Effects of homeopathic arsenic on tobacco plant resistance to tobacco mosaic virus. Theoretical suggestions about system variability, based on a large experimental data set.

CONTEXT: This research aimed at verifying the efficacy of homeopathic treatments by plant-based bioassays, which may be suitable for basic research, because they lack placebo effects and provide large datasets for statistical analyses. OBJECTIVE: To evaluate the effects of homeopathic treatments of arsenic trioxide (As2O3) on tobacco plants subjected to tobacco mosaic virus (TMV) inoculation as biotic stress. DESIGN: Blind, randomized experiment using tobacco leaf disks. MATERIALS AND METHODS: Tobacco plants (Nicotiana tabacum L. cultivar Samsun) carrying the TMV resistance gene N. TMV inoculated leaf disks were floated for 3 days in the following: Distilled water (control). H2O 5 and 45 decimal and centesimal potencies. As2O3 5 and 45 decimal and centesimal potencies. The main outcome measures is the number of hypersensitive lesions observed in a leaf disk. RESULTS: Homeopathic treatments of arsenic induce two effects on the plant: (i) increased resistance to TMV; (ii) decrease variability between experiments (system variability). CONCLUSIONS: In this experimental model two actions of homeopathic treatment were detected: decrease in system variability and enhancement of the natural tendency of the system towards an 'equilibrium point'.

Statistical analysis of the effect of high dilutions of arsenic in a large dataset from a wheat germination model.

This paper describes the statistical analysis of a series of experiments using a simple biological model (wheat germination in vitro), where a large number of wheat seeds were treated with homeopathic potencies of Arsenic trioxide. Some potencies, such as As2O3 40x, 42x and 45x, have repeatedly shown a significant stimulating effect on germination compared to controls, whereas As2O3 35x has a significant inhibiting effect. In some experiments the seeds were stressed before the experiment with a sublethal dose of the same substance. We performed a statistical analysis, both for stressed and non-stressed seed groups, using Poisson distribution as a suitable model for representing the number of non-germinated seeds in a standard experiment with 33 seeds in the same Petri dish. Finally, we have considered the most repeated potencies (30x and 45x), computing the sample odds ratio (OR) and a 95% confidence interval (CI) for the population OR. Our results show significant reproducible effects of some As2O3 decimal potencies, particularly As2O3 45x. In stressed seeds, even decimal potencies of water seem to give significant results compared to control, whereas high dilutions of As2O3 without potentization never show significant effects.

Correlation between plasma HIV-1 RNA levels and the rate of immunologic decline.

To determine the influence of HIV-1 replication on immunologic decline and clinical outcome, we quantified the HIV-1 plasma viral load in 20 patients at different times over a mean period of 10.8 months. Quantitation was performed by branched DNA signal amplification (bDNA) and p24 antigenemia. Immunologic status was assessed through beta 2-microglobulin and CD4+ cell count determinations. CD4+ cell decline was expressed as a slope of the regression line constructed by the logarithms of CD4+ cell count observations. Mean values of plasma viral load were correlated with CD4+ cell decline and mean beta 2-microglobulin levels. Significant correlation was observed between plasma viral load quantified by the bDNA technique and CD4+ cell decline. No significant correlation was observed between plasma viral load quantified by p24 antigenemia and CD4+ cell decline. A significant correlation was observed between plasma viral load and beta 2-microglobulin levels. Immunologic decline was better predicted from HIV-1 RNA levels than from the CD4+ cell count. Significantly higher plasma viral load was observed in patients who had clinical progression of HIV-1 infection. Thus, HIV-1 plasma viral load quantified by a highly reliable technique such as bDNA showed that the immunologic decline is closely related to HIV-1 RNA replication.

Lead toxicosis in a captive bottlenose dolphin (Tursiops truncatus) consequent to ingestion of air gun pellets.

A captive bottlenose dolphin (Tursiops truncatus) in a dolphinarium in Tel Aviv, Israel, had signs of anorexia, weight loss and a reluctance to train over a 4-week period in June 1995 and died shortly thereafter. On necropsy, it had an enlarged, yellow discolored liver, and about 55 air gun pellets in the second stomach. The pellets were composed of 40% lead. Samples of liver and kidney cortex contained 3.6 and 4.2 micrograms/g lead, respectively. There was hemosiderosis in the liver and kidneys, status spongiosus in the brain, and vacuolization in the optic nerve; acid-fast intranuclear inclusion bodies were seen in the kidneys. We propose that chronic lead toxicosis had been induced after the gradual dissolution of the lead-based pellets in the acid environment of the stomach.

Cerebrospinal fluid filtration in a case of severe pneumococcal meningitis.

Current therapy of bacterial meningitis includes high doses of antibiotics and, sometimes, addition of corticosteroids in order to reduce a harmful inflammatory response. The persisting high mortality and rate of sequelae, particularly regarding pneumococcal meningitis, calls for new therapeutic approaches. We report a case of a 71-year-old female with a pneumococcal meningitis treated with extra-corporeal filtration of cerebrospinal fluid (CSF) after 3 days of coma on conventional treatment. The treatment was carried out without complications and the clinical outcome was favourable. We suggest that filtration of the CSF should be considered as a tool to eliminate noxious substances playing a harmful role in deteriorating course of bacterial meningitis.

Improved method for polyamine determination in TMV, a rod-shaped virus.

Polyamines were measured in viruses using different techniques. An improved method of polyamine analysis is reported for tobacco mosaic virus (TMV), a rod-shaped virus (95% protein and 5% RNA), based on HPLC of sonicated PCA-treated highly purified virus suspensions. This method allowed higher and more reliable recovery of TMV polyamines (putrescine, spermidine and spermine) when compared to the HPLC of non-sonicated samples and to thin layer chromatography. It is suggested that sonication acts on PCA-precipitated protein aggregates causing the release of trapped polyamine molecules.

HIV-plasma viral load detection by branched DNA signal amplification.

To detect HIV-1 plasma viral load in seropositive patients, we applied a new molecular quantitation technique: branched DNA signal amplification (bDNA). We performed bDNA with 99 sera from HIV-1 seropositive patients undergoing antiretroviral therapy. We compared the results obtained with p24 antigenemia and CD4+ cell count. The bDNA proved quite sensitive and available for routine use in laboratories.

Quantitation of HCV viraemia by branched DNA signal amplification in patients treated with alpha-interferon--a longitudinal study.

Using bDNA, the plasma viral load trend of HCV-infected patients undergoing IFN therapy was analyzed. Nine patients were enrolled, each assigned to one of three groups, based on IFN response as determined by ALT and AST level trend. HCV was genotyped using DEIA. Each patient's clinical stage was determined by liver biopsy analysis. In nonresponding patients elevated viral loads and biochemical parameters were observed. These values were not influenced by IFN treatment. In relapsed patients the cessation of IFN treatment increased viral load; this was associated with a rise in ALT and AST values. In responders ALT and AST levels remained normal; viral load was low. Patients with elevated HCV viral load showed a worsening in their liver histology during the follow-up period. These results confirm that plasma viral load is a good marker of biochemical change and disease progression.
