Effects of gonadotrophins on the proliferative activity of somatic testicular cells in neonatal and prepubertal rats.

The proliferative activity of the different testicular somatic cell lineages was studied in neonatal and prepubertal rats after treatment with recombinant FSH (recFSH) and human chorionic gonadotrophin (hCG). Male rats were treated on days 5-9 or 15-19 with 1 or 3 IU/rat.day, respectively, of recFSH, 10 IU/rat.day of hCG or vehicle and sacrificed the day following the last injection. Proliferation was studied by the immunohistochemical demonstration of DNA-incorporated 5-bromodeoxyuridine (BrdU) in both paraffin- and Epon-embedded tissues. No differences existed for the labeling indices in paraffin- or Epon-embedded tissues, but the identification of some cell types were easier in these latter one. At 10 days of age, no significant differences were found for the proliferative activity of the different cell types, except for the labeling index of foetal Leydig cells that was increased in hCG-treated animals. At 20 days of age, treatment with recFSH increased the testicular weight and tubule diameter, as well as the size of adult type Leydig cells and the labeling index of macrophages. Treatment with hCG increased the testicular weight and the number and size of adult-type Leydig cells, as well as the labeling indices of mesenchymal, adult-type Leydig cells and macrophages. These results indicate that the effects of gonadotrophin treatment on the different somatic testicular cell types are dependent on both the type of cells and their developmental stage.

[Inflammatory pseudotumor of the spleen].

A case of inflammatory pseudotumor of the spleen in a 69-year-old woman who was studied for elevated globular sedimentation rate is presented. The lesion was observed on echography as a mixed solid-cystic mass. Magnetic resonance showed a slight decrease in signal intensity in potentiated T1 sequences and a heterogenic mass with high signal areas in T2 images and protonic density. Splenectomy was performed and histopathologic study demonstrated an inflammatory pseudotumor of 10 cm in its greatest size. Two year follow up was satisfactory. On review of the literature only 34 cases of inflammatory pseudotumors have been previously published. Half of the cases presented during the 6th and 7th decades of life, with women being the most frequently affected. Inflammatory pseudotumors of the spleen are benign lesions which should be considered in the differential diagnosis of splenic masses. Since diagnosis cannot be made preoperatively, and a malignant tumor is almost always suspected, splenectomy should be performed.

[Cancer of the prostate: evaluation of the expression of the mutated protein of the 53 tumor suppressor gene].

UNLABELLED: Prostate cancer is the most common tumour in men, and is most frequently established late in life. The objective of the article is to determine the protein expression of gene p53 in prostate cancer and to correlate the findings with clinical, morphological and epidemiological parameters. MATERIAL AND METHODS: Immunohistochemical techniques with monoclonal antibody used to study the gene p53 expression. The study included 35 prostate carcinoma: 4 cases in stage A; 5 cases in stage B; 12 in stage C and 14 in stage D. The control group consisted in 10 benign prostate hyperplasia. Average age was 66.8 (SD +/- 6.7 years). RESULTS: Positive staining for gene p53 protein was seen in 11 of the 35 cases (31.4%). All stage A and B tumours were negative; however, stage C and D tumours were positive in 16 and 64% cases respectively. In the control group all cases were gene p53 negative. No statistical relationship was seen between Gleason Index and positive results for p53. There was a significant difference between gene 53 expression and clinical stage. CONCLUSIONS: Results show that the immunohistochemical expression of gen p53 mutated protein keeps no relation to Gleason's rating index and is seen with significantly higher frequency in advanced, primarily stage D tumours.

Prevalence of skin reactions to aeroallergens in asthmatics of Puerto Rico.

In Puerto Rico, although a high prevalence of asthma has been reported, the sensitization rates to aeroallergens in these patients is unknown. The purpose of this study using a case control design, was to determine and compare the rates of sensitization to common aeroallergens in an asthmatic population of 576 asthmatics and 144 healthy controls. A skin prick test was conducted using standardized extracts of Dermatophagoides farinae (Df) and D. pteronyssinus (Dp), house dust (HD), cat hair and epithelium (CT), dog hair and dander (DG), grass pollen mix (PG), tree pollen mix (PT), weed pollen mix (PW), Aspergillus mix (AM), mold mixes A (MA) and B (MB), Periplaneta americana (PA) and Alternaria-Hormodendrum mix (AH). In addition, an extract from the domestic mite Blomia tropicalis (Bt) was also used. A wheal > or = 3 mm2 was considered a positive reaction. In addition, a standardized questionnaire was administered and a preliminary domestic mite identification survey was conducted. The analysis of the data showed that 85.8% of the asthmatics had at least one positive reaction and 61.6% of them had positive skin reactions to atleast one mite species. Asthmatics reacted to domestic mites 6.19 times more than the control group (p < 0.0001) and was the largest significant difference found in this study for any allergen tested. Preliminary identification of the acarologic fauna in southern Puerto Rico demonstrated that Blomia tropicalis, Dermatophagoides pteronyssinus, D. farinae and E. maynei are the dominant domestic mite species found in homes of asthmatic individuals. These results demonstrate that domestic mites are a very important source of sensitizing aeroallergens for asthmatic patients in Puerto Rico. Based upon the mite survey, Blomia tropicalis plays an important role in allergic sensitization, in addition to Dermatophagoides pteronyssinus and D. farinae. The skin prevalence to pollens and to molds may not reflect the true prevalence of sensitization to these allergens. Pollen identification and counts, and a survey of microflora of Puerto Rico are needed in order to identify and validate important allergens that eventually could be incorporated into a more appropriate panel for testing sensitization in susceptible individuals.

Evaluation of CHROMagar Candida medium for the isolation and presumptive identification of species of Candida of clinical importance.

CHROMagar candida is a new differential culture medium that allows the isolation and presumptive identification of species of yeast of clinical importance. We tested 618 strains of yeast, including 128 direct isolates from clinical specimens. After two days of incubation at 37 degrees C, 339 of 341 Candida albicans, 98 of 99 Candida glabrata, all the Candida tropicalis, and all the Candida krusei were identified correctly. The sensitivity and specificity in these cases were both superior to 99%. Easy to prepare, with low cost, CHROMagar Candida proves to be a useful medium for the identification of species of yeast that are isolated with greater frequency in clinical material and for the identification of mixed cultures.

Laboratory determinations in Anisakis simplex allergy.

BACKGROUND: Anaphylactic reactions caused by the fish nematode, Anisakis simplex, after ingestion of parasitized fish, have been described. OBJECTIVE: This study was undertaken to confirm, by histamine release tests, that A. simplex is able to trigger IgE-mediated reactions and to describe the serologic profiles in this sensitization. METHODS: Twelve patients who had anaphylactic symptoms after ingestion of cooked fish and positive prick test results and determinations of IgE to A. simplex were studied by indirect IgG ELISA and IgG and IgE immunoblotting. Sera from subjects parasitized with other nematodes, patients with fish allergy, and healthy donors were included as controls. A histamine release test was performed in a representative case. RESULTS: IgE immunoblotting was a specific test to detect A. simplex allergy. IgE-reacting bands were found in serum samples from 11 of our patients. Specific IgG antibodies were found by ELISA and immunoblotting, but this response was less specific. Histamine release was positive with A. simplex extract and negative with fish. CONCLUSION: A specific and intense immune response to an A. simplex extract was found in our patients. A. simplex is able to elicit anaphylactic reactions, and A. simplex allergy should be suspected in patients with allergic symptoms after ingestion of fish. A positive prick test response to A. simplex and a negative response to fish is a good indication for a diagnosis of A. simplex allergy.

Magnification and visual acuity in highly myopic phakic eyes corrected with an anterior chamber intraocular lens versus by other methods.

PURPOSE: To calculate retinal magnification and visual acuity in highly myopic phakic eyes corrected with anterior chamber intraocular lenses (IOLs) versus correction by other methods. SETTING: Laboratory of Optics, University of Alicante, Alicante, Spain. METHODS: Relative retinal magnification (RRM) was used to study changes in retinal image size induced by different correcting systems. Relative spectacle magnification (RSM) was used to compare these systems with the emmetropic eye. The matrix method was used to calculate RRM, RSM, and visual acuity in 12 theoretical cases of myopia in eyes with axial lengths from 27.0 to 33.0 mm and corneal powers from 42.00 to 48.00 diopters. RESULTS: The RRM, RSM, and visual acuity increased as corneal power increased for a myopic eye of fixed length. Changing the plane of correction from the spectacle or cornea to the intraocular increased retinal magnification, which increased visual acuity. CONCLUSION: Visual acuity in myopic eyes corrected with an IOL will be better than that in myopic eyes corrected by other means because of the retinal magnification induced by the IOL, which increases visual acuity one line or more.

Validation of an automatic urea analyser used in the continuous monitoring of hemodialysis parameters.

The validation of an automatic urea analyser used in the monitoring of hemodialysis processes is reported. The analyser can indirectly determine dialysis parameters as dialysis delivery (KT/V) and protein catabolism (PCRn). These parameters are useful for the prescription and optimization of hemodialysis. The analyser, based on a previously-reported flow-injection analytical biosystem, was connected on-line to the effluent of a dialysis machine during several hemodialysis sessions. The urea concentration data were continuously processed and dialysis parameters were obtained in quasi real time by means of the integration of an adjusted time-dependent exponential function. These values were compared with those obtained by applying the methods traditionally employed in hospital laboratories. The evaluation comprised 24 data sets from several patients of different gender and age. No significant differences were found between the KT/V and PCRn results obtained with the usual method and those results produced by the analyser proposed here.

Heterogeneity in the haemagglutinin gene and emergence of the highly pathogenic phenotype among recent H5N2 avian influenza viruses from Mexico.

Molecular changes in the haemagglutinin (HA)-coding regions and proteolytic cleavage sites from multiple H5N2 subtype viruses isolated during a recent outbreak of avian influenza (AI) in central Mexico have been characterized. Eighteen isolates, collected during a 15 month period (October 1993 to January 1995) from six central states, were sequenced. None of the 18 predicted HA1 amino acid sequences were identical and changes were not restricted to a specific region of the sequence. Phylogenetic analyses of the HA1 sequences demonstrated two virus lineages, designated Puebla and Jalisco, with sequence variation as high as 10.5 percent for amino acid and 6.2 percent for nucleotide sequences. During the latter months of the surveillance period, highly pathogenic (HP) strains of AI emerged causing lethal disease in commercial poultry flocks. In each of the HP strains isolated, the HA protein was cleaved in chicken embryo fibroblast cells in the absence of trypsin, and two alterations not found in earlier non-HP isolates were detected. In the HA protein, HP strains all had a glutamic acid --> lysine substitution at amino acid position 324 and an insertion of arginine and lysine as new residues 325 and 326. The insertion appears to be due to a duplication of the nucleotide sequence AAAGAA at nucleotide positions 965-970 of the HA1-coding region. Computer-assisted secondary structure analyses place the target for the insertion in a predicted RNA stem-loop structure. A mechanism is suggested by which the polymerase duplicates the sequence.

Prostaglandin H-synthase-2 is the main enzyme involved in the biosynthesis of octadecanoids from linoleic acid in human dermal fibroblasts stimulated with interleukin-1beta.

This study was focused on the characterization of the metabolism of linoleic acid by human dermal fibroblasts and the effect of interleukin-1 on the biosynthesis of octadecanoids. Dermal fibroblasts untreated and treated with recombinant IL-1beta were incubated with exogenous labeled linoleic acid. A combination of high performance liquid chromatography and gas chromatography-mass spectrometry was used as the analytic technique. We found that dermal fibroblasts convert linoleic acid mainly into 13-hydroxy-9-cis,11-trans-octadecadienoic acid (13-HODE) and 9-hydroxy-10-trans,12-cis-octadecadienoic acid (9-HODE), 13(S)-HODE and 9(R)-HODE being the predominant enantiomers. IL-1beta increased the formation of both 13-HODE and 9-HODE in a concentration-dependent manner with similar EC50 values as for prostanoid formation. This effect of IL-1beta on HODEs formation was concomitant with the expression of prostaglandin H-synthase-2. Formation of octadecanoids was inhibited in a concentration-dependent manner by acetylsalicylic acid and indomethacin. Dexamethasone, actinomycin D, and cycloheximide abolished the effect of IL-1beta on HODEs biosynthesis. Octadecanoid biosynthetic activity was associated with the microsomal fraction. Dermal fibroblasts incorporated [14C]-9-HODE and [14C]-13-HODE into phospholipids, mainly into phosphatidylcholine. IL-1beta increased significantly the esterification of 13-HODE in all glycerophospholipids, the major increase being observed in phosphatidylinositol. These results indicate that prostaglandin H-synthase-2 is the enzyme responsible for the increase in the ability to form HODEs of dermal fibroblasts stimulated with IL-1beta.

Significantly reduced expression of the proteoglycan decorin in Alzheimer's disease fibroblasts.

Aims-To investigate whether proteoglycan synthesis is altered in skin fibroblasts in patients with Alzheimer's disease compared with normal subjects.Methods-Cell lines obtained from donors with Alzheimer's disease and healthy controls were incubated with radioactive sulphate. The proteoglycans synthesised were determined and analysed by chromatographic, sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and glycosaminoglycans-lyase treatment. The amount of decorin synthesised by each cell line was quantified using western blot analysis. Transcripts for human decorin were determined using northern blot analysis.Results-No significant changes in total sulphate incorporation and glycos-aminoglycan (GAG) composition were detected in the incubation media of these cells. However, chromatographic and SDS-PAGE analysis of the proteoglycans secreted by the cell lines showed that a dermatan sulphate proteoglycan of 150-125 kilodaltons was substantially reduced in Alzheimer's disease fibroblasts. The molecular characteristics of this proteoglycan correspond to decorin. Western blot analysis indicated that decorin was reduced in Alzheimer's disease incubation medium compared with normal medium. Northern blotting indicated that in Alzheimer's disease fibroblasts decorin transcripts were significantly reduced compared with normal fibroblasts. Glypican concentrations, a cell surface heparan sulphate proteoglycan, remained the same.Conclusions-These results strongly suggest that the expression and synthesis of decorin is affected in Alzheimer's disease skin fibroblasts.

Use of species-specific oligonucleotide probes to detect Mycoplasma gallisepticum, M. synoviae, and M. iowae PCR amplification products.

Three digoxigenin-labeled oligonucleotide probes, complementary to the variable region of the 16S ribosomal RNA (rRNA) gene of Mycoplasma gallisepticum, M. synoviae, and M. iowae were designed. The oligonucleotides were used in a dot blot hybridization assay. The target DNA is a 780-bp fragment of the 16S rRNA gene of avian mycoplasmas amplified by a single set of primers (multispecies polymerase chain reaction [PCR]). The oligonucleotide probes were specific for their corresponding PCR products at hybridization conditions of 56 C and 50% formamide. The detection limit of the dot blot hybridization assay was approximately 70, 50, and 30 colony-forming units for M. gallisepticum, M. synoviae, and M. iowae, respectively, per 4 microliters of PCR. In general, the oligonucleotide probe dot blotting assay was a more sensitive and effective method of detecting PCR products than detection by gel electrophoresis.

Detection of nitric oxide synthase (NOS) in somatostatin-producing cells of human and murine stomach and pancreas.

The aim of this study was to identify by immunocytochemistry the distribution of nitric oxide synthase (NOS) in human and murine gastric epithelium. Using two different antisera specific for neuronal NOS (nNOS), we detected nNOS immunoreactivity in endocrine cells of the epithelium of the body and pyloric regions as well as in ganglion cells of the intrinsic plexi of the stomach of the three species studied. Both immunocytochemistry of contiguous sections and double immunolabeling methods showed that the nNOS-immunoreactive cells were also immunoreactive for somatostatin. Co-localization of nNOS and somatostatin has also been found in the pancreatic islets, where strong nNOS immunoreactivity appeared in scattered cells, which were peripheral in rat and mouse islets and more randomly distributed in human. The possibility of crossreactivity between the antisera against nNOS and somatostatin was ruled out by means of absorption controls. Immunocytochemical techniques were also applied to thin sections, confirming the immunostaining of gastric D-cells, which was restricted principally to the secretory granules. The possible functional implications of these findings for gastric and pancreatic physiology are discussed.

In vivo manipulation (depletion versus activation) of testicular macrophages: central and local effects.

Testicular macrophages are a relevant cell type for the regulation of Leydig cell steroidogenesis. The availability of liposome technology allows in vivo manipulation of macrophages in order to analyze their role in the regulation of the hypothalamic-pituitary-testicular axis. In this study, adult (70 days of age) and prepubertal (22 days of age) rats were injected intratesticularly with liposomes containing either dichloromethylene diphosphonate (C12MDP) to deplete testicular macrophages or muramyl tripeptide (MTP-PE) to activate them. Control rats were injected with the corresponding volumes of 0.9% NaCl. Animals were killed 10 days after treatment. Adult rats injected bilaterally or unilaterally with C12MDP liposomes showed increased serum LH and testosterone concentrations, as well as increased testosterone concentrations in the testicular interstitial fluid. In unilaterally injected rats, testosterone concentrations in the interstitial fluid were higher in the macrophage-containing testes than in the contralateral, macrophage-depleted testes. Adult rats treated bilaterally with MTP-PE liposomes showed increased numbers of testicular macrophages, whereas the number of Leydig cells was unchanged. Serum LH concentrations were decreased, but no changes were found in testosterone concentrations. Prepubertal rats treated bilaterally with C12MDP liposomes showed decreased numbers of Leydig cells. However, serum LH and testosterone concentrations were increased. Otherwise, prepubertal rats treated bilaterally with MTP-PE liposomes showed increased numbers of macrophages and Leydig cells, as well as increased serum testosterone concentrations. These data suggest that testicular macrophage-derived factors act at two different levels in the pituitary-testicular axis: first, at a central level by inhibiting LH secretion, and secondly, at a local level by stimulating Leydig cell steroidogenesis.
