A new brain specific protein in the cerebrospinal fluid (CSF) of subacute sclerosing panencephalitis (SSPE).

A rabbit immune serum against SSPE CSF reveals in these CSF, in addition to IgG, another protein with electrophoretic gamma-mobility which is never revealed by anti-normal human serum nor by anti-normal CSF. This protein, called B.T., has been found in 5 out of 8 CSF studied. The same immune serum against SSPE CSF reveals in normal brain extracts a specific protein sharing antigenic identity with the fraction B.T. from SSPE CSF. This brain protein is located in the white matter. It has not been found in the grey matter, nor in other tissue extracts. Due to its solubility and electrophoretic properties, this brain protein B.T. is not comparable with any of the brain specific proteins known at this time.

[Various aspects of lipid metabolism in the rat testicle after sulpiride treatment].

Testicular lipids were extracted from 20 rats which had received 40 mg sulpiride per kg per day for 20 days as well as from 20 controls. The extracts were weighed and chromatographed on thin layer in order to analyse them for the main fatty constituants : phospholipids and neutral fats. In addition total cholesterol, triglycerides and lipid phosphorus were determined. The total testicular lipid content and the phospholipids were unaltered. The distribution of phospholipids wasn't influenced either. Among the neutral fats the precursors of steroid hormones such as cholesterol and its esters didn't show any modification. The triglycerides on the other hand suffered a significant decrease which was partly compensated in situ by the increase of free fatty acids. Since plasma electrophoresis didn't reveal a parallel increase of the complex albumin--free fatty acids, the hypothesis of an in situ lipolysis is proposed. But it is not possible yet to attribute to sulpiride the role of a direct activation on a testicular triglyceride lipase nor that of an indirect activator by way of prolactine for example.

Gamma-globulins patterns in CSF of inflammatory neurological diseases in tropical Africa.

400 CSF electrophoreses were performed on Black West Africans (Ivory Coast) with inflammatory infections of the nervous system. In the normal state, the CSF of the Black African contains a higher concentration of gamma-globulins than the European (15.8% compared to 8--10%). This study permits us to confirm indirectly the classical idea of the extreme rarity of MS in tropical areas: an increase in the gamma-globulins of the CSF has been shown in 22% of the cases and the two principal responsible infections are trypanosomiasis (57.3%) and SSPE (16.8%). The oligoclonal distribution by various authors in trypanosomiases was not found. On the contrary, a nonclonal aspect of gamma-zone was constantly found (49/50). It is possible that the differences are in accordance with the different evolutionary phases of the disease, and that the oligoclonal distribution is a marker of the autoimmune state of the disease.

[Variation of the profile phenotype M of alpha-1-chymotrypsine : trial of interpretation (author's transl)].

Cross immunoelectrophoresis of serum A1AT (Pi M) shows "frequent" patterns with M2 less than M6 less than M4. Other "particular" patterns were found with M6 greater than M4 and M6 less than M2. The most often found patterns among the Negroid population in our study appeared dissimilar to that found among Caucasians. The "super-gene" concept and the variability of genetic flow may explain this difference. The "particular" patterns are often found in cord blood samples and in patients with hepatocarcinoma. They show similaritis between embryonic and cancerous processes. These different patterns also agree with the "super-gene" concept.

[Cerebrospinal fluid gamma globulins in neurological disorders affecting black Africans (author's transl)].

Four hundred C.S.F. electrophoreses were carried out in black West Africans (Ivory Coast) suffering from inflammatory disorders of the nervous system. In the normal state, the C.S.F. of the black African contains more gamma globulins than that of the white (15.8% as opposed to 10%). The study provides indirect evidence of the classical notion of the extreme rarity of DS (Disseminated sclerosis) in the tropics. Hypergammaglobulinorachia was demonstrated in 22% of cases, the two principal disorders responsible being trypanosomiasis (57.3%) and sub-acute sclerosing panencephalitis (16.8%). The oligoclonal distribution described in trypanosomiasis was not found. On the contrary, a homogeneous appearance of the gamma zone was constantly demonstrated (49/50). It is possible that these differences are related to the various phases in the course of the disease, and that oligoclonal disturbation is a marker of the auto-immune phase.

[Primary liver carcinoma: deficiency of alpha 1-antitrypsin].

A preliminary study involving 12 subjects suffering from carcinoma of the liver indicates the value of the estimation of alpha-1 antitrypsin and the determination of its phenotype by bidimensional electrophoresis. Alpha-1 antitrypsin was greater than 3.3 g/l in all cases, with a normal of 2.5 g/l. The MZ phenotype was found in 9 cases out of 12, the MM phenotype in the three recent cases and in particular in the only case of secondary carcinoma. In the Pi system, the Z trait would seem to be a genetic risk factor in primary carcinoma of the liver. Determination of the phenotype of alpha-1 antitrypsin may aid in its laboratory diagnosis.

[Inventory of oligoclonal fractions of subacute sclerosing panenephalitis cere-brospinal fluid in Africa with a specific immuneserum: first results (author's transl)].

The oligoclonal gamma-fractions in the cerebrospinal fluid (C.S.F.) from one case of subacute sclerosing panencephalitis (S.S.P.E.) have been studied by means of immunolectrophoresis. The gamma-zone consisted of IgG and free kappa and lambda chains. Besides that, a rabbit immune serum against S.S.P.E. C.S.F. has been prepared. This immune serum did reveal in the gamma-zone one additional protein which is different from IgG and normal tissue proteins. The possibility for this additionnal protein to be of myelinic origin is postulated, because of the demyelinisating nature of the lesion.

[Phosphlipids and neutral lipids of the testis of the monkey as a function of sexual maturity].

Biochemicals analysis of lipid parameters of testicular extracts of 14 monkeys (Cercopithecide) was found to confirm prior observations made with the same species by histochemical and histoenzymatic methods. Total lipids decrease with sexual activity; contrariwise total cholesterol increase; both parameters, however, showing higher levels in the absence of sexual activity. Phospholipids increase with sexual maturity after puberty but revert to normal with the cessation of spermatogenesis. Chromatographic separation reveals the specific variations of triglycerides the total being higher during pre-puberty and in animals lacking sexual potency. Phospholipid constituents are constant and may be classified in order of their concentration with lecithin being the major constituent. These observations though somewhat fragmentary and representing a rather small population will stimulate new studies and at the present time, agree with similar work done on other species of animals.

A new case of heavy mu-chain disease.

The tenth case of mu chain disease is described. The patient lives in Ivory Coast as in our previously reported case. He was not affected with chronic lymphocytic leukaemia and the main clinical feature was liver cirrhosis of unknown origin. The amount of abnormal protein in the serum was great enough to give an abnormal bond on the routine electrophoresis. The protein was devoid of light chains and was present in the form of disulfide linked polymers of incomplete mu chain. The molecular weight of the monomer was approximately 58,000. The protein comprised the Fc fragment and a part of the Fd segment. Bence Jones protein was not found in the urine.
