[Establishment of a new human ovarian serous cystadenocarcinoma cell line (IM), and influence on cell proliferations by cisplatin with or without hyperthermia].

A new cell line, designated IM has been established from operation material derived from a woman with ovarian serous cystadenocarcinoma. The population doubling time of the 35th passage IM cell was 28.8 hours. And it was successively subcultured 165 times in over 7 years, moreover still kept CA125 production. The nuclear DNA and cell surface CA125 antigen were double stained by propidium iodide and anti CA125 monoclonal antibody-FITC. Then the two color cytogram obtained by flow cytometry was drawn up. For the most part of CA125 positive cell retained G0 + G1 of cell cycle, the lesser part was in G2 + M phase. The S phase rate of IM cell incubated with cisplatin at 37 degrees C or 41 degrees C for 1 hour that estimated by BrdU-propidium iodide double stain method of flow cytometry, it was suggested that the inhibition of DNA synthesis by cisplatin was increased with 41 degrees C low hyperthermia.

Correlation analysis of blood pressure and platelet aggregation/vascular reactivity in SHRSP, WKY, F1 and backcross generations.

1. Hypofunction of stroke-prone spontaneously hypertensive rat (SHRSP) platelets at developmental ages of hypertension is due to the defective protein (p47) phosphorylation which is mediated by protein kinase C. This study was undertaken to examine the genetic association of platelet functions and vascular reactivity with hypertension using male WKY, SHRSP, F1 and backcross generations at 16-18 weeks of age. 2. The distribution of blood pressure was continuous in each generation. 3. Contraction of mesenteric vascular bed with norepinephrine was positively correlated with blood pressure in the five generations (r = 0.77, n = 128). 4. Thrombin-induced platelet aggregation was inversely correlated with blood pressure (r = -0.87, n = 127). 5. The distribution of platelet aggregation and contraction was continuous in each generation, and backcross generations were not likely to have 1:1 segregation. 6. These results suggest the possibility that platelet hypoaggregability and peripheral vascular resistance are due to the pleiotropic effect of hypertensive genes, or that genes controlling these three characters are closely linked each other.

Blood pressure-related changes of endothelium-dependent relaxation in the aorta from SHRSP at developmental ages of hypertension.

1. Shear stress and flow influence endothelial functions to stimulate the release of endothelium-derived relaxing factor (EDRF). However, it is not well defined how hypertension affects endothelial functions. In this study, blood pressure-related changes of endothelium-dependent relaxation (EDR) in the aorta were examined in SHRSP, WKY and F1 hybrid at developmental ages of hypertension, and hypotensively-treated SHRSP. 2. Acetylcholine-induced EDR of aortic rings was significantly enhanced in 8 week old SHRSP compared with that of age-matched WKY. 3. NG-monomethyl L-arginine (L-NMMA) (1 mmol/L), an inhibitor of nitric oxide, greatly reduced the relaxation in both strains. Indomethacin (10(-5) mol/L), a cyclo-oxygenase inhibitor, did not affect the relaxation at this age. 4. EDR was positively correlated (r = 0.81, n = 22) with blood pressure at 9 weeks of age in WKY, SHRSP and F1 hybrid. 5. A five week hypotensive treatment of SHRSP caused a significant reduction in EDR with decreasing blood pressure. 6. It was concluded that the endothelium releases more EDRF in response to increasing blood pressure to regulate vascular tone at developmental ages of hypertension.

[The use of polymerase chain reaction method for the detection of Rickettsia tsutsugamushi in wild rodents].

We studied the applicability of polymerase chain reaction (PCR) method for the detection of R. tsutsugamushi in wild rodents. The PCR method which amplified the gene coding for the group-specific antigen of R. tsutsugamushi was used in this study. Specific PCR products (88 bp) were obtained with the DNAs from three reference strains (Gilliam, Karp, and Kato) and two cell culture adapted field isolates (KN-1 and GJ-1). The minimum number detectable by the PCR method was estimated to be 1.3 copies of rickettsial genome. In a study with experimentally infected mice, the PCR method could detect rickettsial DNA in one of two infected mice at four months after inoculation. Thereafter, fifty five wild rodents were captured in five areas of Okayama Prefecture, and R. tsutsugamushi DNA was detected, by the PCR method, by amplifying DNA from the spleen of each rodent. The rickettsia was also isolated from the same rodents by the mouse inoculation method. By the PCR method, rickettsia DNAs could be detected in 12 of 13 rodents from which the rickettsiae were isolated, and in 10 of 42 rodents from which no rickettsiae were isolated. These findings indicate that the PCR method is a simple and specific procedure to detect R. tsutsugamushi in wild rodents. On the other hand, the results of the PCR method demonstrated that the middle area of Okayama Prefecture was highly (44-81%) contaminated with R. tsutsugamushi.

[Clinical assessment for prognostic factors related to recurrence and progression in superficial bladder cancer].

PURPOSE: The objective of this assessment is to identify prognostic factors of recurrence and disease progression of primary superficial transitional cell carcinoma of the bladder. PATIENTS AND METHODS: TUR-Bt was performed in 150 patients with initial superficial bladder cancer, of which pathological diagnosis was pTa or pT1 transitional cell carcinoma. The recurrence and progression free survival was examined. The clinicopathological factors analyzed were as follows: grade, pT, tumor size, tumor number and tumor form. RESULTS: Median follow-up period was 39 months (range 3-184). The age distribution was from 25 to 98 years old with the average of 67. The patients were 122 males and 28 females. Recurrence was observed in 72 patients. The 5-year recurrence free rate for all cases were 41.6%. The factors found to be of significance for the prognosis for recurrence were tumor size (1 cm <) and tumor multiplicity (p < 0.01). Progression was seen in 15 patients; 8 invasive tumors, 2 lymph node metastases and 5 distant metastases. Twelve cases of those died of cancer. The 5-year progression free rate for all cases were 87.0%. The prognostic factors related to progression found in this study were G3, pT1 and sessile form (p < 0.01). There was no significant difference in recurrence free rate between cases with and without postoperative therapy; UFT p.o., ADM i.i. (intravesical instillation), ADM + Ara-C i.i. and PEP + 5-FU + Ara-C i.i. CONCLUSION: Tumor size more than 1 cm and tumor multiplicity were the high risk factors about recurrence. So were G3, pT1 and sessile form about progression.

[Carcinoma of the renal pelvis and ureter following bladder carcinoma].

We followed 135 primary bladder carcinoma patients for at least 3 years. Subsequent carcinomas of the upper urinary tract were found in 5 patients (3.7 per cent) an average of 67 months after an initial treatment of the bladder tumors. Two patients underwent radical cystectomy and the remaining 3 patients received transurethral resections or partial cystectomy five to seven times for bladder lesions. Primary bladder tumor was multiple in all and one of them was accompanied by carcinoma in situ in the bladder and urethra. Except for one patient who presented with gross hematuria, four patients had no symptoms referable to the upper urinary tract tumor. However two of them had high stage disease. Positive urinary cytology was observed in only one patient. All patients underwent nephroureterectomy and the four got well but one died of acute heart failure. Regular urinary cytology and IVP should be done for an extended period of time for early detection of renal pelvic and ureter cancers in patients who had multiple and recurrent bladder cancers.

N-nitroso compounds in two nitrosated food products in southwest Korea.

Gastric cancer is the commonest malignant neoplasm in Southwest Korea. The possibility of carcinogenic dietary factors led to the investigation of exposure to N-nitroso compound precursors among residents of the City of Chonju and of two outlying rural townships in North Cholla Province. Two traditional and widely consumed home-prepared food products, salted pickled cabbage (kimchi) and salted seafood sauce (chut-kal) were analysed (a) for nitrite, nitrate, total secondary amines and pH in these food products prior to nitrite incubation and (b) for volatile nitrosamines and total N-nitroso compounds before and after incubation with nitrite in simulated human stomach conditions. Nitrate levels were significantly higher in kimchi (median 1550 mg/kg) than in chut-kal (median 140 mg/kg) (P < 0.001). Secondary amine levels in non-nitrosated samples of kimchi (median 5.5 mg/kg) were significantly lower than secondary amine levels in non-nitrosated chut-kal (median 56 mg/kg) (P = < 0.001). Analyses of nitrite-incubated kimchi revealed high levels of total N-nitroso compounds (median 1173 micrograms/kg); the increase with nitrosation was significant (P = 0.001). The concentration of N-nitroso compounds in nitrite-incubated kimchi was significantly greater than that found in nitrite-incubated chut-kal (P = 0.015). The combination of high levels of nitrate in the kimchi, the demonstration of high levels of total N-nitroso compounds in this food after nitrosation, and the volume of kimchi consumed in the traditional diet suggest that salted pickled cabbage may play a role in gastric carcinogenesis in Southwest Korea.

Enhancement of endothelium-dependent relaxation in the aorta from stroke-prone spontaneously hypertensive rats at developmental stages of hypertension.

1. Endothelium-dependent relaxation (EDR) in aortic rings from young (8 weeks) and adult (16 weeks and 20 weeks) stroke-prone spontaneously hypertensive rats (SHRSP) was investigated in comparison with age-matched Wistar-Kyoto rats (WKY). 2. At 8 weeks, acetylcholine (3 x 10(-9)-10(-5) mol/L) and ionomycin (4 x 10(-8)-10(-6) mol/L)-induced EDR in SHRSP aortae was significantly enhanced compared to that in WKY aortae. Mechanical denudation of the endothelium completely abolished, and pretreatment of aortae with NG-monomethyl L-arginine (1 mmol/L), an inhibitor of nitric oxide formation, greatly reduced the relaxation in both strains. Indomethacin (10(-5) mol/L), a cyclo-oxygenase inhibitor that blocks the production of endothelium-derived contracting factors, did not significantly alter the relaxation by acetylcholine at this age. There was no difference in endothelium-independent relaxation of denuded aortae by sodium nitroprusside (10(-9)-10(-6) mol/L) and 8-bromoguanosine 3', 5'-cyclic monophosphate (10(-6)-10(-3) mol/L). 3. In adult SHRSP with established hypertension, however, the acetylcholine (10(-8)-10(-5) mol/L)-induced relaxation markedly diminished at any of the concentrations tested compared to that observed in 8 weeks old SHRSP and WKY at 8-20 weeks of age. This finding differed from other observations where the relaxation in SHRSP was impaired only at higher concentrations of acetylcholine. Indomethacin pretreatment of aortae from 20 week old SHRSP restored acetylcholine-induced EDR to a level comparable with that in age-matched WKY.(ABSTRACT TRUNCATED AT 250 WORDS)

Rapid detection of human group C rotaviruses by reverse passive hemagglutination and latex agglutination tests using monoclonal antibodies.

Reverse passive hemagglutination (RPHA) tests and a latex agglutination test with monoclonal antibodies (MAbs) were developed for the rapid detection of noncultivatable human group C rotaviruses. For RPHA tests, two MAbs, MAb 5A12 recognizing the outer capsid and MAb 13A3 recognizing the inner capsid, were separately used for the coating of sheep erythrocytes (SRBCs). Forty-six fecal samples were examined to confirm the practicality of the tests. As a result, there was concordance between the RPHA test with SRBCs coated with MAb 5A12 and polyacrylamide gel electrophoresis of viral RNA (RNA-PAGE) in 44 (95.6%) of 46 samples, while the diagnoses by the RPHA test with SRBCs coated with MAb 13A3 were in complete agreement with those by RNA-PAGE. Furthermore, a latex agglutination test with MAb 13A3 was also developed, and this test was fast enough and sensitive enough to successfully detect the viruses from most fecal samples within 2 min. The present procedures would be useful for the diagnosis of human group C rotavirus infections in clinical laboratories which are not well equipped.

Results obtained from microsurgical therapy of varicocele.

The microsurgical method of varicocelectomy was performed in 56 patients. In 25 cases the procedure was done because of scrotal symptoms and in 31 cases as a treatment for infertility. Complete regression of varicosities was observed in all the cases including 2 recurrent ones. The conception rate was 56% in the infertility patients excluding azoospermic and severe oligozoospermic ones. This microsurgical method has a good success rate and is also quite satisfactory in terms of its pregnancy rate.

[In vitro study for hormones and growth factors dependent cell proliferation of endometrial adenocarcinoma cells].

Sex steroid hormone dependent cell proliferation and inducing growth factors of endometrial carcinoma cells were investigated using in vitro culture systems. The cell proliferation of Ishikawa cells derived from well-differentiated endometrial adenocarcinoma which possess both estrogen and progesterone receptors were stimulated by either estradiol added to culture media or EGF and TGF-alpha acting through EGF receptors. These stimulatory effects of TGF-alpha were antagonized by the anti TGF-alpha and EGF-receptor antibodies. The cell proliferations of other endometrial cancer cells were also inhibited by those antibodies. All endometrial cancer cells secrete TGF-alpha into their culture media measured by TGF-alpha ELISA methods. The expression of TGF-alpha mRNA and secretion of TGF-alpha of Ishikawa cells were induced by estradiol but not of hormone independent HEC-50 cells. Thus suggest that estradiol dependent growth factor should be TGF-alpha in human endometrial carcinoma cells.

[Canonical discriminant analysis for hematological and serum biochemical changes during pregnancy period in squirrel monkeys (Saimiri sciurea)].

Hematological and serum biochemical data obtained from non-pregnant, pregnant and post-partum squirrel monkeys (Saimiri sciurea) were analyzed by canonical discriminant analysis (discriminant analysis with reduction of dimensionality). All animals were of wild origin and had been maintained under uniform environmental conditions at Tsukuba Primate Center for Medical Science, N.I.H., Japan. Months were standardized by the day of parturition. The calculated arithmetic means and standard deviations were listed for each item of measurement performed. Items detected statistically significant difference (p less than 0.01) between months were as follows: red blood cell count (RBC), mean corpuscular volume (MCV), hematocrit value (Ht), hemoglobin concentration (Hb), white blood cell count (WBC), albumin concentration (ALB), blood urea nitrogen concentration (BUN), total cholesterol concentration (T-CHO), triglyceride concentration (TG), alkaline phosphatase activity (ALP) and calcium concentration (Ca). Results of canonical discriminant analysis showed that the value of the first canonical variate (Z1) decreased from the early period of pregnancy to the middle period, and that the second canonical variate (Z2) decreased from the middle period of pregnancy to the end of pregnancy. The meaning of their changes were discussed.

Purification and properties of gamma-glutamyltranspeptidase from Bacillus subtilis (natto).

To understand the mechanism by which gamma-polyglutamic acid (gamma-PGA) in the sticky material of natto was synthesized, we purified the gamma-glutamyltranspeptidase (gamma-GTP) (EC 2.3.2.2) from the culture broth of Bacillus subtilis (natto) to homogeneity. gamma-GTP was composed of two subunits with molecular weight of 45,000 and 22,000. The N-terminal amino acid sequence of light subunit was homologous with that of gamma-GTP from Escherichia coli. The optimum pH and temperature of activity were 8.5 and 60 degrees C. The enzyme was inactivated by incubation for 15 min at pH 8.0 and 55 degrees C, but little loss of the activity was detected at 40 degrees C. gamma-GTP used glutamine as a gamma-glutamyl donor and acceptor for gamma-PGA synthesis. Dipeptides were better gamma-glutamyl acceptors than free amino acids.

[Management of individual body weight growth of infant squirrel monkey (Saimiri sciureus) in indoor breeding colony].

We biometrically analyzed the body weight growth data of new-born squirrel monkeys, obtained during the nursing period from 0 to 12 weeks of age. Body weight (y in grams) could be expressed as a function of birth weight (a in grams) and age (x in weeks) by the following equation: y = a + b x, where b indicates growth rate. This equation corresponded significantly with actual growth curves (R2 = 0.96). The frequency distribution of b values was demonstrated to be abnormal distribution. This value was used to judge whether the body weight growth of each monkey was normal or abnormal. The lower control limit (LCL) was calculated by using a linear equation with the b value of 9.07 (M-1.25 x S.D.) and each birth weight. For the monkeys whose body weight was above the LCL during the first three weeks after birth, it was determined whether the frequency of weighings could be reduced from 13 to 7. Using the same animals, no significant difference was detected between the b value estimated from 13 measurements and that estimated from 7 measurements. Thus, from the standpoint of management's policy to save labor, the frequency of weighings could be reduced. A new daily routine has been established in our primate center to save labor by reducing the number of body weighings of the many infant monkeys. In the new program, newborn monkeys whose body weight is above the LCL are weighed only 7 times during the nursing period of 12 weeks, while those whose weight is below the LCL are weighed 8 to 13 times.
