Triglyceride dependence of factor VII coagulant activity in deep venous thrombosis.

The effect of incubation of plasma with lipoprotein lipase on factor VII coagulant (FVII:C) activity was examined in 40 patients, 22 male and 18 female, aged 28 to 77 years, with history of venographically proven deep venous thrombosis (DVT). While the mean (+/-SEM) FVII:C activity of the 40 patients was 100.9 +/- 4.1%, 19 patients had FVII:C activity less than 100%, 11 had 100 to 120% activity and 10 patients had greater than 120% FVII:C activity. The mean triglyceride level of all the patients was 84.0 +/- 6.5 mg/dl. The FVII:C activity correlated significantly with triglyceride (r = 0.36; n = 40; p = 0.021). There was about 30% average loss of FVII:C activity upon incubation of plasma with lipoprotein lipase. The mean activity loss increased from 23.8% to 31.5% and 42.6% in patients whose FVII:C activity levels were less than 100%, between 100 and 120% and more than 120% respectively, the variation in the means being statistically significant (p < 0.001). While according to current opinion, FVII:C activity represents the total FVII mass (FVII plus FVIIa) and activity state, the present findings demonstrate a lipid dependence of FVII:C activity, and raises the possibility of a therapeutic option of controlling FVII:C by controlling triglyceride levels.

The potential thrombogenic action of a nonionic radiographic contrast medium used during coronary angiography is offset by heparin during coronary angioplasty.

Iohexol sodium, a nonionic radiographic contrast medium, used in invasive imaging techniques has been shown to be potentially thrombogenic. In the present study, the effect of iohexol sodium on haemostatic factors was evaluated in 20 patients, 16 male and 4 female, 10 undergoing coronary angiography and another 10 undergoing coronary angioplasty. All the patients had angiographically-assessed coronary artery disease. The patients undergoing coronary angioplasty received a significantly larger quantity of the dye as compared with the patients undergoing coronary angiography. The former group of patients also received a bolus of 20,000 units of standard heparin in addition. The levels of thrombin-antithrombin-III complex (TAT), prothrombin fragments 1 and 2 (F1F2), D-dimer and the functional activity of tissue factor pathway inhibitor (TFPI) were assayed. While the baseline and 30-min post-dye levels of TAT and F1F2 were comparable in patients undergoing coronary angioplasty, the 30 min levels were significantly elevated in patients undergoing coronary angiography. The post-dye levels of TFPI activity were significantly increased in the former group due to the heparin-induced release of TFPI. It is concluded that the thrombogenic potential of iohexol sodium was overcome by heparin used routinely during coronary angioplasty.

Electron microscopic characteristics of actinomycetic agents having aetiological association with human leprosy and epizootic ulcerative syndrome of fish.

The transmission electron microscopic (TEM) studies of the human leprosy derived chemoautotrophic nocardio-form (CAN) bacteria and EUS derived CAN bacteria showed presence of double contoured cell-walls consisting of an electron transparent and a dense layer. The fibrillar structures on the surface of these CAN bacterial cells also suggested their similarity to the human tissue derived Mycobacterium leprae cells. These EM studies further revealed mycelial and coccoid bodies in all these bacteria as was observed originally.

On the application of phase relationships to complex structures. XXXV. Some experiments with 2-Zn insulin.

The direct-methods program SAYTAN has been applied to the known structure of 2-Zn insulin with 806 atoms, excluding solvent, in the asymmetric unit. Useful sets of phases can be obtained and selected by figures of merit for data resolutions between 1.5 and 2.25 A and these can be extended by SAYTAN to give mean phase errors of 68 degrees for more than 2000 reflections. A feature of the phases so found is that the phase errors decrease with increasing resolution - which is the opposite of the situation when phases are found by isomorphous-replacement techniques.

Syntheses of several 99mTc and 131I labeled neoglycoalbumins and their differential uptake patterns in animal biodistribution experiments.

Several glycoconjugates, alpha-D-mannopyranosyl, beta-L-fucopyranosyl, alpha-L-rhamnopyranosyl, beta-D-glucopyranosyl and beta-D-galactopyranosyl human serum albumin, were synthesized using C9-tether and radiolabeled with 99mTc and 131I. Both 99mTc and 131I radiolabeled neoglycoalbumins had considerable stability and exhibited similar biodistribution patterns within the experimental limits. The results of biodistribution studies can be explained from the in vitro observations that 99mTc-beta-D-galactopyranosyl albumin binds to hepatic binding protein in liver in a dose-dependent fashion. The radiolabeled glycoalbumins derived from D-mannopyranose and L-fucopyranose also bind in a dose-dependent fashion to the receptors present in the liver sinusoidal cells and spleen macrophages. The beta-D-glucopyranosyl and alpha-L-rhamnopyranosyl neoglycoalbumins accumulate nonspecifically in liver and spleen.

Phage-mediated conjugation in staphylococci as a means of transfer of drug-resistance.

Penicillin resistance plasmid was transferred from Staphylococcus aureus B4 (PcrKms, donor) to S. aureus ML351 (PcsKmr, recipient) by co-cultivation of the donor with the recipient in nutrient broth with or without the modifying effects of CaCl2 or sodium dodecyl sulfate. It was found that the transfer of drug-resistance occurred maximally between 6 and 18 hr postinoculation; however, addition of DNase (200 micrograms/ml) could totally prevent such a transfer up to 6 hr and significantly reduce it thereafter. Cell-free filtrate of the donor culture when mixed with the recipient was ineffective in bringing about the transfer of Pcr.

A pathogenicity model for the agents of epizootic ulcerative syndrome (EUS) in fish.

Chemoautotrophic nocardioform (CAN) bacteria had been repeatedly isolated from fish with ulcerative disease syndromes (EUS) from the massive epizootics that had repeatedly occurred since 1988 in eastern India as the major or only pathogenic agent in the background of distinctive environmental and epizootic data. Since these isolates bear significant similarity to the human and rat leprosy bacilli, attempts had been made to demonstrate the pathogenicity of this fish pathogen in the "Swiss" strain of mice as a convenient model. The studies reveal that the fish CAN bacteria could produce pathogenic effects in mice similar to that of the rat leprosy bacillus.

Atherogenic markers in the offspring vis-à-vis family history of coronary heart disease.

The object of the study was to determine if atherogenic markers in the offspring reflected family history of coronary heart disease (CHD). Two hundred and four male subjects aged 9-18 years, mean age 13.3 +/- 1.7 (S.D.) years, and apparently healthy and normolipidemic, were selected for the study. Information on their family history of fatal or non-fatal heart attack or stroke was obtained on a questionnaire, and a scoring system was devised to grade the positivity of family history. The boys having the highest decile values of triglyceride, ratios of LDL/HDL-cholesterol, total/HDL-cholesterol and apolipoprotein B/A-I, and cumulative skin-fold thickness, a marker of obesity, had higher family history scores and increased occurrence of myocardial infarction in their families than the boys with the lowest decile values of these variables. The reverse was true with variables such as HDL-cholesterol and apolipoprotein A-I, high levels of which are known to be cardioprotective by virtue of controlling reverse cholesterol transport. The mean values of triglyceride and total/HDL-cholesterol ratio were significantly elevated in the boys with positive parental history of CHD as compared with those with negative parental history. Thus, although the boys were normolipidemic, their lipoprotein markers and an obesity marker exhibited a variation which in turn reflected family history of CHD.

Cytopathological study of bone tumours.

Fine needle aspiration biopsy is now widely used for detection of any pathological lesions like neoplasm, specific inflammation, cysts, etc. In a study of 22 cases, 17 (77.2%) were diagnosed by aspiration. Specific diagnosis was found in 59% cases and in 18.2% cases suggestive diagnosis was confirmed by histopathological examinations from biopsy materials. Like other investigative methods it is accurate and informative. It is also simple, can be done quickly and cheaper economically.

Triglyceride and coronary heart disease.

The predictive association of triglyceride (TG) in coronary heart disease (CHD). In patients without CHD levels were elevated in those coronary risk factors, as compared to those without the risk factor the elevation of TG may occur in asymptomatic subjects before the onset of clinical manifestation of CHD. The high-risk, asymptomatic subjects with high levels of TG may be on the "threshold" of the sysptomatic phase of CHD. The levels had a positive correlation with body mass index and increased with increasing number of coronary risk factors.

On the application of phase relationships to complex structures. XXXII. A small protein at low resolution.

The direct-methods program SAYTAN is applied to data at various restricted resolutions for a small protein. It is shown that useful sets of phases can be obtained even down to 3 A resolution. Conventional figures of merit are not very discriminating for the phase sets developed, but modified figures of merit seem capable of selecting the better phase sets, at least for those generated from 2 A or higher resolution data.

An IgG human monoclonal antibody that reacts with HIV-1/GP120, inhibits virus binding to cells, and neutralizes infection.

A human mAb (HmAb) termed F105 was obtained by fusion of antibody-producing EBV-transformed cells with the HMMA2.11TG/O cell line. F105 is an IgG1 kappa antibody that binds to the surfaces of cells infected with all HIV-1 strains tested: MN, RF, IIIB, and SF2, but not uninfected cells. The HmAb immunoprecipitates GP120 from all four strains. F105 does not react with denatured GP120 on Western blots, but does react with viral lysates and purified GP120 dotted onto nitrocellulose filter paper under nondenaturing conditions. rGP120 from SF2 and soluble rCD4 inhibit antibody binding to infected cells in a dose-dependent manner. F105 inhibits the binding of free, infectious virions to uninfected HT-H9 cells with 50% of maximal (100%) inhibition at approximately 1 microgram/ml. F105 inhibits infection of HT-H9 cells by 100 tissue culture infective dose 50% units of MN and IIIB strains with 50% inhibition at concentrations of HmAb readily achievable in man. It appears that the F105 HmAb reacts with a conformationally defined epitope on HIV-1/GP120 that is exposed on the free virion and is important for binding to the cell surface by the virion. The epitope, which is immunogenic in humans, appears to be within, or topographically near, the CD4-binding site. F105 and the F105 epitope are potentially useful in therapy and in the design of peptide or anti-Id based vaccines; monitoring of the expression of the Id may prove useful in evaluating immune responses in infected individuals or vaccinated volunteers.
