[Distribution of functionally differing cells in immunocompetent organs on haloperidol administration].

Haloperidol blockade of the dopaminergic system entails redistribution of functionally different cells. Suppressors of IgM and IgG response migrate, respectively, from the spleen and lymph nodes to the bone marrow whose cell transfer leads to inhibition of immune response. The data obtained indicate common regularities in cell distribution under blockade of the dopaminergic and activation of serotoninergic systems. Therefore, at the basis of inhibition of immunogenesis there lie monotypic processes of suppressor cell migration whatever the system responsible for it.

[Formation of an IgM and an IgG immune response depending on the redistribution of T- and B-subpopulations under the action of serotonin].

The syngeneic transfer of spleen cells or spleen and lymph node cells from donors with an elevated serotonin level stimulated, in comparison with the control animals, immune response in the recipients subjected to sublethal irradiation, which was manifested by an increase in the number of plaque-forming and rosette-forming cells. After the combined transfer of spleen cells and bone marrow cells from similar animals a decrease in the number of plaque-forming and rosette-forming cells was observed, while after the transfer of spleen and thymus cells the intensity of immune response remained unchanged. Serotonin was supposed to induce the redistribution of T and B cells in the non-immunized animals, so that suppressor cells migrated from the spleen and the lymph nodes to the bone marrow.

[Rosette formation in a cell population syngeneic transfer system from donors with an altered serotinin level].

The formation of primary immune response has been studied during transplantation to sublethally irradiated recipients of cells from different immunocompetent organs of non-immune intact donors as well as of those from animals injected serotonin. Considerable variations in the development of the immune reaction have been mainly discovered due to IgM-forming cells, both during the transfer of spleen cells alone and in combined injection of these cells with bone marrow and thymus cells from the same donors. The results obtained suggest a possible serotonine influence on variations in the ratio and migration of T and B cells in different organs and on T and B suppressor cells.

[IgM and IgG rosette formation in the primary and secondary immune response in the system of syngeneic transfer of spleen cells].

In the system of syngenous transfer of cells a study was made of the dynamics of IgM- and IgG- of the rosette-froming cells of the spleen in the primary and secondary immune response in mice of the CBA inbred strain immunized with sheep erythrocytes. It was shown that in prolongation of the interval between donor immunization and the transfer of cells to the recipient with his simultaneous immunization for up to 40 days there occurred an increase of the IgG-memory; as to IgM memory-it is expressed with shorter intervals. It is supposed that rosette-forming cells were not bearers of immunological memory.

[Effect on the immobilization-induced stress on the immune response in mice with various behavioral stereotypes].

CBA and C57BL/6J mice with aggressive and submissive types of zoo-social behavior fixed during 10-day confrontation testing demonstrated different immune response to immobilization stress. In aggressive mice with confrontation experience stress resulted in a decrease in the immune response in comparison with aggressive inexperienced mice. Therewith, the immune response of CBA mice did not differ from the control level (the immunized mice without experience of victories and defeats), and in C57BL/6 mice the immune response was two times lower than in the control. In contrast to the control and aggressive mice, stress did not suppress the immune response in submissive animals. It is suggested that stress-induced changes in the immune response depend on zoo-social rank of an individual underlain by a particular neurochemical pattern of the brain.

[The central mechanism of the immunomodulating action of vasopressin and its structural analogs].

It is shown that the effect of neuropeptides depends on the dose and their structure. Considerable immunostimulation is observed after administration of [8-arginine]vasopressin and des-9-glucine-[8-arginine]vasopressin as against the control. This stimulation is prevented by the preliminary heloperidol blockade of dopaminergic receptors, i.e. it is dopamine-dependent. At the same time, injection of tetrapeptide vesopressin-(2-5) induces deep immunosuppression in all the applied doses. The peptide immunosuppression is shown to be a result of interaction with another neuromediator (serotoninergic); it has no effect under preliminary cuproheptadine blockade of C-2 serotonin postsynaptic receptors. The immunomodulating effect of the neuropeptides studied is central one and is realized via the hypothalamus-hypophysis complex.

[Changes in the immune reaction of animals under conditions of drug-induced activation and blockade of D1 dopamine receptors].

A highly selective dopamine D1 receptor agonist (compound SKF 38393) administered in a dose of 20 mg/kg produced a significant increase in the number of rosette-forming cells (RFCs) in the spleen of Wistar rats on the 5th day upon their immunization with goat erythrocytes (5 x 10(8)). At the same time, a specific blocker of these receptors (compound SCH 23390) in a dose of 0.5 or 1 mg/kg inhibited the immune response of rats. In C57BL/6J mice, SCH 23390 (0.25, 0.5, or 1 mg/kg) also significantly reduced the number of RFCs. Preliminary blockade of the D1 dopamine receptors with SCH 23390 (1 mg/kg) prevented the immunostimulating effect of SKF 38393. These data indicate that D1 dopamine receptors are involved in immunomodulation.

[Activation and blocking of 5-HT1A receptors influence on the immune response in CBA mice].

5-HT1A type serotonin receptors influence the immunomodulating action of the selective preparations 8-OH-DPAT (5-HT1A receptor agonist) and WAY-100635 (5-HT1A receptor antagonist) in CBA mice. The activation of 5HT1A receptors with 8-OH-DPAT (1 mg/kg) decreased, while their blocking with WAY-100635 (1 mg/kg) increased the reaction intensity at the peak of response to immunization with ram erythrocytes. Preliminary blocking of the 5-HT1A receptors with WAY-100635 prevented the inhibiting action of 8-OH-DPAT.

[Significance of initial emotional state for neuroimmunomodulation under conditions of activation or blockade of 5-HT1A receptors].

Selective agonist of 5-HT1A receptors--8-OH-DPAT (1 mg/kg) induced a suppression of the immune reaction in aggressive male CBA mice immunized with SRBC (5 x 10(8)). In submissive mice with 10-day defeat experience in confrontation tests, the activation of 5-HT1A receptors with 8-OH-DPAT did not alter the immune response, whereas the application of selective antagonist of 5-HT1A receptors WAY-100635 increased the immune reaction only in submissive mice. It is concluded that activation or blockade of 5-HT1A receptors produced different effects on the immune function of CBA mice depending on the initial emotional state which is known to be provided in aggressive and submissive animals by different activities of the brain neurotransmitter systems including the 5-HTergic system.

[Influence of prostaglandin synthesis blockade on the immunosuppressive effect of serotonin and haloperidol].

Administration of indomethacin (1.5 and 10 mg/kg) to CBA mice was found to produce a considerable increase of the number of plaque- and rosette-forming cells irrespective of the dose and the interval between its injection and immunization of the mice with ram erythrocytes (5 X 10(6)). The immunogenesis stimulation preserved in the mice after a lesion of the pituitary peduncle. Indomethacin was shown to prevent a decrease of the immune response produced by administration of serotonin (50 mg/kg) and haloperidol (1 mg/kg).

[The peptide modulation of immune reactions].

A dose-dependent immunomodulating effect of synthetic low-molecular weight peptides was found in CBA mice. Talcin and its analogue, rigin, tripeptide SKE, substance P and its analogue, and vasopressin were shown to produce an immunostimulating effect, whereas vasopressin tetrapeptide, arginyl-asparagine, and tripeptide SKD showed an immunosuppressive effect. The immunomodulatory effect of the peptides tested was associated with the presence of amino acids such as arginine, lysine, tyrosine, glutamic acid in their chemical structure. These amino acids are essential in the mechanisms of immune responses.
