Trace elemental analysis of extracted dust from lungs and lymph nodes of domestic animals using X-ray fluorescence technique.

Samples of dust extracted from the lungs and lymph nodes of certain domestic animals from West Bengal, Orissa, Bihar provinces of India were quantitatively analysed using photon excited energy dispersive X-ray fluorescence technique. Thin specimens were prepared for analysis to minimize matrix enhancement and absorption effects. Amongst the various elements analysed, Hg, Fe, Cu, Zn and Pb were found to be present in appreciable amounts. An important finding was the presence of a very high concentration of Hg in extracted dust samples from West Bengal. The significance of the various results obtained in this investigation is discussed.

Multicentric metachronous primary carcinoma of the upper alimentary tract in 'Biri' smokers.

Three Biri smokers with multicentric primary metachronous squamous cell carcinomas of the upper alimentary tract are presented. The susceptibility of the tissues of the upper alimentary tract to multiple malignancies, and the carcinogenic effect of radiation and Biri smoke on these tissues, are discussed. We have tried to assess the role of continuous Biri smoking in the production of a second malignancy of the upper food passages in patients who received radiotherapy for their first tumour.

Effects of superoxide dismutase, dithiothreitol and formate ion on the inactivation of papain by hydroxyl and superoxide radicals in aerated solutions.

Losses in enzyme activity and sulphydryl content have been studied in aerated papain solutions containing formate, superoxide dismutase and dithiothreitol. Both formate and dithiothreitol converted .OH to .O2-, whereas superoxide dismutase completely suppressed the inactivation by .O2-. Using results from all three systems, the fraction of .O2- reactions with papain that caused inactivation of the enzyme was 0.33 +/- 0.07. The results also showed that the fraction of .OH reactions, which cause inactivation of papain, is significantly higher in aerated than in oxygen-free solutions.

The repair, protection and sensitization of papain with respect to inactivation by H2O2 and OH: effects of dithiothreitol, penicillamine, cystine and penicillamine disulphide.

While dithiothreitol repairs the peroxide-produced sulphenic acid derivative of papain in a fast reaction involving only one dithiothreitol molecule, penicillamine reacts with it to form papainCys25SSPen. Disulphide is also formed in the absence of peroxide (see article) by reactions of papainCys25S- AND PenS- radicals derived from -OH reactions in penicillamine-papain mixtures. A similar formation of papainCys25SSCys occurs in mixtures of cysteine and papain. However, unlike papainCys25SSCys, papainCys25SSPen cannot easily be restored to the active form of papain by the exchange reaction with CysSH, and this may have significance for an understanding of the sensitizing action of penicillamine observed in some in vivo systems. Under the action of OH radicals dithiothreitol has less of a tendency to form mixed disulphides and is more effective in repairing papain-OH intermediates than either cysteine or penicillamine. Due to secondary reactions of RSOH and other oxidized species the disulphides of cysteine and penicillamine are less effective than the sulphydryls in protecting papain against inactivation by -OH.
