caused by a C to T transition. This mutation results in the replacement of arginine by cystine at position 548 of ESR1 protein. The mutation created an extra Btsl digest site and made it can be readily identified by PCR-PFLP method. Further detection using this method, and sequencing of cloned exon8 colonies from patients proved that the patient No. 14 is Arg548/Cys548 heterozagous in genotype. This mutation increased hydrophobility of the area dramatically. The position and the conservative of this residue in vertebrates suggested Arg548 may play an important role in ESR1 function. For study the role of this mutation in the onset of precocious puberty, a firefly luciferase reporter plasmid pGL3-promoter-ERE was constructed, and a pCR3. 1-hermut pisimid expressing Cys548 ER was constructed based on wild type pCR3. 1her. Co-transfection of reporter and pCR3. 1 -hermut in CMF-7 cell strain proved that Cys548 mutant can significantly increase the transcription activity over the Arg548 wild type.
