Iron tissue storage and hemoglobin levels of deficient rats repleted with iron bound to the caseinophosphopeptide 1-25 of beta-casein.

Caseinophosphopeptides (CPP) issued from enzyme digestion of caseins bind cations and keep them soluble in the digestive tract. They could be used as ligands to improve iron (Fe) bioavailability. Fe-deficient young rats were repleted with Fe (40 or 200 mg/kg of diet) bound either to the beta-CN (1-25) of beta-casein or to whole beta-casein or as FeSO(4). A control pair-fed group was given 200 mg of Fe (FeSO(4))/kg of diet for 6 weeks. After repletion, hemoglobin concentration of the control group was reached only by the ) animals fed 200 mg of Fe/kg; beta-CN (1-25) bound Fe (40 and 200 mg) produced higher Fe liver and spleen stores than FeSO(4). Binding Fe to the whole, nonhydrolyzed beta-casein gave results intermediate between the other experimental groups. Binding Fe to phosphoserine residues of low molecular weight CPP improved its ability to cure anemia and to restore iron tissue stores, as compared to Fe bound to the whole casein and to inorganic salts.

Oct3/4-associating proteins from embryonal carcinoma and spermatogenic cells of mouse.

The octamer-binding protein Oct3/4 was postulated to active transcription through protein-protein interactions with hypothetical cellular coactivator(s). We have used a bacterially produced Oct3/4, as a protein-binding probe, to detect by far-Western assay the Oct3/4-associating proteins (OTAPs) from the embryonal carcinoma (EC) cells F9 and pachytene spermatocytes. Both common and cell-specific OTAPs were shown to interact directly with Oct3/4. Differentiation of the EC cells results in disappearance of most of OTAPs, supporting their coactivator nature. Several OTAPs detected in pachytene spermatocyte may represent germ cell-specific Oct3/4 coactivators.

Relationships between iron and zinc metabolism: predictive value of digestive absorption on tissue storage.

The responses of animals to intake of a trace element could vary if it is ingested with a single test meal or due to chronic intake. The metabolic relationships between zinc (Zn) and iron (Fe) were assessed in the young animal by comparing their digestive absorption studied at the beginning of the study with their tissue storage after two months of being fed on experimental diet. Diets supplied adequate intakes of Fe (45 and 300mg/kg diet) and Zn (14 and 45 mg/kg). A significant effect of Fe supply (p < 0.0001) but not of Zn was displayed on Fe absorption; both Fe and Zn diet concentrations influenced Zn absorption (p < 0.01, p < 0.0001). Fe and Zn organ contents significantly correlated with the amount absorbed during the metabolic balance (p < 0.0001). There was a positive correlation between liver, bone, and muscle Fe and Fe absorption (mg/d)(p < 0.0001), and Fe absorption and bone and muscle Zn (p < 0.04) and a negative one with liver Zn (p < 0.0001); a positive correlation was displayed between Zn absorption (mg/d) and Zn organ content (p < 0.0001). There was no correlation between Zn absorption and Fe tissue content (p > 0.05). This study suggests that interactions occur at every step of Fe and Zn metabolism; Fe is more efficient in altering Zn storage than the reverse. The organism seems to be unable to diminish the consequences of an unbalanced diet and digestive absorption. Care must be taken to give the young growing balanced diets.

Effect of small variations of aluminum intake on calcium metabolism in young rats.

BACKGROUND: While in the adult Al intoxication requires high dosages, little is known on the threshold level of Al toxicity in the young. METHODS: Weaning rats were fed for 90 days ore of four diets differing by their content in Ca (7.5 vs < 0.5 g/< g diet)(Ca+/-) and Al (10.6 vs 8.4 mg/kg)(Al+/-); Al supplementation was 30% above the standard level of diet. Ca and Al levels were measured in liver, bone (femur), and brain. RESULTS: Ca- had a significant negative effect on growth which was further reduced by Al+; in Ca sufficient/Al+ animals, Al concentrations were significantly increased in bone and brain and tended to increase in liver; Ca decreases observed in these three organs were only significant in brain. Ca deficiency further enhanced the Al deposit in bone at both levels of Al intakes, and reduced Ca concentrations in these three organs in Al+ animals; in Ca-/Al- animals, the decrease in Ca displayed in the three tissues reached a significant level in brain. CONCLUSIONS: This study suggests that in the growing subject the side effects of small variations of Al intake can be enhanced when they are combined with other mineral imbalances.

Chromium and parenteral nutrition in children.

Chromium (Cr) dosage was assayed in i.v. nutrition, serum, and losses of five children on total parenteral nutrition for > or = 4 weeks. The Cr supply (4.7 +/- 1.2 micrograms/kg/day) was above recommended levels (0.5 microgram/kg/day). Serum (18.2 +/- 1.8 micrograms/L) and urine (37.4 +/- 10.5 micrograms/L) were also higher than control values (0.7-0.9 microgram/L and 0.2-0.8 microgram/L, respectively). Serum and urine Cr concentrations displayed a positive correlation. Serum Cr and Fe showed a negative correlation. These results confirm the potential toxicity of Cr previously reported in animals. Cr levels of i.v. nutrition solutes should be checked thoroughly.

Comparison of two techniques used to collect normal and motile sperm.

To improve the quality of sperm for intra-uterine insemination (IUI) or in-vitro fertilization (IVF) swim-up (SU) and migration-sedimentation (MS) methods were compared, for the selection of morphologically normal and motile sperm. In 74 patients consulting for couple infertility, it was shown that MS gave a better yield of motile sperm than did SU. An improvement in morphology and motility was achieved, especially in asthenospermia (motility less than 40%) and/or teratospermia (normal shape less than 40%). The percentage of midpiece and tail abnormalities was lowered by the MS technique. It is proposed that the MS method be used for IUI and IVF, adapting the number of tubes to the initial sperm concentration.

[Plasma hemoglobin. Comparison between 2 methods of determination].

The methods used to titrate plasmatic haemoglobin, an analysis which is of great interest to cardiac surgery, must be very accurate because the levels are much lower than those found in whole blood. This work introduces a comparison between two methods: the ammoniacal water method and Cripps's differential method. After studying various criteria (repeatability, limit of detection, accuracy, reference values, stability of the reaction), and after studying the correlation between each method and the benzidin method, used as reference method, as well as the correlation between all of them, Cripp's method turns out to be the closest to the benzidin method, as well as being simpler, more accurate, more specific, and also easier to use than the ammoniacal water method, the only problem occurring at the stage of the preparation of the standard solution.

[Blood levels of molybdenum, selenium and copper at delivery].

Molybdenum (Mo), selenium (Se) and copper (Cu) plasma levels were studied at delivery in 38 women (1 twin pregnancy). The influence of parity (P) (2.5 +/- 1.7) gestational age (GA) (38.6 +/- 3.2 weeks), birthweight (BW) (3,120 +/- 695 g) and Apgar score upon these levels were assessed. Mother (M) and cord (C) blood values were 15.0 +/- 7.8 and 15.0 +/- 9.3 nmol/l respectively for Mo, 0.54 +/- 0.12 and 0.47 +/- 014 mumol/l for Se and 27.6 +/- 5.9 and 5.5 +/- 3.3 mumol/l for Cu. C and M values for Se and Cu were statistically different (alpha less than 0.02, alpha less than 0.001). There was a significant correlation between C and M for Mo levels (r = 0.41; p = 1.9%) and Cu levels (r = 0.47; p = 0.35%), between Cu C levels and GA (r = 0.54; p = 0.6%) and between Se, C levels and BW (r = 0.66; p = 1.9%). There was no correlation between the levels of the trace elements studied. This study shows that fetal levels of Mo vary with those of the mothers. On the other hand, the levels of Se in the fetus seem to be protected from variations observed in the mothers. The antagonism between Cu and Mo which had been previously demonstrated in the fetal liver was not found in blood.

Molybdenum content of term and preterm human milk during the first 2 months of lactation.

To assess the molybdenum supply and requirements of preterm infants, Mo concentration was determined in milk from mothers of 6 term and 11 preterm newborns; no difference was found between fore- and hindmilk and no diurnal variations were found during 24-h collections. Respective values (means +/- SD) of term and preterm milks were 10.2 +/- 3.7 and 4.0 +/- 3.7 micrograms/L (106.2 +/- 38.5 and 41.7 +/- 38.5 nmol/L) at 3-5 d of lactation, 4.8 +/- 3.9 and 3.7 +/- 3.8 micrograms/L (50.0 +/- 4.6 and 38.5 +/- 39.6 nmol/L) at 7-10 d, 1.5 +/- 1.4 and 1.4 +/- 0.9 micrograms/L (15.6 +/- 14.6 and 14.6 +/- 9.6 nmol/L) at 14 d, 2.6 +/- 2.2 and 1.9 +/- 1.4 micrograms/L (27.1 +/- 22.9 and 19.8 +/- 14.6 nmol/L) at 1 mo, and 0.2 and 1.2 +/- 0.5 micrograms/L (2.1 and 12.5 +/- 5.2 nmol/L) at 2 mo. A statistical difference was found between term and preterm milk at 3-5 d of lactation. During lactation significant changes were found between the periods 3-5 d and 7-10 d, 14 d, 1 mo (p less than 0.01) and 2 mo (p less than 0.05) of lactation and between 7-10 d and 14 d (p less than 0.05). According to the requirements of the preterm infant, a supplementation of 2-3 micrograms.kg-1.d-1 by enteral route is suggested.

[Study of an optimal biological profile for demonstrating pancreatic involvement in acute abdominal syndrome].

The authors have studied several seric, plasmatic and urinary constituents in patients hospitalized for an acute abdominal syndrome to be able to characterize an eventual pancreatic lesion; mainly seric and urinary amylase as well as its isoenzymes, lipase, liver profile and trypsin. In acute pancreatitis, the means of the maximal increases of seric amylase, lipase and trypsin are respectively: 10.7; 21.6 and 19.2 X N (upper normal limit) whereas in chronic pancreatitis, these elevations are 6.5 X N for amylase and 9.5 XN for lipase. The authors observed at J1 (first day of hospitalisation) and at J2 an increase in seric amylase, lipase and/or liver profile respectively in 95, 90 and 25 p. cent of acute pancreatitis; in 86, 86 and 14 p. cent of chronic pancreatitis and 43, 39 and 86 p. cent of bili duct diseases. In conclusion, it appears compulsory to run a liver profile with the pancreatic enzymes (amylase and lipase) to diagnose a pancreatitis in presence of an acute abdominal syndrome.

Isomers of broparoestrol and antiestrogen action: comparison with tamoxifen.

This study compares the relative biological potencies of a known antiestrogen tamoxifen to two triarylethylene compounds which have been shown previously to be potent inhibitors of rodent mammary tumorigenesis. Based on a) uterotrophic and anti-uterotrophic tests, b) indexes of cellularity, and c) protein content, these studies indicate that the trans, as well as the cis, isomers of bromotriphenylethylene are partial estrogen antagonists with no estrogenic effects in rat uteri and partial agonists in mouse uteri, whereas tamoxifen shows partial antiestrogenic/estrogenic effects in rats and is fully estrogenic in mice.

[Increase in the number of Kurloff cells following estrogen stimulation in guinea pigs: demonstrated latency period].

The comparative study between plasma estradiol levels and KURLOFF cell numbers in the blood and some organs of adult, non pregnant female guinea-pigs, leads us to describe a biphasic response of the KURLOFF cells to estrogen stimulation. The first step, a five day lag-phase, is followed by a secondary increase of KURLOFF cell numbers with a peculiarly noticeable storage in the thymus parenchyma.

Androgen binding protein, sperm production, semen output and LH and testosterone profiles in young postpubertal bulls.

An androgen binding protein (ABP), which binds 5alpha-dihydrotestosterone with high affinity (Ka=0.3x10(9) M(-1)), has been demonstrated in testicular and epididymal cytosols of 5 young post pubertal bulls (15-17 months old) of the Montbeliarde dairy breed. Simultaneously, daily sperm production (DSP), semen output and plasma LH and testosterone concentrations (from frequent samplings) were determined. ABP levels were 21 fmoles/mg protein in testis and 59, 22 and 43 fmoles/mg, respectively, in caput, corpus and cauda epididymis. Mean DSP, per gram of testis, was 16.6x10(6) spermatozoa, and the mean sperm output was approximately 1.5x10(9) spermatozoa per ejaculate. Mean LH and testosterone levels were 1.5 ng/ml and 2.1 ng/ml, respectively. One bull (882) was clearly distinguishable from the others, in showing higher ABP and testosterone levels together with a lower daily sperm production. Results of this study may (1) suggest a physiological role of ABP in sperm epididymal maturation and (2) give a new parameter in the evaluation of individual bulls testicular function.

Inhibition of prolactin-induced mammary cancer in C3Hf (XVII) mice with the trans isomer of bromotriphenylethylene.

C3Hf (XVII) mice never develop spontaneous mammary tumors. However, the transplantation of an isologous pituitary gland under their kidney capsule is followed by a 10-fold increase in serum and pituitary prolactin content (180 ng/ml and 20 micrograms/mg of tissue, respectively), concomitant with an increase of prolactin receptors in mammary glands. Under these conditions, mammary tumors appear in 90% of the mice. If a racemic brominated triphenylethylene, i.e., broparestrol, is administered, serum and pituitary prolactin decrease rapidly (10 ng/ml and 4 micrograms/mg of tissue, respectively), and prolactin receptors in the mammary gland are markedly reduced. This compound also inhibits the development of normal mammary glands, prevents mammary carcinogenesis, and unexpectedly causes a significant atrophy of the ovaries. Our study confirms that prolactin is a key hormone involved in murine mammary carcinogenesis and that it can act directly on the mammary gland by stimulaing the level of its own receptor.
