Hemorheological and cerebroprotective activity of Lychnis chalcedonica L. extract in rats with cerebral ischemia.

Considerable hemorheological changes and depression of EEG parameters were revealed in rats with cerebral ischemia. Course peroral treatment with scarlet lightning extract in a daily dose of 150 mg/kg for 5 days reduced the severity of hemorheological disorders. It manifested in a decrease in whole blood viscosity, plasma viscosity, erythrocyte aggregation, and fibrinogen concentration and increase in deformability. The extract of lightning extract improved EEG activity in rats with cerebral ischemia.

Effect of ascovertin on morphological changes in rat retina exposed to high-intensity light.

Experiments on rats showed that high-intensity light exposure (6000 lux, 6 h) caused focal injuries in the retina. The most sensitive structures were neurosensory cells, pigmented epithelium, radial gliocytes, and choroid capillaries. Injection of ascovertin led to disappearance of foci of injuries, limited blood supply disorders in the retina, and destruction of neurosensory and glial cells.

Pharmacological regulation of functional activity of stem cells in restoration of the myocardium during the postinfarction period.

The effect of granulocytic CSF on myocardial tissue recovery after acute myocardial infarction was studied on rats. A course of granulocytic CSF after ligation of the left coronary artery normalized ECG parameters and morphological picture of the myocardium 1 month after treatment. It was shown on mouse model of myocardial infarction that this process was associated with more intensive mobilization and homing of mesenchymal stem cells in the heart.

[Morphological changes of rat ovaries after their monopolar coagulation and ascovertin correction].

Using the methods of light and electron microscopy, morpho-functional changes of the ovaries of albino rats after their monopolar coagulation were studied. Electrocoagulation was shown to cause hemodynamic changes in a perifocal area and short-term stimulation of follicular growth, but starting already at postoperational day 3, their atresia was increased, while by day 30 a significant reduction in number of both primordial and growing follicles, as well as of corpora lutea, was found. Ultrastructural changes of oocytes in primary and secondary follicles outside the coagulation zone were demonstrated at early stages and included the reduction of porous plate, cortical and yolk granule numbers, and the increase in numbers of lysosomes and multivesicular bodies. Ascovertin, due to its antioxidant properties and by improving hemorheological parameters, exerts a protective action on ovarian generative apparatus and the state of microcirculation in the organ.

Hemorheological and antioxidant effects of Ascovertin in patients with sclerosis of cerebral arteries.

The clinical trials on 31 patients with arteriosclerosis and I-II stage discirculatory encephalopathy to assess an ability of Ascovertin to limit hemorheology abnormalities were carried out. In patients with discirculatory encephalopathy was a distinct increase in blood viscosity which was induced by disturbances of cell rheological factors: increase in aggregation of erythrocytes and decrease in their deformability were observed in comparison with indices in the group of healthy volunteers. No difference in plasma viscosity and fibrinogen was found. The treatment with Ascovertin in patients with discirculatory encephalopathy improved their attention, memory, mental performance, normalized sleep, releaved headache, decreased fatiquebility, led to the decrease in blood viscosity values, the reduction of pathological erythrocyte hyper aggregation and the improvement of erythrocyte deformability. We partly connect this clinical effect and hemorheology activity of Ascovertin with its antioxidant property--there was found impressive lipid peroxidation suppression. No significant changes in hemorheological and lipid peroxidation indices were observed in patients without Ascovertin.

[Clinical efficacy of a novel hemorheological drug ascovertin in patients with vascular encephalopathy].

Patients with stages I and II of vascular encephalopathy developing on the background of atherosclerosis were treated with ascovertin during 21 days. Ascovertin is a complex of flavonoid dihydroquercetin and ascorbic acid. The study group included 21 patients aged 45-65 years and a comparison group consisted of 10 age-matched patients un treated with ascovertin. The ascovertin treatment relieved headache, reduced vertigo and fatigability, improved cognitive functions. The reliable diminishing of whole blood viscosity due to improvement of cellular rheology indices (decrease of aggregation and increase of erythrocyte deformability as well as decrease of indices of lipid peroxidation in erythrocyte membrane and blood plasma) was observed in the stydy group but not in the comparison one.

Correction of haemorheological disturbances in myocardial infarction by diquertin and ascorbic acid.

In a model of the high blood viscosity syndrome, developed after myocardial infarction in rats, it was observed that a therapy of a combination of diquertin (20 mg/kg) and ascorbic acid (50 mg/kg) for a -period of 6 days, resulted in an improvement of haemorheological indices. The decrease in blood -viscosity was primarily due to an improved deformability of erythrocytes, and to some extent, due to a decrease in the content of plasma fibrinogen and erythrocyte aggregation.

Correction of the high blood viscosity syndrome by a mixture of Diquertin and Ascorbic Acid in vitro and in vivo.

Diquertin was shown to diminish blood viscosity, to decrease the aggregation of erythrocytes, and to increase their deformability using a model of the high blood viscosity syndrome in vitro. A mixture of Diquertin with Ascorbic Acid was more effective in improving rheological indicators of blood, then either Diquertin or Tanakan. On a model of chronic brain ischemia, which is accompanied by significant deterioration of the rheological properties of blood, it was shown that a therapy with a mixture of Diquertin and Ascorbic Acid decreases the expression of the high blood viscosity syndrome.

Effect of ascovertin on morphofunctional changes in the ovaries in monopolar coagulation.

A course of ascovertin therapy reduced manifestations of perifocal inflammation and atresia of growing follicles, corrected vascular disorders and changes in the tissue volume ratios outside the necrotic zone after monopolar coagulation of the ovaries in rats. The drug prevented the increase in blood viscosity, erythrocyte aggregation, and LPO activation in the early postoperation period.

Ischemia-induced changes in synaptoarchitectonics of brain cortex and their correction with ascovertin and Leuzea extract.

Peroral administration of 70 mg/kg ascovertin and 150 mg/kg Leuzea extract to rats with cerebral ischemia for 5 days prevented destructive changes and decrease in the density of synapses in the cerebral cortex. These preparations activated compensatory and reparative mechanisms underlying plasticity of the synaptic pool, which was realized through hypertrophy and destruction of synaptic contacts. Ascovertin possessed more pronounced cerebroprotective activity than Leuzea extract.

Course of shock in rats with different resistance to shockogenic trauma during Crush syndrome.

Experiments on narcotized rats with crush syndrome showed that low resistant animals developed pronounced hypovolemia, hemoconcentration, blood hyperviscosity, impairment of oxygen metabolism, and central and peripheral hemodynamic disturbances, whereas in highly resistant rats the hemodynamics and oxygen supply to tissues were maintained at a sufficient level, while hemoconcentration and the increase in blood viscosity were less pronounced.

[Disorders of blood rheology in Walker's carcinosarcoma-256 and in cyclophosphamide treatment of rats].

Tumor process development in rats inoculated with cellular suspension of transplantable Walker's carcinosarcoma-256 involved enhanced thickening of blood on day 7. Treatment with 20 mg/kg cyclophosphamide, thrice a day, every other day, retarded tumor process and brought hemorheologic indices further down, at the same time. Similar treatment of intact rats with cyclophosphamide caused hemorheologic disorders too.

Relationship between rheological and hemodynamic changes in rats with crush syndrome.

Narcotized rats with crush syndrome develop severe syndrome of increased blood viscosity. A strict correlation was found between changes in blood viscosity at different shear rates and total peripheral vascular resistance. The severity of central hemodynamic and hemorheological disturbances was different in rats with different reaction of total peripheral vascular resistance to injury accompanied by shock.

Cerebroprotective effects of diquertin and ascorbic acid.

Electron microscopy and electrophysiological studies revealed pronounced structural and functional changes in the brain cortex in rats with experimental cerebral ischemia. Repeated administration of diquertin and ascorbic acid significantly attenuates ischemic damage induced by circulatory disturbances.

[Rheological and hydrodynamic properties of blood from patients with stable angina pectoris: correction with high molecular polyethylene oxide].

Rheological and hydrodynamic properties of blood taken from patients with stable angina pectoris were investigated in different periods (1, 7 and 28 days) of combined treatment (calcium antagonists, nitrates, beta-blockers, aspirin). The high viscosity syndrome (elevated blood viscosity, red cell aggregation, hyperfibrinogenemia) was registered before treatment, 1, 7 and 28 days after beginning of the treatment. Low hydrodynamic index (the speed of blood flow in turbulent regimen) reflected insufficiency of endogenic antiturbulent factor in the blood. The addition of high-molecular-weight polyethylene oxide to blood samples in concentration 2 x 10(-6) promoted normalization of the hydrodynamic index and decline of red cell hyperaggregation.
