[Identification and phylogenetic study of pathogenic bacteria causing ulcer disease of cultured Turbot (Scophthalmus maximus)].

An outbreak of ulcer disease of the cultured turbot Scophthalmus maximus L. occurred in an indoor farm in Haiyang, Shandong Province in the sweltering August of 2004 and caused significant mortality. At the beginning, infected turbots displayed sluggish swimming and anorexia. Several days later, the turbot eyes swelled, the fins and tail turned red, and the back gradually ulcerated. Dissection of the moribund fish showed that the ulcerative gills paled, the liver becamed bloodshot, the kidney and gallbladder swelled, the intestinal wall became filmy and bloodshot. The time course from appearance of disease signs to death lasted about a week. A dominant strain of bacteria, which was Gram-negative and short rod with single polar flagellum under electron microscope, was isolated from the diseased turbot and designated as H1. In artificial infection test, all fish of the experimental groups died in a week after intramuscularly injected with bacterial suspension, while the control group showed no signs in 10 d post-challenge. The moribund experimental fish had similar gross signs as the natural infected fish. The bacteria re-isolated from the challenged fish also had the same characteristics as H1, which proved that the isolate H1 was the pathogenic bacteria that triggered this ulcer disease. Different methods were used to identify the pathogenic bacteria. The identification result by API 20NE and API 20E system indicated that H1 was Aeromonas hydrophila, with 99% reliability and 61.5% respectively. While traditional biochemical identification revealed that H1 exhibited relatedness to Vibrio harveyi. In order to confirm the different result, a 1424 bp sequence of H1's 16S rDNA was amplified and compared with other Vibrio spp. in GenBank, homology analysis and phylogenetic study showed that H1 has the highest similarity to V. harveyi, with 99% identity. According to morphological features, physiological and biological characteristics and 16S rDNA homology comparison of the bacteria, the pathogenic bacteria were V. harveyi. Drugs sensitivity test showed that the pathogenic bacteria were highly sensitive to nitrofurantoinum and ceftriaxone sodium etc. This is the first report that V. harveyi was found as the pathogenic bacteria of cultured turbot in China. The research suggests that V. harveyi should be regarded as an important pathogen of turbot and can causes ulcer disease under conditions of high temperatures. Therefore, it is necessary to prevent against ulcer disease in culture turbot in summer.

[CD4+CD25high regulatory cells in peripheral blood of nasopharyngeal carcinoma].

OBJECTIVE: The current study was designed to investigate the changes in peripheral CD4+ CD25high regulatory T cells (CD4+ CD25high Tr) in nasopharyngeal carcinoma (NPC). METHOD: The peripheral was collected blood from 54 patients with NPC and 15 healthy controls, by using monoclonal antibodies, the blood samples were evaluated with the flow cytometry for lymphocyte subsets and Tr cells. RESULT: The proportion of CD4+ T cells, and the ratio of CD4+/CD8+ in NPC group was significantly less than those of the healthy controls (P < 0.05), but the proportion of CD4+ CD25high Tr cells was higher than those in controls (P < 0.01). The proportion of CD4+ CD25high Tr cells in the late stage was higher than those in the early stage [(stages I + II (2.28 +/- 1.07) %; stage III (3.65 +/- 1.21) %; stage IV (4.72 +/- 0.87) %)]. Comparing with the stages I + II, the proportion of CD4+ CD25high Tr cells in the stage III or IV was higher than those in controls (P < 0.01); and more, the proportion of CD4+ CD25high Tr cells in the stage IV was higher than those in the stage III (P < 0.01). CONCLUSION: The relative increase in CD4+ CD25high Tr cells in peripheral blood may be related to immunosuppression and tumor progression in patients with NPC. This finding suggests that CD4+ CD25high Tr cells in peripheral blood of NPC may be positive for prognosis analysis. Deletion of the CD4+ CD25high Tr cells may induce tumor immunity effectively and may be a new tumor treatment method.

[The preparation of the multilevel holes and analysis of the pure titanium surface].

PURPOSE: Using sandblast, double acid-etching and H(2)O(2)/HCl solution treatment to establish a new implant surface, and to analyze the surface's morphology. METHODS: After being polished, sandblasted, cleaned, the titanium specimens were divided into six groups, i.e.A to F group. The specimens of group A were treated with low concentration of HF/HNO(3) solution, Group B was treated with low concentration of HF/HNO(3) solution and then H(2)SO(4)/HCl solution, Group C was treated after Group B and further with H(2)O(2)/HCl solution at 80degrees centigrade for 30 minutes, and subsequently heated at 400degrees centigrade for 1 hour. Group D was treated with high concentration of HF/HNO(3) solution, Group E was treated after Group D and then H(2)SO(4)/HCl solution, Group F was treated after E group and further with H(2)O(2)/HCl solution at 80degrees centigrade for 30 minutes, and subsequenty heated at 400degrees centigrade for 1 hour. RESULTS: The titanium surface sandblasted and ultrasonically cleaned exhibited a great deal of particles. The titanium plates treated with low concentration of HF/HNO(3) solution still had some particles, but there were much nano-holes on the titanium surface. The titanium plates treated with high concentration of HF/HNO(3) solution had no particles on the surface, but showed much micron-holes on the titanium surface. The titanium plates treated with H(2)SO(4)/HCl solution after HF/HNO(3) solution etching had all particles removed and got multilevel holes on the surface. The last H(2)O(2)/HCl method produced an anatase titania gel layer on the specimen surface. CONCLUSION: By altering the concentration of HF/HNO(3) solution, we could make different sizes of apertures on the titanium surface. Double acid-etching can remove all sandblasting particles embedded in titanium surfaces and form multilevel holes. The H(2)O(2)/HCl method can turn the rough non- crystallographic TiO(2) layer into the anatase titania gel layer.

[Design of a quantitative analysis software system for myocardial contrast echocardiography].

This article reports a quantitative analysis software system for myocardial contrast echocardiography (MCE). It can measure the signal intensity of grayscale images and power Doppler images, draw the time-intensity curves of variations on the intensity of microbubbles scattering in subendocardial layer and subepicardial layer with the pulsing intervals, and estimate the hemodynamic parameters by nonlinear regression analysis. This system has been applied to a study on 20 healthy volunteers, and the results suggest that the software has the capacity for bringing the quantitative analysis of MCE to success. The MCE software system conforms to the DICOM standard and can be integrated into PACS.

[Effective control of excitable waves in 2D cardiac excitable media].

We propose a method for effective control of patter on dissipative system by use of little perturbation analysis, and apply this nonuniform feed back method to control the polarization wave of heart represented by FHN equation. In the numerical experiment, we successfully alter the propagating direction of planar waves and drift the spiral waves to boundary without resonant repulsion. The effective control of the excited system will be used to study the mechanism of defibrillation and that is our interesting work.

[Arthropod community structure and its fuzzy clustering analysis in jujube orchard].

Based on the principles of community ecology and by the method of fuzzy clustering, this paper studied the temporal and quantitative dynamics of the arthropod community and sub-community in jujube orchard (Feidong 2003), aimed to utilize natural enemy resources for pest control. There were 52 species of arthropod belonging to 47 families in the orchard. Arachnida and Cecidomyidae had a higher relative abundance than other families being 0.3465 and 0.2309, respectively. The individuals of the arthropod community were the highest on 8 June and 21 September. The total community and pests-neutral insect sub-community could be clustered into 4 clusters, respectively. For total community, the clusters (T = 0.92) were those on 22 March, 6 April, 22 November, and on other dates, and for pests-neutral insect sub-community, they (T = 0.94) were those on 22 March, 6 April, 8 June, and on other dates. The natural enemy sub-community could be clustered into 3 clusters (T = 0.94), i. e., those on 22 March, on 6 April, and on other dates. The results of cluster analysis partly reflected the seasonal differences of total community and sub-community, while the temporal overlaps of cluster results reflected the complexity of community structure.

[Spatial distribution pattern and space dependence of Contarinia sp. and Erigonidium graminicola in jujube orchard].

With the principles and methodologies of geostatistics, this paper studied the spatial structure and space dependence of Contarinia sp. and its natural enemy Erigonidium graminicola in a jujube orchard at different date. The results indicated that the semivariogram of Contarinia sp. on 8 June, 27 August and 19 October could be described by spherical model, showing an aggregated spatial distribution with the ranges of spatial dependence being 26.3, 7.8 and 22.0 m, and the degrees of spatial dependence being 88.05%, 85.77% and 87.58%, respectively. The semivariogram of Erigonidium graminicola could be also described by spherical model, showing an aggregated distribution with the ranges of spatial dependence being 28.3, 22.5 and 22.2 m, and the degrees of spatial dependence being 90.09%, 87.96% and 85.72%, respectively. It is suggested that Erigonidium graminicola is the dominant natural enemy against Contarinia sp. population in the examined jujube orchard.

[Tiam1 expression correlates with the biological behavior of human colorectal carcinoma cells].

OBJECTIVE: To investigate whether the Tiam1 gene expression enhances the invasive and metastatic capabilities of colorectal carcinoma cells. METHODS: Endogenous expression of Tiam1 in five colorectal carcinoma cell lines was investigated by RT-PCR. Tiam1/C1199HA cDNA was transfected into HT29, a colorectal carcinoma cell line without endogenous Tiam1 expression. RNA and protein expression of Tiam1 gene in the transfectants were detected by RT-PCR, immunohistochemistry and Western blot respectively. The biological behaviors of the transfectants were investigated by MTT and in-vitro invasion assays. RESULTS: Tiam1 gene was highly expressed in LoVo and SW620 cells. Low level expression was seen in HCT116 and SW480 and no expression was found in HT29. Transfection of Tiam1 significantly increased the proliferation of HT29 cells along with markedly enhanced in-vitro invasion and metastasis. CONCLUSIONS: Tiam1 gene plays an important role in the invasion and metastasis of colorectal carcinoma. It may be a useful marker for metastasis of colorectal carcinoma.

[Probing R6G single molecules by surface-enhanced resonance Raman scattering].

R6G was incubated in three kinds of silver colloid with different low concentrations, and its surface-enhanced resonance Raman scattering (SERRS) was studied with a confocal microscopy system. Each sample exhibited different spectrum character. At the single-molecule level the SERRS-spectra were recorded in 10(-13) mol x L(-1) dye colloidal solution. Some spectral inhomogeneous behavior was recorded such as spectral diffusion, intensity fluctuation of vibrational lines and spectral polarization, and even on/off blinking and splitting of some lines within the spectrum of one molecule. Polarization of excitation light and background subtraction of signal have an important influence on the SERRS spectra detection of single-molecule, which was analyzed in the present paper.

BAG5 inhibits parkin and enhances dopaminergic neuron degeneration.

Loss-of-function mutations in the parkin gene, which encodes an E3 ubiquitin ligase, are the major cause of early-onset Parkinson's disease (PD). Decreases in parkin activity may also contribute to neurodegeneration in sporadic forms of PD. Here, we show that bcl-2-associated athanogene 5 (BAG5), a BAG family member, directly interacts with parkin and the chaperone Hsp70. Within this complex, BAG5 inhibits both parkin E3 ubiquitin ligase activity and Hsp70-mediated refolding of misfolded proteins. BAG5 enhances parkin sequestration within protein aggregates and mitigates parkin-dependent preservation of proteasome function. Finally, BAG5 enhances dopamine neuron death in an in vivo model of PD, whereas a mutant that inhibits BAG5 activity attenuates dopaminergic neurodegeneration. This contrasts with the antideath functions ascribed to BAG family members and suggests a potential role for BAG5 in promoting neurodegeneration in sporadic PD through its functional interactions with parkin and Hsp70.

Novel biodegradable polymers as gene carriers.

This study investigated two new biodegradable polymers as gene controlled-released coatings for gene transfer. Poly(ethylene glycol)-co-poly(D,L-lactic acid) (PELA) and poly(ethylene glycol)-co-poly(lactic acid)-co-poly(glycolic acid) random copolymer (PELGA) were synthesized and used as microspheres matrices with encapsulated plasmid pCH110. The plasmid loading efficiency, cytotoxicity, transfection efficiency and in vitro degradation and release profiles of microsphere complexes were evaluated in details. The biodegradable polymers showed high DNA loading efficiency and low cytotoxicity as gene controlled-released coatings, and the poly(ethylene glycol) (PEG) contents of polymer matrices influenced the diameter, loading efficiency and transfection efficiency of plasmid DNA within the microspheres. The average diameters of PELA and PELGA microspheres were between 0.5 and 1.5 microm, and the plasmid loading efficiency was 62 and 73% for PELA and PELGA microspheres with 10% PEG content, respectively. In vitro testing showed a gradual release profile of DNA from polymeric matrices. The polymers/DNA microspheres had high transfection efficiency and early gene expression and maintenance of gene expression level for up to 96 h, although transfection efficiency were slightly lower than that of liposome in the initial 24 h. The biodegradable polymeric materials possess potential superiority as gene carriers.

Tetraspanin CD82 attenuates cellular morphogenesis through down-regulating integrin alpha6-mediated cell adhesion.

Tetraspanin CD82 has been implicated in integrin-mediated functions such as cell motility and invasiveness. Although tetraspanins associate with integrins, it is unknown if and how CD82 regulates the functionality of integrins. In this study, we found that Du145 prostate cancer cells underwent morphogenesis on the reconstituted basement membrane Matrigel to form an anastomosing network of multicellular structures. This process entirely depends on integrin alpha6, a receptor for laminin. After CD82 is expressed in Du145 cells, this cellular morphogenesis was abolished, indicating a functional cross-talk between CD82 and alpha6 integrins. Interestingly, antibodies against other tetraspanins expressed in Du145 cells such as CD9, CD81, and CD151 did not block this integrin alpha6-dependent morphogenesis. We further found that CD82 significantly inhibited cell adhesion on laminin 1. Notably, the level of alpha6 integrins on the cell surface was down-regulated upon CD82 expression, although total cellular alpha6 protein levels remained unchanged in CD82-expressing cells. This down-regulation indicates that the diminished cell adhesiveness of CD82-expressing Du145 cells on laminin likely resulted from less cell surface expression of alpha6 integrins. As expected, CD82 physically associated with the integrin alpha6 in Du145-CD82 transfectant cells, suggesting that the formation of the CD82-integrin alpha6 complex reduces alpha6 integrin cell surface expression. Finally, the internalization of cell surface integrin alpha6 is significantly enhanced upon CD82 expression. In conclusion, our results indicate that 1) CD82 attenuates integrin alpha6 signaling during a cellular morphogenic process; 2) the decreased surface expression of alpha6 integrins in CD82-expressing cells is likely responsible for the diminished adhesiveness on laminin and, subsequently, results in the attenuation of alpha6 integrin-mediated cellular morphogenesis; and 3) the accelerated internalization of integrin alpha6 upon CD82 expression correlates with the down-regulation of cell surface integrin alpha6.

Piperidine-containing beta-arylpropionic acids as potent antagonists of alphavbeta3/alphavbeta5 integrins.

The synthesis and SAR of a new class of piperidine-based alphavbeta3/alphavbeta5 integrin antagonists is described. Replacement of an amide bond in a prototype isonipecotamide by a C-C isostere, and adjustment of the spacer length between the carboxylic acid and basic moieties, led to low nanomolar antagonists of alphavbeta3 and/or alphavbeta5 integrins with excellent selectivity versus alpha(IIb)beta3.
