Destruction and reorganization of the receptor membrane in labellar chemosensory cells of the blowfly. Long-lasting latent action of colchicine.

Electron micrographs of sections of the labellar chemosensilla of the blowfly, Phormia regina, showed that treatment with sodium deoxycholate (DOC; 7.2 mM for 2 min) destroyed the distal processes of the receptors from up to 10 microns from the tip of the sensillum, but these processes regenerated almost completely within 0.5 h. However, when DOC treatment was preceded by colchicine treatment (25 mM for 2 min), greater than 10 h was required for complete regeneration. Sugar receptor responses supported these findings and disclosed a more detailed time course of regeneration after DOC treatment: without colchicine pretreatment, the destroyed distal process completely regenerated in 0.3-1.0 h, but with pretreatment, regeneration began at 3 h and reached the chemosensillar tip at 8 h at the earliest. Hardly any depression of the response was observed for 8 h after treatment with colchicine alone, but a transient depression was detected at 12 h. Based on these results, the role of microtubules in the maintenance of the receptor membrane is discussed.

Destruction and reorganization of the receptor membrane in labellar chemosensory cells of the blowfly. Recovery of responses to sugar after destruction.

Recovery from destruction by sodium deoxycholate (DOC) was studied with the receptor membrane of the blowfly, Phormia regina. The recovery can be divided into two processes, colchicine dependent and colchicine independent. The colchicine-dependent process was completely depressed by pretreatment with colchicine at 5 mM for 2 min (partially at 0.1 mM for 10 min), but the colchicine-independent one persisted. Vinblastine also caused depression but lumicolchicine did not. Records of responses obtained from the DOC-treated sugar receptor showed long response latencies that gradually became indistinct with recovery. Colchicine also affected this change in response latency after the DOC treatment. These results suggest that the colchicine-dependent recovery process is related to microtubules in the distal process of the receptor cell. The recovery time course and the change in response latency could be quantitatively explained by the simple assumptions that DOC underwent desorption from the receptor membrane (colchicine-independent recovery process) and that regeneration of the disrupted distal process of the receptor cell accompanied recovery in the number of available receptor sites (colchicine-dependent recovery process).

Effects of sensitization of hemolytic streptococcal M-protein fraction on embryonic heart in the rat.

After sensitization by M-protein fraction (MP) of hemolytic streptococci for two generations, Wistar rats were injected with 9 ml/kg of MP on day 9 of gestation (plug day = day 0). The incidence of malformations was about 12% whether anti-MP antibody was positive or not in the maternal sera. The primary malformations were ventricular septal defect, anophthalmia, and microphthalmia. Light microscopic study revealed no myocarditis nor endocarditis in either the maternal or fetal heart. No antinuclear antibody was detected in the maternal sera by the fluorescent antibody technique. No complete heart block was found in the fetus by electrocardiogram. These results were similar to previous findings using a single injection of MP, and therefore further demonstration that the principal factor in the teratogenesis induced by MP administration in the rat is not an inflammation in the fetal organs caused by an autoimmune mechanism but rather yolk sac dysfunction.

Des-O-methylolivoretin C is a new member of the teleocidin class of tumor promoters.

Des-O-methylolivoretin C, a demethylated form of olivoretin C, is a naturally occurring compound in Streptomyces mediocidicus and Streptoverticillium olivoreticuli. Des-O-methylolivoretin C is a regioisomer of teleocidin B, which has the same activity as teleocidin. The tumor-promoting activity of des-O-methylolivoretin C was studied in a two-stage carcinogenesis experiment on mouse skin in comparison with that of teleocidin. Treatments with 7,12-dimethylbenz[a]anthracene (DMBA) plus des-O-methylolivoretin C and DMBA plus teleocidin induced tumors in 63.3% and 84.6% of the mice, respectively, in week 30. The difference in the tumor-promoting activities of des-O-methylolivoretin C and teleocidin is presumably related to the regioisomeric difference in the cyclohexene ring.

[Study on the induction of immunological suppression by blood transfusion].

There have been several reports that suggest the possibility that blood transfusion induces the suppression of immunity. For the purpose of studying immunological effect of blood transfusions, in vivo and in vitro experiments were performed. Using C3H/He (H-2k), DBA/2 (H-d), C57BL/6 (H-2b) and BDF1 (H-2b,d) mice, blood transfusions were done each other. In in vitro experiment, the effect of blood transfusion was examined with the survival of allogenic tumor, Meth-A (H-2d), transplanted in C3H/He (H-2k). And in in vitro experiment, natural Killer activity and CTL activity were investigated following blood transfusions. These experiments led to the following results: Some allogenic blood transfusions enhanced allogenic tumor survival. Only leucocytes transfusion showed such effects. The effective factor existed in the transfused serum. NK activity was suppressed by blood transfusion. CTL activity was not correlated with blood transfusion. From these results, it was concluded that allogenic blood transfusion could cause immunosuppression, especially suppression of NK activity and it was considered that clinical blood transfusion can also result in a tendency towards acceleration of tumor growth in the cancer-carrying body.

[Clinical and immunological study on the high incidence of malignant neoplasms in patients with chronic renal failure].

With the advancement of dialysis techniques, rehabilitation and high morbidity are becoming serious problems for patients with chronic renal failure. The authors, with the purpose of investigating the carcinogenic state of such patients, studied the incidence of malignant neoplasms and their immunity both clinically and fundamentally. Out of 499 patients with chronic renal failure, malignant neoplasms were found in 17 cases (3.40%), which is obviously a high rate with a statistically significant difference from that of the ordinary healthy population. In the fundamental immunity study, OKM1+ cells of the patients were significantly lower than those of healthy people, and a decrease in NK and K cells was inferred. Incidentally, the lymphocytes of patients strongly inhibited the mitogen response of healthy subjects. This decline in cellular immunity is considered to be closely related to the carcinogenicity of patients with chronic renal failure.

Synergistic effect of subteratogenic doses of antisera on the development in the rat.

Wistar rats were administered rabbit anti-rat-umbilical-cord serum (AUCS) alone on day 9 of gestation or in combination with subteratogenic doses of rabbit anti-rat-kidney serum (AKS). The results showed that the incidence of malformation was low (13.1% +/- 7.0 S.E.) even when very high doses of AUCS alone were administered. However, the incidence markedly increased to 61.0% +/- 9.3 S.E. when a small dose (0.10 ml/100 gm) of AKS was administered on day 9 followed by 0.90 ml/100 gm of AUCS on day 10. The malformations included microphthalmia, hydrocephaly, aortic arch anomalies, ventricular septal defect, and anophthalmia, being similar to the effects of a medium dose of AKS. These results suggest that a combination of subteratogenic doses of teratogenic antisera produces malformations, possibly by mechanisms that potentiate the teratogenic action of the sera.

The effect of a high fat diet on pyruvate decarboxylase deficiency without central nervous system involvement.

A nine-year-old Japanese boy with low pyruvate decarboxylase activity in fibroblasts showed no central nervous symptoms except for muscle fatigue. The pyruvate decarboxylase activities in fibroblasts of the patient and two control subjects were 0.407 +/- 0.083, 1.029 +/- 0.137 and 1.607 +/- 0.096 mumoles/g protein/30 min, respectively. The Michaelis-Menten constant (Km) was the same in the patient and controls. There was no inhibitor of pyruvate decarboxylase in the patient's fibroblasts. A high fat diet has been given to the patient for five years. At present he does not complain of any kind of muscle fatigue, except after severe exercise. Mental and physiological development of the patient are within the normal ranges. However, trials of orally administered thiamine hydrochloride or thiamine hydrochloride combined with lipoamide did not improve his muscle fatigue.

Plasma levels of 25-hydroxyvitamin D2 and 25-hydroxyvitamin D3 in maternal, cord and neonatal blood.

A high-performance liquid chromatographic method for simultaneous assay of 25-hydroxyvitamin D2 (25-OH-D2) and 25-hydroxyvitamin D3 (25-OH-D3) proposed in a previous paper (1) was applied to determine the plasma levels of the metabolites in perinatal and postnatal periods. The plasma samples of maternal, cord and neonatal blood were collected in summer and winter seasons. 25-Hydroxyvitamin D3 was detected in all the samples. The plasma levels of the metabolite in mothers, cords and newborn infants (at life within 24 hr) in summer were 33.9 +/- 12.5, 18.9 +/- 8.4 and 16.6 +/- 6.4 (mean +/- S.D.) ng/ml, respectively, while those in winter were 15.8 +/- 6.6, 8.8 +/- 3.4 and 7.7 +/- 3.2 ng/ml, respectively. The data in summer were significantly higher than the respective data in winter and there were high significant correlations between the mothers and cords and between mothers and newborns. In both seasons, the plasma levels of mothers were about two times higher than the respective data of cords and newborns which were nearly identical with one another. The similar tendency was always observed in the individual data of mothers-cords-newborns pair samples. In this study, many plasma samples from mothers, cords and newborns were examined, but 25-OH-D2 was detected in only few samples (6/41 for mothers, 3/36 for cords and 2/34 for newborns). However, the metabolite began to appear in all the samples during nursing with vitamin D2-fortified dry milk to show 4.6 +/- 1.3 and 4.8 +/- 1.2 ng/ml in the summer and winter samples of neonates at life of 5-6 days, respectively. When the variation of plasma 25-OH-D2 and 25-OH-D3 levels was examined in postnatal periods until one month, the levels of exogenous 25-OH-D2 was increased while those of endogenous 25-OH-D3 was decreased. The sum of vitamin D2 intake from fortified dry milk was highly significantly correlated with the plasma levels of 25-OH-D2, which indicates that daily uptake of exogenous vitamin D2 is very important in nutrition during postnatal periods of bottle-fed infants.
