[Food- and water- intake, urinary and fecal output, and urinalysis in the wild-originated cynomolgus monkeys (Macaca fascicularis) under the indoor individually-caged conditions].

Daily food- and water- intake, and urinary and fecal output were determined with 253 wild-originated cynomolgus monkeys kept in individual cages. In addition, urinalysis was done for freshly collected urine. A statistically significant correlation was observed between total water intake and urinary output as well as between urinary output and urinary specific gravity (p less than 0.01).

[Responsiveness and mechanisms of action of steroid hormones in human endometrial adenocarcinoma cells].

The responsiveness and action mechanisms of steroid hormones and epidermal growth factor on human endometrial carcinoma cells are analyzed by using in vitro culture system. 1) The Ishikawa cells, derived from a well differentiated endometrial adenocarcinoma and possess ER and PR, are shown to respond to estrogens by increasing a variety of parameters, viz cell proliferation, PR levels, ALP and DNA polymerase activities. 2) ER and PR of those cells are localized in the nuclei by immunocytochemical staining using the monoclonal antibodies against to ER and PR, confirming the correctness of Gorski and Greene's one step theory involving the action mechanisms of steroid hormones. 3) Progestins reduced the ER level and stimulate E2DH activities and glycogen content, which are completely abolished by anti-progestin (RU486), suggesting that PR of those cells should be functional. 4) These responses to steroid hormones of Ishikawa cells are synergistically enhanced or appeared earlier by addition of EGF. 5) The main metabolite of E2 incubated with Ishikawa cells is E2-3-sulfate instead of E1, indicate that the higher estrogenic status may be persisted in endometrial cancer tissues.

[Mechanism of action of endocrine therapy of endometrial carcinoma].

The mechanism of action of sex steroid hormones on endometrial carcinoma was analyzed by a cell culture system. 1) Estrogenic actions such as enhancement of ALP activity, PR level and cellular growth were confirmed in Ishikawa endometrial carcinoma cells which possess ER. 2) ER and PR are both localized immunocytochemically in the nuclei of Ishikawa cells, confirming the correctness of Gorsky and Greene's theory in endometrial carcinoma. 3) In Ishikawa cells with PR, progestins stimulated E2DH activity and glycogen synthesis and reduced ER level, thus proving the existence of their hormonal action. We cannot, however, rule out the pharmaceutical action of progestin, because a decrease in nucleic acid syntheses and ALP activity, alteration in electron microscopic observation and finally cellular death were observed in endometrial cancer cells without PR. 4) Conversely, Tamoxifen, anti-estrogen, stimulated the cellular growth in serum-free environment of culture. We must therefore be careful in using it clinically.

[Pregnancy diagnosis by the ultrasonographical device and observation of fetal growth in the squirrel monkey (Saimiri sciureus)].

This paper describes usefulness of the ultrasonographical device (USD) for the diagnosis of pregnancy and the observation of fetal growth in the squirrel monkeys (Saimiri sciureus) conceived under group breeding conditions. Pregnancy was diagnosed on the basis of the detection of gestational sac (GS) in the uterus. The GS was first detected 127 +/- 10 days before delivery. The heart beat of embryo was detected around 114 days before delivery. It was able to judge conditions of fetal growth by measuring the size of GS and the biparietal diameter. No difference in uterine size between nonpregnant and pregnant animals was observed 135 days before delivery.

[Establishment and characterization of a new SCC antigen producing cell line (HCS-2) from a carcinoma of the uterine cervix].

The cell line designated HCS-2 established from a squamous cell carcinoma of the uterine cervix has been subcultivated 77 times since Nov. 8, 1983. The cultured cells appear epithelial in shape, with a pavement-like arrangement and grow without contact inhibition. In electron microscopy, the cells are characterized by desmosomal cell contacts and a few tonofilaments. The cells are transplanted subcutaneously to nude mice and produce tumor which resembles the original tumor of large cell non-keratinizing squamous cell carcinoma. The growth rate of subculture has increased gradually, and population doubling time of cells at 17th passage was about 65 hours. The chromosome studies show aneuploidy and chromosomal number was mainly from hypertriploid to hypotetraploid range. The modal number of cells at 33rd passage was 81. Specific marker chromosome is not realized. The production of SCC antigen is detected from the cells and the amount of SCC antigen in cultured media was recorded from 1.5 to 2.0 ng per 1 x 10(4) cells for 48 hours. CEA synthesis is also confirmed immunohistochemically.

Binding strength of calcium ion to bovine alpha-lactalbumin.

A Ca2+-sensitive electrode was used for determination of the binding strength of Ca2+ to bovine alpha-lactalbumin in 60 mM Tris buffer (pH 7.8-8.5) in the presence of various concentrations of NaCl. The dependence of the apparent binding constant on the concentration of NaCl was consistent with competitive binding of Ca2+ and Na+, and the binding constants of Ca2+ and Na+ were found to be 2.2 (+/- 0.5) X 10(7) M-1 and 99 (+/- 33) M-1, respectively, at 37 degrees C and pH 8.0. The temperature dependence of the binding constant of Ca2+ was examined between 30 and 45 degrees C; extrapolation of the dependence led to a binding constant of approximately 1 X 10(8) M-1 at pH 8.4 and 25 degrees C. The electrostatic contribution and conformational effect of the protein were also taken into consideration, and the intrinsic binding constant of Ca2+ to native alpha-lactalbumin was calculated to be (1.2-1.5) X 10(10) M-1 at 37 degrees C and pH 8.0.

Analysis and occurrence of total N-nitroso compounds in the Japanese diet.

A quantitative technique for differentiating the total N-nitrosamides from the total N-nitroso compounds has been devised. The principle of this method is based on the chemiluminescence response of a nitroso compound under different denitrosating conditions. We also examined the effect of nitrite scavengers on the stability of N-nitroso compounds and found that both ammonium sulfamate and hydrazine sulfate can be satisfactorily used for the detection of chemiluminescence response with hydrogen bromide-acetic acid reagent. A survey of the occurrence of total N-nitroso compounds, total N-nitrosamides, volatile N-nitrosamines, nitrates and nitrites in the Japanese diet has been conducted. Fairly high levels of total N-nitroso compounds were detected in salt-fermented vegetables, the highest being 2 500 micrograms/kg in hakusai-zuke (salt-fermented Chinese cabbage), followed by 253 micrograms/kg in takuan (salt-fermented radish roots). It is noteworthy that the amounts of total N-nitroso compounds in these products almost coincided with those of total N-nitrosamides, although no appreciable amounts, or only trace quantities, of volatile nitrosamines could be detected in these vegetable products.

Formation of N-nitroso compounds during cooking of Japanese food.

N-Nitrosamines in several traditional Japanese meals, such as 'Nebemono', 'Ten-pura' and Kara-age', were analysed. No appreciable amount, or only trace quantities, of NDMA and NPYR could be detected in these meals. The effects of various gases on nitrosamine formation in dried squid have been examined at higher cooking temperatures and the highest yield of NDMA was observed by exposure to the gas produced by the combustion of kerosene, followed by that of city gas, while very low levels of NDMA were formed when samples were wrapped in aluminium foil or broiled in air or argon atmospheres. The principal precursor of NDMA in dried squid was found to be TMAO, which is abundant in the squid muscle and from which nitrosatable DMA can be produced during broiling at higher temperatures.

[Study on so called "cellular effect" of methotrexate on choriocarcinoma and its mode of manifestation (author's transl)].

The "cellular effect" i.e. temporary elevation of HCG titer may not be infrequently experienced during course of chemotherapy against choriocarcinoma. Present paper is to clarify real view of the effect and mechanism of its manifestation by using a cell-line (GCH-nu). 1) Cellular growth was suppressed moderately by 10(-7), 10(-6) and 10(-5)M MTX. 2) During 2-4 days of administration at these concentrations of MTX HCG per ml medium was elevated 5 times. HCG in cells per mg protein were also higher 5-7 times. 3) HCG-beta in media was enhanced 5-7 times in MTX group. 4) Estrone, estradiol and progesterone were not altered. 5) Choriocarcinoma cells developed marked enlargement of nuclei and cells, multinucleation and cytoplasmic vacuolization. 6) Cytochemistry of heat stable ALP revealed increased activity especially in cells with morphological changes. ALP examined by biochemical method was enhanced 5 times. 7) Amount of protein per cell increased 10 times after giving MTX. Real view of so-called "cellular effect" includes elevations of HCG-beta and ALP activity as well as HCG, whereas sex steroids are not apparently related. The data in addition to morphological and cytochemical changes reveal that MTX may induce differentiation of choriocarcinoma cells into those of syncytial type, which then sustain the "cellular effect".

Effects of oestradiol and progesterone on the alkaline phosphatase activity of a human endometrial cancer cell-line.

The alkaline phosphatase (ALPase) activity of a human endometrial cancer cell-line, established and designated as HEC-50-B in our laboratory, was investigated biochemically and histochemically in relation to its cell growth pattern and to the effects of the sex steroid hormones, oestradiol and progesterone. The ATPase activity increased sharply in the early stationary phase to reach an activity almost 2.5 times higher than that obtained in earlier stages of the culture. On administration of oestradiol to the culture medium, a sharp elevation of the ALPase activity was induced on an average of 2 days earlier (late logarithmic phase) than in the case of an ordinary culture (no hormone administration), without causing a notable change in cell growth pattern. It should be noticed, however, that progesterone at such a low concentration that had very little effect on cell growth in the culture could clearly prohibit the elevation of ALPase activity. This hormonal effect on the ALPase activity resembled that on the enzyme activity of the endometrium of adult women. The ALPase activity of both the cultured endometrial cancer cells and the endometrium was found to be a sensitive indicator of the effect of progesterone. It would be a useful tool for future study in elucidating the mechanism of hormonal control of the neoplasm.

Occurrence, formation and precursors of N-nitroso compounds in the Japanese diet.

A survey has been made of volatile nitrosamines in 123 fermented foods. These included salt-fermented vegetables, soy sauce, "miso", Japanese "sake" and other fermentation products. No appreciable amount, or only trace quantities, of NDMA and NPYR were detected in vinegar, "sake", wines and whiskies, while minute quantities of NDMA were detected in 31 out of 33 beer samples, where the levels ranged from traces to 13.8 micrograms/l, with an average of 5.1 micrograms/l. The nitrosamine content of salt-dried fish apparently increased when these products were broiled in a city gas range. A method for the determination of N-alkylureas in foods has been developed. A total of 57 food samples, both uncooked and broiled in a gas range or fried, were analysed. No alkylureas could be detected in these samples.

Detection of HeLa cell contamination--presence of human papillomavirus 18 DNA as HeLa marker in JTC-3, OG and OE cell lines.

There are warnings of the contamination of cell cultures with HeLa cells in many laboratories in the world. The cell lines JTC-3, OG and OE that were established in Okayama in 1959, 1969 and 1971, respectively, were examined for human papillomavirus (HPV) 18 DNA by Southern blot hybridization. The HPV 18 DNA detected in these three cell lines showed hybridization patterns characteristic of the HPV 18 DNA in the HeLa cell line established in 1951. Southern hybridization patterns of HPV 18 DNA in the cellular DNA of the C4-II cervical cancer cell line that was established in the USA in 1962 was different from that of HeLa cells. These results suggest that the JTC-3, OG and OE cell lines have been contaminated by HeLa cells.
