Protection against keratoconjunctivitis shigellosa induced by immunization with outer membrane proteins of Shigella spp.

Active immunization of guinea pigs and rabbits with outer membrane proteins (OMP) isolated from Shigella flexneri 3a and Shigella sonnei phase I protected the animals against keratoconjunctivitis shigellosa induced with the homologous or heterologous strain. Protection was also achieved in rabbits after passive immunization with anti-OMP immune serum. Active immunization with lipopolysaccharide of S. flexneri 3a did not protect rabbits against keratoconjunctivitis shigellosa.

Transferable drug resistance of Escherichia coli isolated from infant diarrhoea.

The resistance to several drugs was determined in 29 pathogenic strains of Escherichia coli (026, 055 and 0111) isolated from infant diarrhoea and 18 non-pathogenic E. coli strains isolated from the same individuals. Both pathogenic and nonpathogenic strains were resistant to at least 1 to 10 drugs, but only in four cases resistance patterns of the pathogenic strains were identical with those of non pathogenic ones. The majority of the strains were resistant to sulfonamide, tetracycline, ampicillin, carbenicillin, neomycin and kanamycin. The drug resistance (except the resistance to nalidixic acid and rifampicin) was associated with conjugative R-plasmids. Some of the tested strains carried two R-plasmids in one cell, being in hetero R-state.

Studies on virulence of Shigella flexneri.

The antigenic structure and some biological properties were compared in virulent S. flexneri strains of differences in: 1) antigenic composition; 2) biochemical activity; 3) sensitivity to a set of Shigella phages and 4) susceptibility to phagocytosis. The only difference found concerned the LD50 for mice; it was 10 to 100 times larger for avirulent than for virulent strains of S. flexneri. The toxic products of the strains were also compared. The lipopolisaccharide and free endotoxin purified from virulent and avirulent variants behaved similarly when tested for: LD50, pyrogenicity and the local Shwartzman reaction. A striking difference was demonstrated in the ultrastructure of lipopolisaccharide and free endotoxin isolated from virulent and avirulent variant of S. flexneri 3a.

Fimbriae and haemagglutinating properties in Edwardsiella and Levinea.

Using the haemagglutinating procedure, the presence of fimbriae was revealed in all 34 Edwardsiella strains tested and in 3 Levinea strains out of 17 examined. The great majority of fimbriate Edwardsiella strains caused haemagglutination of MR type; only two strains of Edwardsiella and all fimbriate strains of Levinea brought about haemagglutination of MS type. All the strains exhibited a similar spectrum of haemagglutinating activity when they were tested against red cells of various animal species. Under the electron microscope MR fimbriae of Edwardsiella were found to be thicker than MS fimbriae (6.5 nm and 6.0 nm respectively). MS fimbriae of Levinea had 6.5 nm in diameter.

Serologic relationship of fimbriae among Enterobacteriaceae.

The existence of serological relationship of fimbriae among serotypes of the same genus and among genera of Enterobacteriaceae was determined by agglutination and absorption procedures. It was found that fimbriae of different serotypes of the same genus and among the genera a) Escherichia, Shigella and Klebsiella, and b) Salmonella, Arizona and Citrobacter were more or less antigentically related. No relationship was found among fimbriae of the Edwardsiella, Enterobacter, Hafnia, Serratia, Proteus and Providencia. It was also found that in Klebsiella, MS and MR fimbriae differed antigenically. On the basis of the results obtained nine antigentically distinct types of fimbriae were distinguished.

Immunochemical studies on the serotype of Shigella flexneri and its recombinant.

Chemical and serologic analysis of polysaccharides of Shigella flexneri 2b and its recombinant revealed complete defect of the gene responsible for type II specificity as a result of recombination. At the same time, function of the gene in the same locus responsible for 7,8 group specificity was partially inhibited. Attenuation of group antigen 7,8, associated with partial uncovering of the structure of the basic chain, causes appearance of a distinct 3,4 group antigen. Changes of this type in Shigella flexneri have not been described hitherto.
