[Effect of clofelin and apressin on blood circulation and metabolic indices of the brain].

Experiments on cats were made to study the effects of clopheline (5-6 and 10 micrograms/kg) and apressin (1 mg/kg) on the time-course of changes in the cerebral circulation, total arterial and venous pressures, oxygen and glucose concentrations, pH in the arterial and venous blood. It was discovered that clopheline and apressin appreciably reduce the total blood pressure and minimize the volumetric velocity of the cerebral circulation. Brain oxygen and glucose consumption was found to be reduced under the effect of these drugs.

[Mechanism of action of reserpine on the blood circulation and oxygen regimen of the brain].

Resistography was used in experiments on anesthetized cats to show that reserpine produced biphasic (dilatation and constrictor) reactions on the part of the cerebral, extracranial and peripheral vessels. The dilatation reaction occurred immediately after the drug administration, it was intermittent and preserved after the blockade of the adreno- and cholinoreceptors. During electric stimulation of the cervical sympathetic ganglia, reserpine insignificantly reduced constrictor reactions of the vessels and inhibited depressor reactions. After intravenous injection of rausedyl the cerebral blood flow and oxygen consumption by the brain tissue decreased.

[Effect of clofelin and apressin on the autoregulation of cerebral blood flow].

Acute experiments on cats under mild sodium ethaminal anesthesia and local novocaine anesthesia were made to determine the autoregulative reactions (AR) of the cerebral vessels to different levels of arterial pressure (AP). After clofelin administration the autoregulation borders shifted to the right i. e. towards higher AP levels. Unlike clofelin, apressin made the AP borders shift to the left, i. e. towards lower AP levels. Possible mechanisms of the effects seen and their importance for medicine are discussed.

[Effect of pentoxyfylline and xanthinol nicotinate on the overall blood pressure and tonus of cerebral and peripheral vessels].

It has been demonstrated in cat experiments that pentoxyphylline (10 and 20 mg/kg intravenously, 1 mg/kg intraarterially) and xanthinole nicotinate (75 mg/kg intravenously) first produce dilatation on the part of the perfused vessels, with this dilatation progressing to the constriction in some of the experiments. As the dose of pentoxyphylline raised the arterial blood pressure, whereas after pentoxyphylline administration the phase alterations of AP were observed not infrequently: at first the pressure decreased and then rose as compared to the initial level.

[Effect of strophanthin and celanide on the autoregulation of the cerebral blood flow].

It has been demonstrated in acute experiments on anesthetized cats that the control group animals manifested (before glycoside administration) a comparatively steady brain circulation accompanied by changes in the arterial pressure (AP) within 60-150 mm Hg. Intravenous injection of strophanthine (0.05 mg/kg) and celanide (0.1 mg/kg) improved autoregulatory reaction of the brain vessels during AP falling to lower levels as compared to control.

[Effect of strophanthin and celanide on the tonus of the cerebral and peripheral vessels].

Resistography was used in acute experiments on anesthetized cats to demonstrate that cardiac glycosides, particularly celanide, often cause phasic (constrictive-dilative) responses of the perfused vessels and general arterial pressure. The constrictive response of the perfused vessels did not depend on the concentration of calcium ions in the blood and was not prevented by preliminary administration of dihydroergotoxin. However, glycosides prolonged the vasoconstrictive effect of noradrenaline. The vasodilative response to glycosides was reversed by atropine. It is assumed that the constrictive response is myotropic, whereas the dilative one is cholinergic in nature.

[Effect of clofeline and apressin on systemic arterial pressure and the tone of cerebral and peripheral vessels].

Resistography was used to demonstrate in acute experiments on anesthetized cats that intravenous injection of clofelin (2,6 and 10 microgram/kg) and apressin (1, 2.5 and 5 microgram/kg) induced a dose-dependent decline in the arterial blood pressure which was not infrequently preceded by short-lived hypertension. Clofelin induced an increase in the tone of cerebral vessels while apressin reduced it. As regards femoral vessels both the drugs produced diverse (sometimes phasic) reactions, with vasodilation being predominant.

[Effect of thiamine bromide on cerebral circulation and arterial pressure (an experimental and clinical study)].

The effect of thiamine-bromide (5-7 mg/kg, intravenously) on the cerebral blood flow circulation rate; total arterial pressure, oxygen tension in the brain tissues (measured polarographically) and pressure in the venous vessels of the brain was investigated on anesthetized cats with controlled respiration. In patients with hypertensive disease (essential hypertension) the action of the agent (1 ml of a 6% solution, intramuscularly) on the rheoencephalographic readings and overall arterial pressure was studied. Thiamine-bromide is shown to lower the tonicity of intra- and extracranial vessels and to increase pulsed variations in the volume of blood filling of the cranial vessels. The hypotensive effect of the drug is pronounced but mildly. The oxygen tension in cerebral vessels depended mainly on changes in the cerebral circulation rate.

[The effect of cytochrome c preparations on the cerebral circulation in cerebral ischemia].

Biotechnological cytochrome c, heme-tetradecapeptide (HTDP) and animal cytochrome c were studied for their effects on intact and brain ischemic rats. In the latter case, the compounds were administered before ischemia induction and 15 min after artery ligation. It was found that the cytochrome c preparations did not virtually affect the cerebral circulation in intact rats. In cerebral ischemia, the cytochrome C preparations increased circulation, showing their more profound effects in case of preadministration of the drugs. The dose-independent effects of HTDP may be associated with the higher transmembranous permeability and the saturation phenomenon of this compound.

[The effect of lithium derivatives of GABA on blood circulation and metabolic indices in the brain under ischemia].

In acute experiments on anesthetized cats with cerebral ischemia in conditions of autohemoperfusion of cerebral and peripheral vessels with a stable volume of blood we found that lithium oxybutirate and new GABA derivatives: LOS 1-84 and LOS 5-79 affect the cerebral blood flow and metabolism in the brain. Prophylactic intravenous injection of lithium oxybutirate did not affect the development of postischemic hyperperfusion but inhibited postischemic hyperperfusion. The compounds prevent the development of postischemic phenomena and promote the recovery of metabolism in the brain.

[The effect of cytochrome c preparations on the autoregulation of the cerebral blood flow in ischemia].

The effect of animal cytochrome C (Ca), biotechnological cytochrome (Cb) and its hemtetradecapeptide (HTDP) on cerebral blood flow autoregulation during rapid decrease of systemic arterial pressure (SAP) was studied in acute experiments on rats. Cytochrome C preparations caused no effect on the autoregulatory responses of the cerebral vessels in animals with normal cerebral circulation. Injection of 5 mg/kg Ca and Cb and 0.8 mg/kg HTDP promoted restoration of the phenomenon of cerebral blood flow autoregulation in ischemic brain damage in change of SAP from 120 to 60 mm Hg. Prophylactic injection of 20 mg/kg Ca and Cb and 3.3 mg/kg HTDP prevented cerebral blood flow autoregulation disturbance caused by transitory brain ischemia.

[Effect of diethyphen on cerebral vessel tonus and on general arterial pressure].

The investigation was carried out on anesthetized cats under controlled respiration. The tonicity of cranial vessels was judged about by the perfusion pressure figures in the system of intra- and extra-cranial vessels. With its intravenous and intracarotid administration diaethiphenum lowered the one of intra- and extra-cranial vessels. An intravenous introduction of its high doses was followed also by a fall of the total arterial pressure. An effect on the brain vasculature similar to that of diaethiphenum is also produced by diethylstibestrol.

[The effect of 3-hydroxypyridine derivatives on postischemic disorders in the autoregulatory reactions of the cerebral vessels].

Acute experiments on nembutal anesthetized rats showed improvement of cerebral blood flow autoregulation in the postischemic period under the effect of mexidol and the new 3-hydroxypyridine (3-HOP) derivatives LBK-10 and LBK-38 administered for prophylactic and therapeutic, purposes. The role of dopaminergic activity, solubility in lipids, and other factors in the mechanism of the activity of 3-HOP derivatives is analysed.
