[Characteristics of the pharmacological action of piracetam].

As concerns most generally accepted characteristics adopted in pharmacological studies of psychotropic drugs pyracetam is little active, has no effect on the EEG of the cortex and subcortical structures of the brain. At the same time, in a number of tests this drug displayed a well-marked influence on the central nervous system. It restrains convulsive activity provoked by application to the cerebral cortex of strychnine and penicillin solutions, reduces EEG disturbances produced by barbiturates poisoning and acute hypoxia of the brain. Pyracetam also facilitates the process of teaching in rats, improves the execution of a developed conditioned avoidance reaction, protects rats against manifestations of retrograde amnesia induced by an electric shock, applied after development of the conditioned reflex of passive avoidance.

[Effect of the antihistamine preparation, fencarol, and other quinuclidyldiarylcarbinols on tissue aminoxidase activity].

In experiments in vivo, the derivatives of quinuclidylarylcarbinol, possessing an antihistaminic activity, fencarol, its di(o-tolyl)- and di(o-methoxyphenyl)-derivatives activate histamine oxidative deamination by diaminoxidase of the rat lungs and increase the histamine tissue level. Quinuclidyl-3-dithienyl carbinol inhibits the activity of diaminoxidase, but decreases the histamine tissue level. On the contrary, dimedrol (diphenhydramine), pipolfen (promethazin), pyrilamine and cyproheptadin exerted no effect on the activity of diaminoxidase or the histamine tissue level. All the compounds studied did not alter oxidative deamination of the rat brain serotonin by monoamine oxidase or the serotonin content in brain tissues.

[Effect of the antihistamine preparation, phencarol, on the central nervous system].

The new antihistaminic and antiallergic drug phencarol (quinuclidy-3-diphenylcarbinol hydrochloride) in contradistinction to dimedrol (diphenhydramine) and diprazine (promethazin) exerts no pronounced depressant effect on the central nervous system. It does not significantly influence the period of action of hypnotic drugs, the effects produced by alcohol, spontaneous motor activity, summation capacity of the central nervous system, conditioned reflexes or electroencephalogram. That phencarol possesses no marked central action may be accounted for by its low lipophilic activity and little permeability of the blood brain barrier by this drug.

[Comparative effect of antiglaucoma preparations on brain bioelectrical activity].

Experiments on unanesthetized rabbits with electrodes implanted in different areas of the cortex and subcortical structures of the brain have demonstrated that when administered to the conjunctival sac of the eye, the drugs used as eye drops for glaucoma treatment affect brain bioelectrical activity and the ECG. Aceclidine, physostigmine and clofelin produce the most remarkable action. Phosphacol and pilocarpine are less active. Aceclidine and physostigmine effects are characterized by pronounced activation of the EEG. On the contrary, clofelin produces the synchronization of bioelectrical activity. The activation effect of aceclidine and physostigmine is reduced or blocked by conjunctival administration of atropine and benactizine.

[Effect of the tetracyclic antidepressants pyrazidol and inkazan on the adrenergic nerve transmission in the rat vas deferens].

The effect of new tetracyclic antidepressants pyrazidol and inkazan on adrenergic neurotransmission in the isolated rat vas deferens was studied by examining vas deferens contractions in response to the transmural electric stimulation of the postganglionic sympathetic nerves and addition of noradrenaline (NA) or BaCl2. Pyrazidol and inkazan were found to be capable to increase vas deferens contractions in response to transmural electric stimulation or addition of NA. When given in high concentrations, these antidepressants inhibit the adrenergic neurotransmission since inkazan possesses low sympatholytic activity and pyrazidol exhibits certain alpha-adrenolytic effect.
